Several species of cyanobacteria biomineralizing intracellular amorphous calcium carbonates (ACC) were recently discovered. However, the mechanisms involved in this biomineralization process and the determinants discriminating species forming intracellular ACC from those not forming intracellular ACC remain unknown. Recently, it was hypothesized that the intensity of Ca uptake (i.e., how much Ca was scavenged from the extracellular solution) might be a major parameter controlling the capability of a cyanobacterium to form intracellular ACC. Here, we tested this hypothesis by systematically measuring the Ca uptake by a set of 52 cyanobacterial strains cultured in the same growth medium. The results evidenced a dichotomy among cyanobacteria regarding Ca sequestration capabilities, with all strains forming intracellular ACC incorporating significantly more calcium than strains not forming ACC. Moreover, Ca provided at a concentration of 50 μM in BG‐11 was shown to be limiting for the growth of some of the strains forming intracellular ACC, suggesting an overlooked quantitative role of Ca for these strains. All cyanobacteria forming intracellular ACC contained at least one gene coding for a mechanosensitive channel, which might be involved in Ca influx, as well as at least one gene coding for a Ca2+/H+ exchanger and membrane proteins of the UPF0016 family, which might be involved in active Ca transport either from the cytosol to the extracellular solution or the cytosol toward an intracellular compartment. Overall, massive Ca sequestration may have an indirect role by allowing the formation of intracellular ACC. The latter may be beneficial to the growth of the cells as a storage of inorganic C and/or a buffer of intracellular pH. Moreover, high Ca scavenging by cyanobacteria biomineralizing intracellular ACC, a trait shared with endolithic cyanobacteria, suggests that these cyanobacteria should be considered as potentially significant geochemical reservoirs of Ca. 相似文献

Land use can cause significant impacts on ecosystems and natural resources. To assess these impacts using life cycle assessment (LCA) and ensure adequate decision-making, comprehensive national inventories of land occupation and transformation flows are required. Here, we aim at developing globally differentiated inventories of land use flows that can be used for primary use in life cycle impact assessment or national land planning.
Methods
Using publicly available data and inventory techniques, national inventories for several land use classes were developed. All land use classes were covered with the highest retrievable level of disaggregation within urban, forestry, agriculture and other land use classes, thus differentiating 21 land use classes. For illustrating the application of this newly developed inventory, two different application settings relevant to life cycle impact assessment were considered: the calculation of global normalisation references for 11 land use impact indicators related to soil quality assessment (adopting the methods recommended by the EU Commission) and the determination of generic globally applicable characterisation factors (CFs) resulting from aggregation of country-level CFs for situations for use when land use location is unknown.
Results and discussion
We built national inventories of 21 land occupation and 17 land transformation flows for 225 countries in the world for the reference year 2010. Cross-comparisons with existing inventories of narrower scopes attested its consistency. Detailed analyses of the calculated global normalisation references for the 11 land use impact categories showed different patterns across the land use impact indicators for each country, thus raising attention on key land use impacts specific to each country. Furthermore, the upscaling of country-level CFs to global generic CFs using the land use inventory revealed discrepancies with other alternative approaches using land use data at different resolutions.
Conclusions
In this study, we made a first attempt at developing national inventories of land use flows with sufficient disaggregation level to enable the calculation of normalisation references and differentiated impacts. However, the findings also demonstrated the need to refine the consistency of the inventory, particularly in the combination of land cover and land use data, which should be harmonised in future studies, and to expand it with differentiated coverage of more land use flows relevant to impact assessment.
