Geoactive fungi play a significant role in bioweathering of rock and mineral substrates. Monazite is a phosphate mineral containing the rare earth elements (REE) cerium, lanthanum and neodymium. Little is known about geomicrobial transformations of REE-bearing minerals which are also relevant to REE biorecovery from terrestrial and extra-terrestrial reserves. The geoactive soil fungus Aspergillus niger colonized monazite in solid and liquid growth media without any apparent growth inhibition. In a glucose-minerals salts medium, monazite enhanced growth and mycelium extensively covered rock particle surfaces, probably due to the provision of phosphate and essential trace metals. Teeth-like and pagoda-like etching patterns indicated monazite dissolution, with extensive precipitation of secondary oxalate minerals. Biomechanical forces ensued causing aggressive bioweathering effects by tunnelling, penetration and splitting of the ore particles. High amounts of oxalic acid (~46 mM) and moderate amounts of citric acid (~5 mM) were produced in liquid media containing 2% (wt./vol.) monazite, and REE and phosphate were released. Correlation analysis suggested that citric acid was more effective than oxalic acid in REE mobilization, although the higher concentration of oxalic acid also implied complexant activity, as well as the prime role in REE-oxalate precipitation.  相似文献

There are a need for novel, economical and efficient metal processing technologies to improve critical metal sustainability, particularly for cobalt and nickel which have extensive applications in low-carbon energy technologies. Fungal metal biorecovery processes show potential in this regard and the products of recovery are also industrially significant. Here we present a basis for selective biorecovery of Co and Ni oxalates and phosphates using reactive spent Aspergillus niger culture filtrate containing mycogenic oxalate and phosphate solubilized from struvite. Selective precipitation of oxalates was achieved by adjusting phosphate-laden filtrates to pH 2.5 prior to precipitation. Co recovery at pH 2.5 was high with a maximum of ~96% achieved, while ~60% Ni recovery was achieved, yielding microscale polyhedral biominerals. Co and Ni phosphates were precipitated at pH 7.5, following prior oxalate removal, resulting in near-total Co recovery (>99%), while Ni phosphate yields were also high with a recovery maximum of 83.0%.  相似文献

Intracellular pH is a potent modulator of neuronal functions. By catalyzing (de)hydration of CO2, intracellular carbonic anhydrase (CAi) isoforms CA2 and CA7 contribute to neuronal pH buffering and dynamics. The presence of two highly active isoforms in neurons suggests that they may serve isozyme‐specific functions unrelated to CO2‐(de)hydration. Here, we show that CA7, unlike CA2, binds to filamentous actin, and its overexpression induces formation of thick actin bundles and membrane protrusions in fibroblasts. In CA7‐overexpressing neurons, CA7 is enriched in dendritic spines, which leads to aberrant spine morphology. We identified amino acids unique to CA7 that are required for direct actin interactions, promoting actin filament bundling and spine targeting. Disruption of CA7 expression in neocortical neurons leads to higher spine density due to increased proportion of small spines. Thus, our work demonstrates highly distinct subcellular expression patterns of CA7 and CA2, and a novel, structural role of CA7.  相似文献

We have previously reported that EBNA-5, one of the Epstein–Barr virus-encoded proteins, accumulates in the nuclear bodies containing PML, the promyelocytic leukemia associated protein. In this study, we examine the intranuclear distribution of SV40 large T-antigen (SV40T), the p53 tumor suppressor protein (p53), and PML in a conditionally immortalized cell line, IDH4. In IDH4 cells, the expression of SV40T is regulated by a dexamethasone (Dex)-driven promoter. Withdrawal of Dex results in down-regulation of SV40T and growth arrest, whereas addition of Dex to the growth-arrested cells results in up-regulation of SV40T and proliferation. In proliferating IDH4 cells, SV40T is concentrated in nuclear dots that are also positive for p53. Many of these dots are juxtaposed to PML positive structures but do not colocalize with them. After removal of Dex, SV40T–p53 dots gradually disappear, while the PML structures remain. Induction of SV40T in nonproliferating IDH4 cells causes a coordinated redistribution of SV40T and p53. The immunostaining for SV40T and p53 is first weak, then strong with a homogeneous distribution, and 3–4 days later becomes dot-like again. This reappearance of SV40T–p53 dots coincides with the recovery of proliferation in restimulated IDH4 cells. Also, the p53 pattern correlates with the SV40T pattern with regard to both morphology and intensity during both suppression and induction of SV40T. Taken together, our data suggest that (i) the level of p53 is coregulated with the level of SV40T in a dose-dependent fashion; (ii) the formation of SV40T–p53 nuclear dots correlates with the transformed phenotype; (iii) the SV40T–p53 dots localize preferentially to the neighborhood of PML bodies which are already present in normal cells.  相似文献

Many applications of diversity indices are only valid if they are first transformed into their equivalent number of species. These equivalent numbers of species can be multiplicatively partitioned into independent alpha, beta and gamma components, and can be formed into mathematically consistent similarity measures. The utility of beta diversity and similarity measures that incorporate information about the degree of ecological dissimilarity between species is becoming increasingly recognized. The concept of equivalent number of species is here extended to Rao’s quadratic entropy, opening the way to methods of diversity partitioning that take into account taxonomic or ecological differences between species.  相似文献
