We synthesized the results from a flow cytometric seed screen and the literature to infer the phylogenetic origin and the geographical and taxonomic distribution of apomixis in tribe Potentilleae (Rosaceae). We distinguished between regular sexuality and apomixis, the zygotic and parthenogenetic origin of the embryo, and the pseudogamous (i.e. sexual) versus autonomous origin of the endosperm. The combined evidence provides information on reproductive modes for 11 genera and 120 species. For the first time records on reproductive mode are provided for the genus Farinopsis, 29 species (from five genera), and seven series of Potentilla. Regular sexuality was observed in Aphanes, Argentina, Comarum, Dasiphora, Drymocallis, Farinopsis, Fragaria, Horkeliella, Potentilla, and Sibbaldia. Reliable evidence for apomixis is restricted to two evolutionary lineages of Potentilleae: the Potentilla core group and Alchemilla/Aphanes. Early evolutionary divergence of these lineages (approximately 50 Mya), characterized by pseudogamous and autonomous apomictic seed formation, respectively, suggests parallel origins of apomixis. Apomixis is shown to be taxonomically widespread in the whole Northern Hemisphere distribution range of Potentilla, a pattern that is explained by hybrid transfer and repeated intercontinental dispersals. Bone marrow, spleen, liver and kidney proton transverse relaxation rates (R2), together with cardiac R2* from patients with sickle cell disease (SCD), paroxysmal nocturnal hemoglobinuria (PNH) and non-transfusion dependent thalassemia (NTDT) have been compared with a control group. Increased liver and bone marrow R2 values for the three groups of patients in comparison with the controls have been found. SCD and PNH patients also present an increased spleen R2 in comparison with the controls. The simultaneous measurement of R2 values for several tissue types by magnetic resonance imaging (MRI) has allowed the identification of iron distribution patterns in diseases associated with iron imbalance. Preferential liver iron loading is found in the highly transfused SCD patients, while the low transfused ones present a preferential iron loading of the spleen. Similar to the highly transfused SCD group, PNH patients preferentially accumulate iron in the liver. A reduced spleen iron accumulation in comparison with the liver and bone marrow loading has been found in NTDT patients, presumably related to the differential increased intestinal iron absorption. The correlation between serum ferritin and tissue R2 is moderate to good for the liver, spleen and bone marrow in SCD and PNH patients. However, serum ferritin does not correlate with NTDT liver R2, spleen R2 or heart R2*. As opposed to serum ferritin measurements, tissue R2 values are a more direct measurement of each tissue’s iron loading. This kind of determination will allow a better understanding of the different patterns of tissue iron biodistribution in diseases predisposed to tissue iron accumulation.  相似文献

Social insects exhibit a great variability in their social organization,and this affects colony kin structure, relatedness among nestmates, and population genetic structure. In the mosaic of arborealants of neotropical habitats, mutually exclusive dominant antspecies occupy different territories, and their nest distributionis spatially aggregated in patches influencing patterns of populationgenetic structure. In this study, we performed an analysis ofthe population and colony genetic structure of the facultativepolygynous ant Ectatomma tuberculatum to investigate how theparticular breeding and social system of this species can explainits ecological dominance in the mosaic. Within-nest geneticanalysis revealed that relatedness between nest mate workerswas significantly greater than zero (r = 0.30) with an effectivenumber of queens per nest of Ne = 2.5–3, indicating thatpolygyny is functional in this species. Moreover, we found thatqueen number was highly variable, probably due to queen adoptionevents, leading to the prevalence of polygyny over monogyny.Finally, the strong population genetic structure and the significantisolation by distance suggested that both budding and polydomytake place in this species. The respective role of secondarypolygyny, budding, and polydomy are then discussed in the contextof the mosaic of arboreal ants, and we propose that this particularsocial organization ensures the ecological dominance of E. tuberculatumby optimizing the colonization of new available nesting sitesand by increasing territory size.  相似文献

