To date, no genome of any of the species from the genus Spiroplasma has been completely sequenced. Long repetitive sequences similar to mobile units present a major obstacle for current genome sequencing technologies. Here, we report the assembly of the Spiroplasma melliferum KC3 genome into 4 contigs, followed by proteogenomic annotation and metabolic reconstruction based on the discovery of 521 expressed proteins and comprehensive metabolomic profiling. A systems approach allowed us to elucidate putative pathogenicity mechanisms and to discover major virulence factors, such as Chitinase utilization enzymes and toxins never before reported for insect pathogenic spiroplasmas.  相似文献

It has been demonstrated by the methods of histochemical and biochemical examination of the activity of the enzymes that the mucus layer covering the small intestinal wall contains active enzymes (alkaline phosphatase, leucin aminopeptidase IV, saccharase, lactase) and pancreatic enzymes (alpha-amylase and trypsin). Emphasis is laid on the enrichment of the mucus layer with pancreatic enzymes as compared with small intestinal juice. A hypothesis has been advanced according to which the mucus layer undergoes degradation of polymeric and oligomeric substrates, which plays a physiological part in the digestion of nutritive substances and protection of the internal medium against immunoactive biopolymers. The digestion occurring in the mucus layer is proposed to be called mucus digestion.  相似文献

Adrenal glands of rats were immersed in Ringer-Lock solution (used as control), other rat adrenal glands were immersed in human blood plasma. The amount of corticosterone in the extracts received was estimated after two hours of incubation. The activity of adrenocorticotropic function of the pituitary gland of the patients examined was judged by the difference in the corticosterone indices in these extracts. ACTH (1.5 and 10 mkg) was added to the plasma to determine the sensitivity of this technique. Regular and significant increase in corticosterone was revealed. Modified Pierre's technique can be applied for ACTH determination in man.  相似文献

A considerable increase in several heat shock proteins (HSPs) amount in Acholeplasma laidlawii cells has been revealed after temperature rising of liquid culture; and the quantity of small HSP, named p17, was increased in a hundred of times. The p17 protein was isolated and identified as HSP of alpha-crystallin type (alpha-HSP). It became possible as a result of sequencing of 15 amino acids from N-terminal of the p17 polypeptide chain, followed by revealing of a corresponding open reading frame (ORF) in a completely sequenced genome of A. laidlawii PG 8A. Computer-based search for homologous ORFs in all 17 genomes of Mycoplasmataceae family (the mycoplasmas themselves) that had been completely sequenced to date, gives negative result. But among the representatives of Mollicutes (mycoplasma) class, the genes coding alpha-HSPs were found in two Phytoplasma species (Phytoplasmataceae family) and the acholeplasma examined (Acholeplasmataceae family). It supposed that presence or absence of alpha-HSPs in microorganisms might be connected with the fact that representatives of Acholeplasmataceae and Phytoplasmataceae families inhabit principally in plant tissues in contrast to majority of Mycoplasmataceae family, that inhabit animal and human tissues, i. e. use ecological niches with relatively constant temperature.  相似文献

The invasiveness of 2 grud sensitive and 8 holy-resistant M. tuberculosis clinical isolated was evaluated in experiments on BALB/c mice. Mycobacterial suspension was injected into the caudal vein of experimental animals. The results were evaluated by the degree of contamination of lungs and spleen of infected animals euthanized at different periods on time. The study revealed high variability in the degree of contamination of the organs of the animals infected with M. tuberculosis drug-resistant clinical isolates.  相似文献

The in vivo action of the antimicrobial peptide melittin, expressed from a recombinant plasmid vector, on chickens experimentally infected with Mycoplasma gallisepticum was studied. The plasmid vector pBI/mel2/rtTA includes the melittin gene under the control of an inducible tetracycline-dependent human cytomegalovirus promoter and the gene coding for the trans-activation protein rtTA. Aerosol administration of the vector, followed by infecting the chickens with M. gallisepticum 1226, is shown to inhibit development of infection. The inhibitory action was confirmed by a complex of clinical, pathomorphological, histological and serological studies, and also by comparing the M. gallisepticum reisolation frequency from the respiratory tract and internal organs. The data suggest that plasmid vectors expressing genes of antimicrobial peptides can be considered as potential agents for the prevention and treatment of mycoplasma infections in poultry farming.  相似文献

The formation of a peaked bell-shaped profile of the electron density n e (r) in the T-11M tokamak (B t=1 T, R/a = 0.7/0.2 m, I p = 100 kA, t shot ≤ 300 ms, Li and C limiters) was observed in Li experiments carried out in the near-plateau collisionality regime (the collisionality parameter at one-half of the minor radius was v* ≥ 0.5) under the conditions of low hydrogen recycling and intense hydrogen influx from the plasma edge. It is well known that peaked n e (r) profiles are observed in collisionless regimes at v* values as low as 10?1–10?2 or in impurity-contaminated discharges, in which this effect can be attributed to the impurity accumulation on the plasma column axis. Moreover, a bell-shaped n e (r) profile in discharges with low n e can result from the ionization of hydrogen atoms at the column axis, where they arrive from the plasma edge due to cascade charge-exchange. In quasi-steady lithium discharges in T-11M, however, peaked n e (r) profiles were observed at a relatively high central electron density n e (0) and relatively high collision frequency, such that the influence of impurities on the n e (r) profile could be ignored (Z eff = 1.1±0.1). To explain this effect, one has to assume that the pinching of hydrogen ions in T-11M is anomalous. The lower estimate of the observed pinch velocity is 4 ± 1 m/s, which is three to five times higher than the velocity of the neoclassical (Ware) pinch, characteristic of these conditions. The work is devoted to the experimental study of this effect.  相似文献
