Abstract. The concentrations of putrescine, spermidine and spermine, the only polyamines detectable in normal and habituated calli of Beta vulgaris L. ssp. altissima , were much higher in the habituated callus than the normal callus, irrespective of experimental conditions. These results suggest that, in normal (tolerant to NaCl) and habituated (sensitive to NaCl) calli, there exists a competition for the common precursor of ethylene and polyamine biosynthesis viz. S-adenosylmethionine. A disequilibrium favouring the synthesis of putrescine and spermidine in the habituated callus might be linked to structural deterioration of the cell membrane following extended culture or severe osmotic stress (68 mol m−1 NaCl). The maintenance of membrane integrity by the normal callus coincides with ethylene production at the expense of polyamine synthesis. In contrast to the habituated callus, the salinity tolerance of the normal callus is accompanied by the accumulation of proline under hypersaline conditions (274mol m−3). The important osmoregulatory role played by quaternary ammonium compounds in the-aerial parts of Chenopodiaceae, especially the sugarbeet, is not observed in the calli, these compounds being found in very low concentrations in saline conditions.  相似文献

Epimastigotes of Trypanosoma cruzi, Peru strain, incubated in Contreras' artificial triatomine urine transformed into metacyclic trypomastigotes when 10 mM L-glutamine, L-asparagine or D-fructose was added to the medium. Metacyclogenesis with these substrates was comparable to the percent metacyclic morphotype formation induced by L-proline and significantly greater than that stimulated by 10 mM D-glucose. Sodium acetate (10 mM) increased transformation induced by L-proline, and L-hydroxyproline (10 mM) increased transformation induced by D-fructose. Phosphoenolpyruvate (10 mM) inhibited L-proline-induced metacyclic trypomastigote stage formation. Three antimetabolites, azetidine 2-carboxylate (5 mM), malonic acid (1 mM), and desthiobiotin (5 mM), completely inhibited D-fructose-induced but not L-proline-induced transformation. The Costa Rica, Y, and CL strains of T. cruzi showed different patterns of percent metacyclogenesis with substrates that induce transformation in the Peru strain.  相似文献

1. Climate is an important source of selection on life histories, and local adaptations to climate have been described in several cline studies. Temperature is the main climatic factor that has been considered as an agent of selection, whereas other factors may vary with it, such as precipitation. 2. We compared life‐history traits of five populations of Leptopilina boulardi, a Drosophila parasitoid, originating from contrasting climates. Referring to cline studies, we hypothesised shorter lifespan, earlier reproduction, and lower lipid content in populations from the hottest and driest areas if life histories have been selected in response to temperature and/or humidity. 3. Our results are opposite to these predictions. Females from humid and mild climates invested more in early reproduction and lived for fewer days than females from dry and hot areas, which were synovigenic (i.e. they matured additional eggs during adult life) and able to synthesise lipids during adult life. 4. We suggest that life histories are more adapted to host distribution than to climatic factors. Drosophila patches are more abundant in the humid area, allowing the parasitoids to spend less energy and time finding hosts. This may result in selection for early reproduction traded‐off against longevity. In the hot and dry climate, females have to fly large distances to find host patches. Synovigeny, a long lifespan, lipogenesis, and high dispersal ability may be adaptive there. This is the first time that between‐population differences in the ability to synthesise lipids have been described in parasitoids.  相似文献

By a combination of thin-layer chromatography and gas liquid chromatography, a complete study of the development of the different lipid classes and of their fatty acids, during the development of the fruit of Hedera helix L., the English Ivy, has been achieved. In any part of the fruit observed, at any particular stage, the phospholipids and the neutral lipids are the most abundant lipid classes. They accumulate during the entire process of maturation, whereas significant changes occur in their relative proportions, phospholipids being largely dominant until fruit blackening. The accumulation of fatty acids during maturation is characterized by large amounts of C18:1 in the neutral lipids, especially in the seed, where petroselinic acid (C18:1Δ6) reaches 86% of the total fatty acids. To a smaller extent, the phospholipids also accumulate and thus have the character of reserve molecules. However, their composition remains more stable, which relates them to the “structural lipids” such as galactolipids that maintain their characteristic fatty acid composition, despite the radical changes occurring in the fatty acid metabolism during fruit ripening.  相似文献

A survey of amplified fragment length polymorphism (AFLP) was conducted to examine genetic diversity in 146 individuals of British, native black poplar ( Populus nigra subsp. betulifolia ) and three individuals considered by collectors to be non- betulifolia poplars. Using two primer pairs, a total of 147 bands were detected of which 82 (56%) were polymorphic in at least one individual. Cluster analysis and principal coordinates analysis of the calculated similarity matrix revealed a low level of genetic diversity, although a loose clustering into five groups could be identified, one of which contained the non- betulifolia individuals. Examination of the spatial distribution of the other four groups (all betulifolia ) revealed a general correlation between geographic proximity and genetic similarity. On the basis of the polymorphism observed, it was possible to identify a small number of individual plants which exhibit maximum diversity and might therefore be suitable for inclusion in a replanting programme designed to maintain at least the existing low level of polymorphism observed within British black poplar.  相似文献

The activities of DNA polymerases α, β, and γ and of thymidine kinase were determined in the chick neural retina at different stages of embryonic development (starting at seven days) and after hatching (up to five years). Crude extracts of neural retinae were fractionated by centrifugation on sucrose gradients and the enzymatic activities measured using specific assays. The DNA polymerase a activity decreases greatly between 7 and 11 days of incubation. This decrease parallels the decline in mitotic activity. However, a constant residual activity remains after hatching, even in the oldest animals. DNA polymerase β activity increases slightly between 7 and 14 days of incubation; it then decreases slowly until seven days after hatching and remains constant thereafter. DNA polymerase γ activity is maximal between 7 and 14 days of incubation and then decreases until hatching. The activity of thymidine kinase increases slightly during the embryonic life until hatching and remains almost constant thereafter. The implication of these enzymes in DNA replication and repair processes is discussed.  相似文献
