The muscularis mucosae, a type of smooth muscle located between the urothelium and the urinary bladder detrusor, has been described, although its properties and role in bladder function have not been characterized. Here, using mucosal tissue strips isolated from guinea pig urinary bladders, we identified spontaneous phasic contractions (SPCs) that appear to originate in the muscularis mucosae. This smooth muscle layer exhibited Ca(2+) waves and flashes, but localized Ca(2+) events (Ca(2+) sparks, purinergic receptor-mediated transients) were not detected. Ca(2+) flashes, often in bursts, occurred with a frequency (~5.7/min) similar to that of SPCs (~4/min), suggesting that SPCs are triggered by bursts of Ca(2+) flashes. The force generated by a single mucosal SPC represented the maximal force of the strip, whereas a single detrusor SPC was ~3% of maximal force of the detrusor strip. Electrical field stimulation (0.5-50 Hz) evoked force transients in isolated detrusor and mucosal strips. Inhibition of cholinergic receptors significantly decreased force in detrusor and mucosal strips (at higher frequencies). Concurrent inhibition of purinergic and cholinergic receptors nearly abolished evoked responses in detrusor and mucosae. Mucosal SPCs were unaffected by blocking small-conductance Ca(2+)-activated K(+) (SK) channels with apamin and were unchanged by blocking large-conductance Ca(2+)-activated K(+) (BK) channels with iberiotoxin (IbTX), indicating that SK and BK channels play a much smaller role in regulating muscularis mucosae SPCs than they do in regulating detrusor SPCs. Consistent with this, BK channel current density in myocytes from muscularis mucosae was ~20% of that in detrusor myocytes. These findings indicate that the muscularis mucosae in guinea pig represents a second smooth muscle compartment that is physiologically and pharmacologically distinct from the detrusor and may contribute to the overall contractile properties of the urinary bladder.  相似文献

Nutritional restrictions in winter may reduce the availability of protein for reproduction and survival in northern ungulates. We refined a technique that uses recently voided excreta on snow to assess protein status in wild caribou (Rangifer tarandus) in late winter. Our study was the first application of this non-invasive, isotopic approach to assess protein status of wild caribou by determining dietary and endogenous contributions of nitrogen (N) to urinary urea. We used isotopic ratios of N (δ15N) in urine and fecal samples to estimate the proportion of urea N derived from body N (p-UN) in pregnant, adult females of the Chisana Herd, a small population that ranged across the Alaska-Yukon border. We took advantage of a predator-exclosure project to examine N status of penned caribou in April 2006. Lichens were the primary forage (>40%) consumed by caribou in the pen and δ15N of fiber tracked the major forages in their diets. The δ15N of urinary urea for females in the pen was depleted relative (−1.3 ± 1.0 parts per thousand [‰], ) to the δ15N of body N (2.7 ± 0.7‰). A similar proportion of animals in the exclosure lost core body mass (excluding estimates of fetal and uterine tissues; 55%) and body protein (estimated by isotope ratios; 54%). This non-invasive technique could be applied at various spatial and temporal scales to assess trends in protein status of free-ranging populations of northern ungulates. Intra- and inter-annual estimates of protein status could help managers monitor effects of foraging conditions on nutritional constraints in ungulates, increase the efficiency and efficacy of management actions, and help prepare stakeholders for potential changes in population trends. © 2010 The Wildlife Society.  相似文献

Identifying patterns of fine-scale genetic structure in natural populations can advance understanding of critical ecological processes such as dispersal and gene flow across heterogeneous landscapes. Alpine ungulates generally exhibit high levels of genetic structure due to female philopatry and patchy configuration of mountain habitats. We assessed the spatial scale of genetic structure and the amount of gene flow in 301 Dall’s sheep (Ovis dalli dalli) at the landscape level using 15 nuclear microsatellites and 473 base pairs of the mitochondrial (mtDNA) control region. Dall’s sheep exhibited significant genetic structure within contiguous mountain ranges, but mtDNA structure occurred at a broader geographic scale than nuclear DNA within the study area, and mtDNA structure for other North American mountain sheep populations. No evidence of male-mediated gene flow or greater philopatry of females was observed; there was little difference between markers with different modes of inheritance (pairwise nuclear DNA F ST = 0.004–0.325; mtDNA F ST = 0.009–0.544), and males were no more likely than females to be recent immigrants. Historical patterns based on mtDNA indicate separate northern and southern lineages and a pattern of expansion following regional glacial retreat. Boundaries of genetic clusters aligned geographically with prominent mountain ranges, icefields, and major river valleys based on Bayesian and hierarchical modeling of microsatellite and mtDNA data. Our results suggest that fine-scale genetic structure in Dall’s sheep is influenced by limited dispersal, and structure may be weaker in populations occurring near ancestral levels of density and distribution in continuous habitats compared to other alpine ungulates that have experienced declines and marked habitat fragmentation.  相似文献

