Three new diterpenoids and one known diterpenoid have been isolated from a sea anemone of the order Actiniara, and the structures of the new compounds, actiniarins A-C (1-3) were established on the basis of extensive 1D and 2D NMR spectroscopic data interpretation. Compound 1 has a six-membered ring hemiacetal ring, and the equilibrium of this ring is discussed. All the isolates were evaluated for their inhibition of Cdc25B and for cytotoxicity against the A2780 ovarian cancer cell line.  相似文献

Synaptosome-associated protein-23 (SNAP23) is a component of the cellular mechanism required for specific membrane fusion and targetting of intracellular vesicles. We have cloned the full-length human cDNA and the SNAP23 gene. The SNAP23 gene has eight exons, with the initiation codon located in exon 2, and maps to the human chromosome 15q21-22 region. The human SNAP23 gene can generate two types of message, the full-length message (SNAP23A) and a shorter message (SNAP23B). The latter is the result of alternative splicing where exon 5 is joined to exon 7 and the skipping of exon 6; it thus lacks a region that is required for non-specific binding to plasma membranes. The two isoforms, expressed as fusion proteins with glutathione-S-transferase, interact in vitro with human syntaxin 6, thus retaining the specific protein interaction required for membrane fusion. Alterations in the SNAP23 gene might be involved in neurological and other diseases with defects in vesicle-membrane fusion processes that map to 15q15-21.  相似文献

Oxidative stress in biological membranes can regulate various aspects of apoptosis, including phosphatidylserine (PS) externalization. It is not known, however, if the targets for these effects are lipids or proteins. Nitric oxide (NO), a bifunctional modulator of apoptosis, has both antioxidant and prooxidant potential. We report here that the NO donor PAPANONOate completely protected all phospholipids, including PS, from oxidation in HL-60 cells treated with 2,2'-azobis(2,4-dimethylisovaleronitrile) (AMVN), presumably via the ability of NO to react with lipid-derived peroxyl radicals and terminate the propagation of lipid peroxidation. PAPANONOate, however, had no effect on PS externalization or other markers of apoptosis following AMVN. Therefore, PS oxidation is not required for PS externalization during AMVN-induced apoptosis. PS externalization was accompanied by inhibition of aminophospholipid translocase (APT). NO potentiated AMVN inhibition of APT. Treatment with PAPANONOate alone produced modest (20%) inhibition of APT without PS externalization. NO did not reverse AMVN-induced oxidation of glutathione and protein thiols. We speculate that APT was sensitive to AMVN and/or NO via modification of protein thiols critical for functional activity. Therefore, the lipoprotective effects of NO were insufficient to prevent PS externalization and apoptosis following oxidative stress. Other targets such as protein thiols may be important redox-sensitive regulators of apoptosis initiation and execution. Thus, in the absence of significant peroxynitrite formation, NO's antioxidant effects are restricted to protection of lipids, while modification of protein substrates continues to occur.  相似文献

This report describes structural and kinetic properties of the purified α-galactosidase from Saccharomyces carlsbergensis. This galactosidase has many similar properties to other exocellular enzymes in yeast which have been reported. Its molecular weight of 300,000 is comparable; it has similar carbohydrate content (57%) and amino acid and carbohydrate composition. That is, 35% of its amino acid residues can be accounted for by threonine, serine, and aspartic acid. Its carbohydrate composition is primarily mannose (90–95%) with approximately 7% glucose and 1% glucosamine. The enzyme is very stable to both acidic and alkaline conditions as well as to heating to 50 °C. α-Galactosidase remains active after incubation with as much as 1% sodium dodecyl sulfate at 30 °C. However, the enzyme is denatured with urea and guanidine hydrochloride. The loss of activity is proportional to the urea concentration, the nondenatured enzyme being responsible for the remaining activity. Inactivation by urea is partially reversible. With urea or 60 °C heat denaturation, the enzyme dissociates into two types of subunits as revealed by polyacrylamide gel electrophoresis with sodium dodecyl sulfate. Thus, α-galactosidase is the first external enzyme from yeast in which an oligomeric structure is reported. The enzyme catalyzes the hydrolysis of p-nitrophenyl-α-d-galactoside, melibiose, and raffinose with similar pH optima and Vmax. However, the affinity is 20-fold lower for raffinose than for the other two substrates. Sugars having the same configuration in carbons 2, 3, and 4 as galactose competitively inhibit the enzyme.  相似文献

Serpulids and other related tube-dwelling polychaetes are often ignored when found as fossil remains. They are, however, a widespread and important group today, and abundant literature has been published on them. Knowledge of fossil serpulids is centered on European material, and little has already been done on South American fossil calcareous tubes. In this paper, seven serpulid and sabellid morphotypes are described from the Early Cretaceous marine Agrio Formation of Argentina, revealing a diversity of worms recorded as encrusters on bivalves, ammonites and corals. Sabellids are represented by Glomerula cf. serpentina. Serpulids are represented by two subfamilies: “Serpulinae” includes Mucroserpula mucroserpula, Parsimonia antiquata, Placostegus cf. conchophilus, Propomatoceros semicostatus and P. sulcicarinatus; Spirorbinae is represented by heavily worn tiny coiled tubes assigned to ?Neomicrorbis. Serpulids and sabellids are one of the main components of the mollusk-encrusting fauna recorded in the Agrio Formation, along with small oysters but much more diverse. They are most commonly found as post-mortem encrusters, but some cases of unquestionable living interaction are also found, such as serpulid tubes embedded on coral branches. They are often overgrown by bryozoans, and sometimes by oysters; they frequently occur aggregated.  相似文献

Neurons are highly polarized cells that critically depend on long‐range, bidirectional transport between the cell body and synapse for their function. This continual and highly coordinated trafficking process, which takes place via the axon, has fascinated researchers since the early 20th century. Ramon y Cajal first proposed the existence of axonal trafficking of biological material after observing that dissociation of the axon from the cell body led to neuronal degeneration. Since these first indirect observations, the field has come a long way in its understanding of this fundamental process. However, these advances in our knowledge have been aided by breakthroughs in other scientific disciplines, as well as the parallel development of novel tools, techniques and model systems. In this review, we summarize the evolution of tools used to study axonal transport and discuss how their deployment has refined our understanding of this process. We also highlight innovative tools currently being developed and how their addition to the available axonal transport toolkit might help to address key outstanding questions.  相似文献
