We have studied the response of the interphase and mitotis microtubule arrays in root meristem cells of spring and winter cultivars of wheat Triticum aestivum L. (Moskovskaya 35 and Moskovskaya 39) during cold stress (1 h at 0 degrees C) and acclimation to cold (3-48 h at 0 degrees C). Our data show that interphase microtubules are more resistant to cold than mitotic arrays in both cultivars. During cold stress the density of endoplasmic microtubules increases in interphase cells of winter plants, yet no changes are detected in cells of spring plants. In mitotic cells of both wheat cultivars the density of microtubules within the kinetochore fibers decreases, yet this effect is more evident in the cells of spring plants. During acclimation to cold of both cultivars, we have observed the disorganization of the interphase cortical arrays and the enhanced growth of endoplasmic microtubule arrays, composed of microtubule converging centers. However, the reaction of mitotic microtubule arrays differs in the cells of winter and spring plants. In winter plants, during prophase diffuse tubulin "halo" accumulates first at perinuclear area, followed by the appearance of the microtubule converging centers. In spring plants, we have observed the formation of the prophase spindle, yet later the prophase spindle is not detected. Metaphase cells of both cultivars show similar aberrations of the mitotic spindle, accumulation of abnormal metaphases and the excessive formation of microtubule converging centers. In telophase cells of both cultivars, acclimation induces similar reaction, resulting in the disorganization of the phragmoplast and the formation of multiple microtubule converging centers. The latter are detected in the perinuclear areas of the daughter cells in winter plants and in the cortical cytoplasm of cells in spring plants. Our data point to the common pathways of microtubule response to cold treatment (0 degrees C). The excessive formation of the microtubule converging centers indicates the activation of microtubule assembly during prolonged cold treatment. 相似文献

Male Wistar rats with different types of behavior in "emotional resonance" test ("active" and "passive") were studied one week after the global ischemia induced by cardiac arrest. Recovery of some physiological functions as well as free-radical-mediated processes and NO-synthase activity were studied in cerebral structures and blood serum. The "open-field" behavior normalized more rapidly in the "active" rats than in the "passive" ones, though the time course of the neurologic deficit compensation did not differ in these groups. A decrease in superoxide scavenging activity and in the content of 2-thiobarbituric acid-reactive material was revealed in the cerebral structures of both "active" and "passive" rats. Increased levels of free-radical generation in the hippocampus of the "passive" rats and in the cerebellum of the "active" rats were found. Higher NO-synthase activity was demonstrated in the cerebellum of the "passive" rats. Taken together, these data suggest that there are specific patterns of free-radical-mediated processes in the brain of rats with different types of behavior in "emotional resonance" test. 相似文献

In acute experiments with narcotized and paralyzed cats, we studied responses of 74 striate neurons to cross-like figure under synchronous and asynchronous presentation of its lines. The aim of the study was to characterize the temporal course of interaction between three RF zones: main excitatory, end-inhibitory, and side disinhibitory ones. Previously we have found that this interaction is responsible for sensitivity to a cross in near 3/4 of cat striate cells with such sensitivity. In neurons with sensitivity to a cross, we found two types of temporal interaction between zones of RF. In the 1st type cells (14/23), the response significantly increased if the disinhibitory and the main excitatory zones of RF were stimulated simultaneously. Neurons of the 2nd type (9/23) revealed opposite temporal function: synchronous activation of RF zones evoked a minimal response. Simulation shows that the 1st type of behavior is connected with disinhibitory mechanism, while that of the 2nd type--with combination of this mechanism with convergence of orientation detectors of the previous functional level. 相似文献

Dynamics of tuning to the shape of cross-like figure flashed in receptive field was studied in 83 striate neurons by the method of temporal slices. Tuning was estimated by the total number of spikes in the response and by this number in successive fragments of the response with 20 ms steps. It was found that only in 11.7% cases neurons showed stable tuning to the same shape of the preferred figure (an angle between its lines), in other cases (88.3%) during response generation this tuning changes being one-phase (7.2%) or two-phase (27.0%), or undulatory (54.1%). Different dynamical reorganization of receptive field zones is discussed as a possible mechanism of the revealed effects as well as their correlation with previously described dynamics of tuning to orientation of a single bar and a cross in striate cells. 相似文献