Background

The impact of the intrauterine environment on the developmental programming of adult female reproductive success is still poorly understood and potentially underestimated. Litter size variation in a nonhuman primate, the common marmoset monkey (Callithrix jacchus), allows us to model the effects of varying intrauterine environments (e.g. nutrient restriction, exposure to male womb-mates) on the risk of losing fetuses in adulthood. Our previous work has characterized the fetuses of triplet pregnancies as experiencing intrauterine nutritional restriction.

Methodology/Principal Findings

We used over a decade of demographic data from the Southwest National Primate Research Center common marmoset colony. We evaluated differences between twin and triplet females in the number of pregnancies they produce and the proportion of those pregnancies that ended in fetal loss. We found that triplet females produced the same number of total offspring as twin females, but lost offspring during pregnancy at a significantly higher rate than did twins (38% vs. 13%, p = 0.02). Regardless of their own birth weight or the sex ratio of the litter the experienced as fetuses, triplet females lost more fetuses than did twins. Females with a male littermate experienced a significant increase in the proportion of stillbirths.

Conclusions/Significance

These striking findings anchor pregnancy loss in the mother’s own fetal environment and development, underscoring a "Womb to Womb" view of the lifecourse and the intergenerational consequences of development. This has important translational implications for understanding the large proportion of human stillbirths that are unexplained. Our findings provide strong evidence that a full understanding of mammalian life history and reproductive biology requires a developmental foundation.  相似文献

1. Eutrophic acid lakes are not common. Delamere Lake in Cheshire, U.K. is shallow and acid (mean pH 4.5) with a very high phytoplankton crop (mean 290 μg chlorophyll a L?1), dominated by Dictyosphaerium pulchellum. Rotifers were dominant in the pelagic waters but small cladocerans (Alona guttata, Chydorus sphaericus and Scapholeberis mucronata) were occasional in the littoral waters. Chaoborus flavicans larvae were the top predators in this fishless lake. Two mesocosm experiments were carried out in which pH and Chaoborus populations were manipulated. 2. Progressively higher concentrations of D. pulchellum were maintained in the elevated pH treatments (pH 6 and 8; P < 0.001) with increased amounts of a Chlamydomonas species at the end of the experiment. Highest species richness was seen at ambient pH. Thus the low pH of Delamere Lake alone did not control the structure of the phytoplankton community. Keratella quadrata showed significantly higher abundance at pH 6 than in other pH treatments (P < 0.001). Species richness of rotifers was unaffected by pH. 3. Most Cladocera were C. sphaericus. Although never seen in the open lake, Daphnia pulex appeared in all the pH treatments. Low pH did not control small Cladocera abundance in Delamere Lake, but probably hampered reproduction in Daphnia. Negative correlations between chlorophyll a concentrations and Daphnia in the mesocosms (r2 = 0.215, P < 0.05), however, indicated the potential of large‐bodied daphniids in controlling phytoplankton. 4. Neither different combinations of Chaoborus instars (none, instars 1 and 2 and instars 3–5) nor different densities of instars 3–5 (0.15, 0.5 and 1.0 L?1) had a negative impact on Cladocera. Daphnia pulex remained unaffected in the experiment, perhaps because of its large size, and C. sphaericus because of its high reproductive rate compensating predatory losses. 5. Very low pH in Delamere Lake might suppress Daphnia by hampering its reproduction. Consequently, Daphnia may be vulnerable to invertebrate predation even at low predator density in the lake.  相似文献

Wolf packs generally consist of a breeding pair and their maturingoffspring that help provision and protect pack young. Becausethe reproductive tenure in wolves is often short, reproductivelymature offspring might replace their parents, resulting in siblingor parent-offspring matings. To determine the extent of incestuouspairings, we measured relatedness based on variability in 20microsatellite loci of mated pairs, parent-offspring pairs,and siblings in two populations of gray wolves. Our 16 sampledmated pairs had values of relatedness not overlapping thoseof known parent-offspring or sibling dyads, which is consistentwith their being unrelated or distantly related. These resultssuggest that full siblings or a parent and its offspring rarelymate and that incest avoidance is an important constraint ongray wolf behavioral ecology  相似文献
