Systemic acquired resistance (SAR) is a broad-spectrum, systemic defense response that is activated in many plant species after pathogen infection. We have previously described Arabidopsis mutants that constitutively express SAR and concomitantly develop lesions simulating disease (lsd). Here, we describe two new mutants, lsd6 and lsd7, that develop spontaneous necrotic lesions and possess elevated levels of salicylic acid (SA) as well as heightened disease resistance, similar to the previously characterized lsd and accelerated cell death (acd2) mutants. Genetic analysis of lsd6 and lsd7 showed that the mutant phenotypes segregated as simple dominant traits. When crossed with transgenic Arabidopsis plants containing the SA-degrading enzyme salicylate hydroxylase, the F1 progeny showed suppression of both SAR gene expression and resistance. In addition, salicylate hydroxylase suppressed lesion formation in the F1 progeny, suggesting that SA or some SA-dependent process may have a role in pathogen-associated cell death. Surprisingly, lesions were restored in the lsd6 F1 progeny after the application of either 2,6-dichloroisonicotinic acid or SA. Lesions were not restored by treatment with either compound in the lsd7 F1 plants. Our findings demonstrate that steps early in the signal transduction pathway leading to SAR and disease resistance are potentiated by later events, suggesting feedback control of lesion formation. 相似文献

Several patterns in the distribution and abundance of organisms have now been documented. They include broad (but not universal) positive correlations between range sizes and population abundances; a decline in the proportion of sites occupied and in average population densities from the centre to the edge of a species' range, with either unimodal or multimodal peaks of abundance and occupancy in the core of the range; and intriguing, but still poorly documented phylogenetic effects on both range size and abundance. All these patterns require further work to establish their generality, and all of them lack generally agreed explanations. They are important, however, not only theoretically but also practically, because of the constraints and opportunities they appear to provide for the management and conservation of species. 相似文献

To clarify the role of ethylene in systemic acquired resistance (SAR), we conducted experiments using Arabidopsis ethylene response mutants. Plants that are nonresponsive to ethylene (i.e., [theta]tr1 and [theta]in2) showed normal sensitivity to the SAR-inducing chemicals salicylic acid (SA) and 2,6-dichloroisonicotinic acid with respect to SAR gene induction and pathogen resistance. This indicated that chemically induced SAR is not an ethylene-dependent process in Arabidopsis. Ethephon, an ethylene-releasing chemical, induced SAR gene expression in both the wild type and ethylene mutants, whereas ethylene alone did not, suggesting that induction of these genes by ethephon is not due to the action of ethylene. Furthermore, transgenic plants expressing salicylate hydroxylase, a bacterial enzyme that degrades SA to catechol, did not accumulate SAR mRNAs in response to ethephon. Thus, SAR gene induction by ethephon appears to be mediated through SA. Other experiments suggested that ethylene may play a role in SAR by enhancing tissue sensitivity to the action of SA. 相似文献

Theoretically, there are three principal ways in which ecosystem processes might respond to reductions in species richness. These theories are reviewed, and then considered in the context of a study of the diversity of soil nematodes and termites in near-primary forest sites at Mbalmayo, Cameroon, and the contribution made by these two taxa to carbon fluxes (CO2 and CH4) from the forest floor. Nematode abundances average 2.04 × 106 m-2, and termites between 2933 and 6957 m-2. The site is the most species-rich yet investigated for both groups anywhere in the world, so that a very large number of species contribute to carbon fluxes. We speculate about how much redundancy might be built into the functioning of both assemblages, and point out the enormous difficulties of resolving such questions, and of producing such detailed species-inventories. 相似文献

Benzothiadiazole (BTH) is a novel chemical activator of disease resistance in tobacco, wheat and other important agricultural plants. In this report, it is shown that BTH works by activating SAR in Arabidopsis thaliana. BTH-treated plants were resistant to infection by turnip crinkle virus, Pseudomonas syringae pv ‘tomato’ DC3000 and Peronospora parasitica. Chemical treatment induced accumulation of mRNAs from the SAR-associated genes, PR-1, PR-2 and PR-5. BTH treatment induced both PR-1 mRNA accumulation and resistance against P. parasitica in the ethylene response mutants, etr1 and ein2, and in the methyl jasmonate-insensitive mutant, jar1, suggesting that BTH action is independent of these plant hormones. BTH treatment also induced both PR-1 mRNA accumulation and P. parasitica resistance in transgenic Arabidopsis plants expressing the nahG gene, suggesting that BTH action does not require salicylic acid accumulation. However, because BTH-treatment failed to induce either PR-1 mRNA accumulation or P. parasitica resistance in the non-inducible immunity mutant, nim1, it appears that BTH activates the SAR signal transduction pathway. 相似文献

Concanavalin A (Con A) stimulated the IgM-binding subpopulation of human T cells (TM) to suppress the pokeweed mitogen-induced differentiation of B lymphocytes to plasma cells. Control TM cells that had not been Con A activated did not suppress. The degree of suppression was related to the number of Con A-TM cells added to the cultures and it was abolished by irradiation of the T lymphocytes either before or after the 24-hr culture period with Con A. Suppression did not require the presence of TG cells, whose suppressor potential has been previously established. These findings indicate that suppressor activity is not confined to the TG subpopulation but may be expressed by TM cells also. 相似文献

Chronic treatment of mice from birth with anti-mu antibodies aborts development of B lymphocytes and plasma cells. In these studies we show that bone marrow from anti-mu-treated mice contains a population of cells with cytoplasmic IgM, but which lack detectable cell-surface IgM. These cells are analogous to pre-B cells, defined in ontogenetic studies as the immediate precursors of B lymphocytes. Pre-B cells from bone marrow of anti-mu treated mice retain their functional integrity, as evidenced by their ability to give rise to sIgM+, LPS-responsive lymphocytes in culture. We also show that cyclophosphamide treatment destroys pre-B cells and that recovery of pre-B cells in bone marrow precedes the regeneration of sIgM+ B lymphocytes. Generation of B lymphocytes in adult mice apparently occurs exclusively in the bone marrow because induction of extramedullary hemopoiesis in spleen was not accompanied by the appearance of pre-B cells in that organ. 相似文献

A previously developed immobilization technique involving latex coatings on solid particulate supports was investigated further for penicillin G production by Penicillium chrysogenum. Several modifications were found to decrease the germination lag time, including a higher spore concentration, a thinner latex layer, an increased latex porosity, and a decreased drying time. This approach enabled the development of immobilized mycelial pellets within 2-3 days from the onset of biocatalyst preparation and incubation.A continuous immobilized-cell airlift bioreactor produced penicillin G in a series of runs in which the production phase lasted up to 30 days. The productivity of this system was 3-6 times greater than the productivity of the corresponding free-cell shake flask fermentation. 相似文献