Hidden Markov models (HMMs) are a class of stochastic models that have proven to be powerful tools for the analysis of molecular sequence data. A hidden Markov model can be viewed as a black box that generates sequences of observations. The unobservable internal state of the box is stochastic and is determined by a finite state Markov chain. The observable output is stochastic with distribution determined by the state of the hidden Markov chain. We present a Bayesian solution to the problem of restoring the sequence of states visited by the hidden Markov chain from a given sequence of observed outputs. Our approach is based on a Monte Carlo Markov chain algorithm that allows us to draw samples from the full posterior distribution of the hidden Markov chain paths. The problem of estimating the probability of individual paths and the associated Monte Carlo error of these estimates is addressed. The method is illustrated by considering a problem of DNA sequence multiple alignment. The special structure for the hidden Markov model used in the sequence alignment problem is considered in detail. In conclusion, we discuss certain interesting aspects of biological sequence alignments that become accessible through the Bayesian approach to HMM restoration. 相似文献

Orientation tuning of 148 primary visual cortical neurons was studied in acute experiments on unanesthetized, curarized cats by analysis of their spike responses to flashes in a receptive field of a bar of light of optimal size. Orientation tuning of 88 neurons (59%) was found to be bimodal: Besides the principal preferred orientation there was a second, making an angle with the first. The second tuning maximum in some cases (64%) was exhibited only with a change in stimulus intensity or background brightness. Analysis of orientation tuning by the time-slice method, i.e., on the basis of individual cuts of the spike trace, showed double tuning to be present in 69% of cases only at certain moments after the beginning of stimulation. The results of analysis of the model showed that the double orientation tuning effect may be the result of the specific configuration of the receptive field, the use of a stimulus longer than the receptive field, the presence of a series of alternating excitatory and inhibitory zones in the receptive field, and also of end inhibitory zones on the narrow ends of the field. The unequal change in zones of the receptive fields in time explains the appearance of double orientation tuning in individual fragments of the spike trace. The functional role of double, "cross-wise" tuning in some primary visual cortical neurons and their role in the detection of the features of visual patterns are discussed. 相似文献

Children categorize stimuli at the basic level faster than at the superordinate level. We hypothesized that between-category similarity may affect this basic level superiority effect. Dissimilar categories may be easy to distinguish at the basic level but be difficult to group at the superordinate level, whereas similar categories may be easy to group at the superordinate level but be difficult to distinguish at the basic level. Consequently, similar basic level categories may produce a superordinate-before-basic learning trend, whereas dissimilar basic level categories may result in a basic-before-superordinate learning trend. We tested this hypothesis in pigeons by constructing superordinate level categories out of basic level categories with known similarity. In Experiment 1, we experimentally evaluated the between-category similarity of four basic level photographic categories using multiple fixed interval-extinction training (Astley and Wasserman, 1992). We used the resultant similarity matrices in Experiment 2 to construct two superordinate level categories from basic level categories with high between-category similarity (cars and persons; chairs and flowers). We then trained pigeons to concurrently classify those photographs into either the proper basic level category or the proper superordinate level category. Under these conditions, the pigeons learned the superordinate level discrimination faster than the basic level discrimination, confirming our hypothesis that basic level superiority is affected by between-category similarity. 相似文献

The changes in the cAMP level during the cell cycle in the synchronous cultures of E. coli were demonstrated. Two maxima in the cAMP level were revealed during each generation period. In the cell cycle of 40-45 min duration the first increase was observed approximately in the middle of the cycle, i. e., it was coincident with the initiation of DNA synthesis. Under these conditions the cAMP level increased 8-10 times (from 0.5 to 5.0 pmole per ml of cell suspension). Rats received intracerebroventricular injections of z-DEVD-FMK (caspase-3 inhibitor) or z-FA-FMK (control peptide) in a dose of 3 nmol. Administration of z-DEVD-FMK significantly decreased the number of avoidance reactions in some blocks of trials in active avoidance (shuttle box) learning. However, only slight effect of the caspase inhibitor across the session was found. Z-DEVD-FMK impaired development of some essential components of the two-way active avoidance performance, such as escape reaction, conditioned fear reaction, and inter-trial crossings. Z-DEVD-FMK did not impair working memory in the spontaneous alternation behavior paradigm. Z-DEVD-FMK affected neither emotionality nor locomotor activity in the open-field test. It also did not influence behavior in the light-dark chamber. Measurement of caspase-3 activity in rat brain regions involved in active avoidance learning revealed z-DEVD-FMK-related inhibition of the enzyme activity most pronounced (about 30%) in the fronto-parietal cortex; a similar effect was close to significant in the hippocampus.
