Unlike in the adult brain, the newborn brain specifically takes up serum albumin during the postnatal period, coinciding with the stage of maximal brain development. Here we report that albumin stimulates oleic acid synthesis by astrocytes from the main metabolic substrates available during brain development. Oleic acid released by astrocytes is used by neurons for the synthesis of phospholipids and is specifically incorporated into growth cones. Oleic acid promotes axonal growth, neuronal clustering, and expression of the axonal growth-associated protein-43, GAP-43; all these observations indicating neuronal differentiation. The effect of oleic acid on GAP-43 synthesis is brought about by the activation of protein kinase C, since it was prevented by inhibitors of this kinase, such as H-7, polymyxin or sphingosine. The expression of GAP-43 was significantly increased in neurons co-cultured with astrocytes by the presence of albumin indicating that neuronal differentiation takes place in the presence of oleic acid synthesized and released by astrocytes in situ. In conclusion, during brain development the presence of albumin could play an important role by triggering the synthesis and release of oleic acid by astrocytes, which induces neuronal differentiation. 相似文献

Enzyme-linked immunosorbent assays (ELISA) proved to be a fast and simple method for the detection of mycotoxins and other undesired contaminants in food and feed. The present study is focused on the optimisation and exploitation of the egg yolk antibody technology in order to develop competitive ELISAs for the detection of mycotoxins in cereals. Due to its importance as one of the most relevant Fusarium mycotoxins, the trichothecene deoxynivalenol (DON) was selected as representative. Chickens were immunised with different protein conjugates performing varying booster intervals. The antibodies were isolated by the poly(ethylene glycol) precipitation method according to Polson. By use of these antibodies an indirect competitive ELISA was developed for the detection of DON. First investigations of naturally contaminated wheat samples showed a good correspondence with results obtained by GC-ECD when calibration in blank wheat extracts was performed. 相似文献

Among the cytokines involved in rheumatoid arthritis (RA), IL-17 is an important inflammatory mediator. This research aims to assess the effect of the application of biosolids compost and phytoremediation on the mobility of total and biodisponibles (DTPA) fractions of cadmium, copper, lead, and zinc from different horizons of a superficially contaminated soil. Leaching experiment in soil columns was proposed. Treatments contemplated application of compost biosolid and phytoremediation. Two destructive samplings were performed. Total and DTPA trace metals were identified in each horizon. The overall performance of the various elements in its total and DTPA forms show greater concentration in horizon A and fewer gradients between horizons Bt and BC, thus assuming that the high content of clay in horizon Bt (62.9%) limits its movement through the horizons. In the mobile nutrients, a greater mobility was evidenced in DTPA fractions if compared to Total fractions. In the horizon A, the more mobile metals, such as Zn and Cd, evidenced a greater percentage of DTPA/Total fractions in all treatments. The application of compost with or without plant diminished the mobilization of Zn, Cu, and Cd Total, thus limiting a potential leaching to inferior horizons. Comparison analysis of the sequences of the mouse and human genomes has proven a powerful approach in identifying functional regulatory elements within the non‐coding regions that are conserved through evolution between homologous mammalian loci. Here, we applied computational analysis to identify regions of homology in the 5′ upstream sequences of the human tyrosinase gene, similar to the locus control region (LCR) of the mouse tyrosinase gene, located at ?15 kb. We detected several stretches of homology within the first 30 kb 5′ tyrosinase gene upstream sequences of both species that include the proximal promoter sequences, the genomic region surrounding the mouse LCR, and further upstream segments. We cloned and sequenced a 5′ upstream regulatory sequence found between ?8 and ?10 kb of the human tyrosinase locus (termed h5′URS) homologous to the mouse LCR sequences, and confirmed the presence of putative binding sites at ?9 kb, homologous to those described in the mouse tyrosinase LCR core. Finally, we functionally validated the presence of a tissue‐specific enhancer in the h5′URS by transient transfection analysis in human and mouse cells, as compared with homologous DNA sequences from the mouse tyrosinase locus. Future experiments in cells and transgenic animals will help us to understand the in vivo relevance of this newly described h5′URS sequence as a potentially important regulatory element for the correct expression of the human tyrosinase gene. 相似文献

Rapeseed (Brassica napus) is a crop relatively tolerant to salt and sodium. Our objective was to study the interactions between Na, K and Ca and their relationship with its yield under the isolated effects of soil salinity or sodicity.Two experiments were carried out using pots filled with the Ah horizon of a Typic Natraquoll. There were three salinity levels (2.3 dS m-1; 6.0 dS m-1 and 10.0 dS m-1) and three sodicity levels, expressed as sodium adsorption ratios (SAR: 12; 27 and 44). The soil was kept near field capacity.As soil salinity increased, the K/Na and Ca/Na ratios in the tissues decreased markedly but yields and aerial biomass production were not affected. As soil SAR value increased, the K/Na and Ca/Na ratios in plants and K-Na and Ca-Na selectivities decreased. Plants could not maintain their Ca concentration in soil with a high SAR. The grain yield and biomass production diminished significantly in the highest SAR treatment. Our results are consistent with those showing detrimental osmotic effects of salts in Brassica napus. Conversely, under sodicity, the K/Na and Ca/Na ratios in plant tissues decreased considerably, in accordance with grain and biomass production. Recently an association between a genetic variation in TRAF1/C5 and mortality from sepsis or cancer was found in rheumatoid arthritis (RA). The most prevalent cause of death, cardiovascular disease, may have been missed in that study, since patients were enrolled at an advanced disease stage. Therefore, we used an inception cohort of RA patients to investigate the association between TRAF1/C5 and cardiovascular mortality, and replicate the findings on all-cause mortality. Nitrogen (N) losses from agriculture are negatively impacting groundwater, air, and surface water quality. New tools are needed to quickly assess these losses and provide nutrient managers and conservationists with effective tools to assess the effects of current and alternative management practices on N loss pathways. A new N-Index tool was developed in spreadsheet format, allowing prompt assessments of management practices on agricultural N losses. The N-Index tool was compared with experimental field data and shown to estimate the effects of management practices on N loss pathways (probability, P < 0.001). The N-Index correctly assessed the nitrate nitrogen (NO3-N) leaching losses when tested against measured NO3-N leaching data and atmospheric N losses collected over multiple years (annual basis) and locations. Biallelic mutations in the gene encoding centrosomal CDK5RAP2 lead to autosomal recessive primary microcephaly (MCPH), a disorder characterized by pronounced reduction in volume of otherwise architectonical normal brains and intellectual deficit. The current model for the microcephaly phenotype in MCPH invokes a premature shift from symmetric to asymmetric neural progenitor-cell divisions with a subsequent depletion of the progenitor pool. The isolated neural phenotype, despite the ubiquitous expression of CDK5RAP2, and reports of progressive microcephaly in individual MCPH cases prompted us to investigate neural and non-neural differentiation of Cdk5rap2-depleted and control murine embryonic stem cells (mESC). We demonstrate an accumulating proliferation defect of neurally differentiating Cdk5rap2-depleted mESC and cell death of proliferative and early postmitotic cells. A similar effect does not occur in non-neural differentiation into beating cardiomyocytes, which is in line with the lack of non-central nervous system features in MCPH patients. Our data suggest that MCPH is not only caused by premature differentiation of progenitors, but also by reduced propagation and survival of neural progenitors. 相似文献