We have shown previously that the 134-residue endonuclease domain of the bacterial cytotoxin colicin E9 (E9 DNase) forms channels in planar lipid bilayers (Mosbahi, K., Lema?tre, C., Keeble, A. H., Mobasheri, H., Morel, B., James, R., Moore, G. R., Lea, E. J., and Kleanthous, C. (2002) Nat. Struct. Biol. 9, 476-484). It was proposed that the E9 DNase mediates its own translocation across the cytoplasmic membrane and that the formation of ion channels is essential to this process. Here we describe changes to the structure and stability of the E9 DNase that accompany interaction of the protein with phospholipid vesicles. Formation of the protein-lipid complex at pH 7.5 resulted in a red-shift of the intrinsic protein fluorescence emission maximum (lambda(max)) from 333 to 346 nm. At pH 4.0, where the E9 DNase lacks tertiary structure but retains secondary structure, DOPG induced a blue-shift in lambda(max), from 354 to 342 nm. Changes in lambda(max) were specific for anionic phospholipid vesicles at both pHs, suggesting electrostatics play a role in this association. The effects of phospholipid were negated by Im9 binding, the high affinity, acidic, exosite inhibitor protein, but not by zinc, which binds at the active site. Fluorescence-quenching experiments further demonstrated that similar protein-phospholipid complexes are formed regardless of whether the E9 DNase is initially in its native conformation. Consistent with these observations, chemical and thermal denaturation data as well as proteolytic susceptibility experiments showed that association with negatively charged phospholipids destabilize the E9 DNase. We suggest that formation of a destabilizing protein-lipid complex pre-empts channel formation by the E9 DNase and constitutes the initial step in its translocation across the Escherichia coli inner membrane.  相似文献

Our objective was to evaluate factors that affected the success of embryo transfer programs in large dairy herds. Non-lactating donor cows produced a larger number of ova/embryos (P<0.01) and viable embryos (P<0.01) than lactating cows. The interaction between season and donor class was correlated with the proportion of ova/embryos classified as fertilized (P=0.03), because lactating donors had fewer fertilized ova in the summer. There was no correlation between 305-day mature equivalent milk yield and response to superstimulation. Although the interval between superstimulation protocols was correlated with the number of ova/embryos (P=0.03), there was no correlation with the number of viable embryos. Pregnancy per embryo transfer (P/ET) in heifer recipients was correlated with embryo quality grade (P<0.01), season (P=0.04), and whether embryos were fresh or frozen/thawed (P<0.01). Lactating recipient cows tended to have a lower rate of P/ET during the summer (P=0.12 to P=0.08). Synchronization protocols tended to be (P=0.06; Herd 1) or were (P=0.02; Herd 2) correlated with P/ET. Lactating cows receiving vitrified IVF embryos had a lower (P=0.01) P/ET than those receiving fresh IVF embryos, especially in the summer (P=0.09). Milk yield was not correlated with P/ET. The use of heat abatement systems is critical to improve embryo production and P/ET. Synchronization protocols that optimized synchrony of ovulation may increase fertility of recipient cows and eliminate the need for estrous detection.  相似文献

Phosphoenolpyruvate carboxykinase (PEPCK) was shown to be present in a range of developing seeds by measurement of its activity and by immunoblotting. Its function was investigated during grape (Vitis vinifera L.) seed development. The maximum abundance of PEPCK coincided with the deposition of storage reserves. At this stage of development, immunohistochemistry showed that PEPCK was very abundant in a layer of cells located at the boundary of developing storage tissues and in the chalaza (close to the termination of the vascular supply to the seed) and was also present in the palisade layer of the seed coat (the inner layer of the outer integument). Earlier in development PEPCK was also present in the developing palisade layer and in the inner region of the nucellus which surrounds the developing endosperm. At later stages of development, PEPCK was located in the outer region of the endosperm. However, PEPCK was present in the phloem of the seed at all stages of development. Feeding of asparagine to developing grape seeds led to a strong induction of PEPCK. We suggest that, in developing grape seeds, both the chalaza and palisade tissue may distribute imported assimilates from the vasculature to the developing storage tissues and that PEPCK may play a role in the metabolism of nitrogenous assimilates during their delivery from the vasculature to the storage tissues.
