A pollen profile from Marais du Sagnas at the northern edge of the Etang de Berre lagoon, west of Marseille, is compared and correlated with a profile (C10) from the southern margin of the lagoon. This new profile extends the pollen record for the area to recent times. The palaeoecological data now available indicate distinct phases of marine influence in the lagoon from about 900 A.D. (non-calibrated) to the 16/17 century. These correspond to historically known drier periods during which it appears that reduced freshwater input facilitated increased influx of saline water. 相似文献

Beta-blockers are used to treat acquired heart failure in adults, though their role in early muscular dystrophy cardiomyopathy is unclear. We treated 2 different dystrophic mouse models which have an associated cardiomyopathy (mdx: model for Duchenne Muscular Dystrophy, and Sgcd-/-: model for limb girdle muscular dystrophy type 2F) and wild type controls (C57 Bl10) with the beta blocker metoprolol or placebo for 8 weeks at an early stage in the development of the cardiomyopathy. Left and right ventricular function was assessed with cardiac magnetic resonance imaging (MRI) and in-vivo myocardial calcium influx with manganese enhanced MRI. In the mdx mice at baseline there was reduced stroke volume, cardiac index, and end-diastolic volume with preserved left ventricular ejection fraction. These abnormalities were no longer evident after treatment with beta-blockers. Right ventricular ejection fraction was reduced and right ventricular end-systolic volume increased in the mdx mice. With metoprolol there was an increase in right ventricular end-diastolic and end-systolic volumes. Left and right ventricular function was normal in the Sgcd-/- mice. Metroprolol had no significant effects on left and right ventricular function in these mice, though heart/body weight ratios increased after treatment. In-vivo myocardial calcium influx with MEMRI was significantly elevated in both models, though metoprolol had no significant effects on either. In conclusion, metoprolol treatment at an early stage in the development of cardiomyopathy has deleterious effects on right ventricular function in mdx mice and in both models no effect on increased in-vivo calcium influx. This suggests that clinical trials need to carefully monitor not just left ventricular function but also right ventricular function and other aspects of myocardial metabolism. 相似文献

The repair of 2,6-diamino-4-hydroxy-5-N-methyl-formamidopyrimidine (Fapy) residues in DNA is performed by a Fapy-DNA glycosylase activity which is encoded for by the fpg gene in Escherichia coli. Besides DNA glycosylase activity, this protein, the FPG protein, is endowed with an EDTA-resistant activity nicking DNA at apurinic/apyrimidinic (AP) sites. To overproduce the FPG protein, the fpg gene was placed under the control of the tac promoter in the expression vector pKK223-3 yielding the pFPG230 plasmid. The production of the FPG protein in cells harboring the pFPG230 plasmid was 800-fold higher than that of the wild type strain after induction by isopropyl-beta-D-thio-galactopyranoside. From these cells, the FPG protein was purified to homogeneity in sufficient quantity to study its physical and catalytic properties. In its active form, the FPG protein is a globular monomer of 31 kDa and has an experimentally measured isoelectric point of 8.5. When the FPG protein is heat-denatured in the presence of EDTA the two activities are more rapidly inactivated than when heated in the absence of EDTA, suggesting that the FPG protein possesses a tightly bound metal ion. Atomic absorption spectrophotometric analysis shows that there is one zinc/FPG protein molecule. The FPG protein is different from previously described DNA glycosylases and AP-nicking enzymes in E. coli. The contribution of the AP-nicking activity associated with the FPG protein represents 10-20% of the total EDTA-resistant AP-nicking activities in E. coli. 相似文献

The diploid number of Phronima sedentaria and P. atlantica is 30, all the chromosomes are metacentric or submetacentric; the caryotypes of these two species are compared with those of other Amphipods. Electrochemical regeneration of NAD was performed in a bench scale reactor in which yeast alcohol dehydrogenase catalyzed the oxidation of ethanol. By recycling one of the products of the reaction, it was possible to displace the equilibrium and favor the production of acetaldehyde. The flow-through electrode was made of graphite felt and had a specific area of 275 cm(-1). A mathematical model taking into account the enzymatic and electrochemical reaction rates as well as the mass transfer to the electrode was used to analyze the results. An actual yield of 99.97% (i.e., 3000 potential regenerating cycles) has been obtained for the electrochemical regeneration of NAD(+). The deactivation of the coenzyme is not related to the number of cycles; it is only time dependent. A reactor, using the enzymatic electrocatalysis scheme, was developed on a laboratory preparative scale for the catalytic oxidation of glucose into gluconic acid. Glucose oxidase was directly immobilized on the surface of a carbon felt electrode and a solution of glucose and benzo-quinone passed through the electrode in order to regenerate the electron acceptor. The reactor was able to produce continuously 1.5 g gluconate/h with a catalytic current of 0.4 A. This gave a high productivity ca. 100 g/h/L reactor. A one-dimensional model was developed which demonstrated the efficiency of coupling between enzymatic and electrochemical reactions due to the proximity of the two reaction sites. For example the catalytic current was practically independent of mass transfer parameters. Two chromatographically distinct endonucleases from Micrococcus luteus, specific for apurinic and apyrimidinic sites (AP-endonucleases A and B), have been extensively purified and characterized. Both are free from DNA glycosylase, unspecific endonuclease, and phosphatase activities. The two enzymes behave as monomeric proteins of approximately 35000 daltons. In addition to their different chromatographic properties on CM-cellulose, P-cellulose, hydroxylapatite, and DNA--Sepharose, both AP-endonucleases can be distinguished as follows: AP-endonuclease A has an isoelectric point of 4.8, shows a half-life of 4 min at 45 degrees C, reacts optimally at pH 7.5 and has a KM value of 2.3 X 10(-6) M. AP-endonuclease B has a pI of 8.8, is more stable at 45 degrees C (half-life of 10 min), and reacts optimally between pH 6.5 and pH 8.5; its KM value is 3.7 X 10(-6) M. 相似文献

Variations of the pancreatic parenchyma, the gastric mucosa and the intestinal mucosa were studied in adult male Wistar rats on day 8 and 15 after hypophysectomy. All results were compared with those obtained in pair-fed control rats. Hypophysectomy affected small intestine as well as gastric mucosa. Hypotrophy was observed on day 8 as most of the morphological parameters reached the maximal decrease. By contrast, hypoplasy occurred on day 15, when the labeling index (LI) decreased significantly. In the intestine, however, a decrease of the LI was observed only for the upper proliferative cells of the crypts. In the gastric mucosa, the LI was reduced only in the proliferative zone containing progenitor cells (isthmic region). Consequently, the cell differentiation is not similarly affected on all levels of the digestive tract. 相似文献