Stationary batteries are projected to play a role in the electricity system of Switzerland after 2030. By enabling the integration of surplus production from intermittent renewables, energy storage units displace electricity production from different sources and potentially create environmental benefits. Nevertheless, batteries can also cause substantial environmental impacts during their manufacturing process and through the extraction of raw materials. A prospective consequential life cycle assessment (LCA) of lithium metal polymer and lithium‐ion stationary batteries is undertaken to quantify potential environmental benefits and drawbacks. Projections are integrated into the LCA model: Energy scenarios are used to obtain marginal electricity supply mixes, and projections about the battery performances and the recycling process are sourced from the literature. The roles of key parameters and methodological choices in the results are systematically investigated. The results demonstrate that the displacement of marginal electricity sources determines the environmental implications of using batteries. In the reference scenario representing current policy, the displaced electricity mix is dominated by natural gas combined cycle units. In this scenario, the use of batteries generates environmental benefits in 12 of the 16 impact categories assessed. Nevertheless, there is a significant reduction in achievable environmental benefits when batteries are integrated into the power supply system in a low‐carbon scenario because the marginal electricity production, displaced using batteries, already has a reduced environmental impact. The direct impacts of batteries mainly originate from upstream manufacturing processes, which consume electricity and mining activities related to the extraction of materials such as copper and bauxite. 相似文献
Material and energy flows among ecosystems can directly and indirectly drive ecosystem functions. Yet, how populations of consumers respond to allochthonous inputs at a macroecological scale is still unclear. Using a meta‐analysis spanning several biomes, we show that the abundance of recipient populations is 36–57% larger with increased allochthonous inputs. The strength of direct effects on the recipients of these inputs as well as the indirect effects on the consumers of these recipients (i.e. ascending indirect effects) are constant across a latitudinal gradient spanning subtropical, arid, temperate, boreal and arctic ecosystems. However, indirect effect on the in situ resources of the input recipient (i.e. descending indirect effects) decreases with latitude. Our results suggest that the influence of allochthonous inputs can vary across large‐scale gradients of ecosystem productivity and may be driven by the types of trophic interactions within recipient food webs. 相似文献
Coccidia are obligate intracellular protozoan parasites responsible for human and veterinary diseases. Eimeria tenella, the aetiologic agent of caecal coccidiosis, is a major pathogen of chickens. In Toxoplasma gondii, some kinases from the rhoptry compartment (ROP) are key virulence factors. ROP kinases hijack and modulate many cellular functions and pathways, allowing T. gondii survival and development. E. tenella's kinome comprises 28 putative members of the ROP kinase family; most of them are predicted, as pseudokinases and their functions have never been characterised. One of the predicted kinase, EtROP1, was identified in the rhoptry proteome of E. tenella sporozoites. Here, we demonstrated that EtROP1 is active, and the N‐terminal extension is necessary for its catalytic kinase activity. Ectopic expression of EtROP1 followed by co‐immunoprecipitation identified cellular p53 as EtROP1 partner. Further characterisation confirmed the interaction and the phosphorylation of p53 by EtROP1. E. tenella infection or overexpression of EtROP1 resulted both in inhibition of host cell apoptosis and G0/G1 cell cycle arrest. This work functionally described the first ROP kinase from E. tenella and its noncanonical structure. Our study provides the first mechanistic insight into host cell apoptosis inhibition by E. tenella. EtROP1 appears as a new candidate for coccidiosis control. 相似文献
Advances in microscopy with new visualization possibilities often bring dramatic progress to our understanding of the intriguing cellular machinery. Picosecond optoacoustic micro‐spectroscopy is an optical technique based on ultrafast pump‐probe generation and detection of hypersound on time durations of picoseconds and length scales of nanometers. It is experiencing a renaissance as a versatile imaging tool for cell biology research after a plethora of applications in solid‐state physics. In this emerging context, this work reports on a dual‐probe architecture to carry out real‐time parallel detection of the hypersound propagation inside a cell that is cultured on a metallic substrate, and of the hypersound reflection at the metal/cell adhesion interface. Using this optoacoustic modality, several biophysical properties of the cell can be measured in a noncontact and label‐free manner. Its abilities are demonstrated with the multiple imaging of a mitotic macrophage‐like cell in a single run experiment. 相似文献
