Fatty acid-binding protein (FABP) from bovine heart, a 15 kDa cytoplasmic protein has been investigated by multidimensional homonuclear and heteronuclear NMR-spectroscopy. Perdeuterated palmitic acid has been used as fatty acid ligand. The tertiary structure has been determined from distance geometry calculations with the variable target functions algorithm (DIANA) [1] utilizing 1027 interproton distance constraints, which were obtained from1H-homo-nuclear NOESY spectra. Overlapping NOE crosspeaks were assigned by heteronuclear multidimensional NMR-experiments with a15N-labelled sample. The tertiary structure resembles a -barrel (-clam) consisting of ten anti-parallel -strands and a short helix-turn-helix motif. The -strands are arranged in two nearly orthogonal -sheets composed of 5 strands each. Two forms of E. coli initiation factor IF2, IF2 alpha and IF2 beta, have been known for several years. Both forms are products of the gene infB with translational initiation at codon 1 (AUG) and codon 158 (GUG) in the same reading frame. In this work we demonstrate that IF2 beta exists in two forms, IF2 beta and IF2 beta' with initiation codons 158 (GUG) and 165 (AUG) and molecular masses of 79.7 kDa and 78.8 kDa respectively. We have recently described a fast purification method for IF2 alpha, using an FPLC procedure consisting of ion-exchange liquid chromatography on Q Sepharose HP, Mono Q and Mono S. After the Mono Q step, an apparently homogeneous IF2 beta was observed when analyzed by SDS-PAGE. However the chromatography on Mono S results in the elution of two peaks containing IF2 beta. The N-terminal amino acid sequence of the two proteins identified the first peak to be IF2 beta and the second as a protein which we term IF2 beta' starting seven residues downstream at the AUG codon 165. The activity in vitro of the two purified forms of IF2 beta was tested by measuring the stimulation of binding of the initiator fMet-tRNA(fMet) to 70S ribosomes in the presence of GTP and poly(A,U,G) as messenger-RNA. In this assay no difference in activity is detected.  相似文献

Abstract The manufacturing of a new spherical fibre-optic microsensor is described. The microsensor measures scalar irradiance, i.e. the spherically integrated light at a point in space. The light collector of the probe was a 70-μm diffusing sphere cast on the tip of a 125-μm wide optical fibre tapered down to 15–20 μm diametre. The microsensor had an isotropic (±10%) response from −160° to +160° over the whole spectral range from 400–900 nm in air as well as in water. The microsensor was coupled to a sensitive spectroradiometre and the spectral distribution of scalar irradiance in sediments was measured at 100 μm spatial resolution. Light was available for photosynthesis near the sediment surface at a higher intensity and a different spectral composition than could be expected from the illumination. By the combination of oxygen microelectrodes and the present fibre-optic microsensor it is now possible to study the depth distribution of microbenthic photosynthesis in relation to the available photosynthetically active radiation at ≤ 100 μ m resolution.  相似文献

Aggregation of α‐synuclein is a hallmark of Parkinson's disease and dementia with Lewy bodies. We here investigate the relationship between cytosolic Ca2+ and α‐synuclein aggregation. Analyses of cell lines and primary culture models of α‐synuclein cytopathology reveal an early phase with reduced cytosolic Ca2+ levels followed by a later Ca2+ increase. Aggregated but not monomeric α‐synuclein binds to and activates SERCA in vitro, and proximity ligation assays confirm this interaction in cells. The SERCA inhibitor cyclopiazonic acid (CPA) normalises both the initial reduction and the later increase in cytosolic Ca2+. CPA protects the cells against α‐synuclein‐aggregate stress and improves viability in cell models and in Caenorhabditis elegans in vivo. Proximity ligation assays also reveal an increased interaction between α‐synuclein aggregates and SERCA in human brains affected by dementia with Lewy bodies. We conclude that α‐synuclein aggregates bind SERCA and stimulate its activity. Reducing SERCA activity is neuroprotective, indicating that SERCA and down‐stream processes may be therapeutic targets for treating α‐synucleinopathies.  相似文献

Habitat loss and anthropogenic climate change are important threats to biodiversity conservation. Owing to the concomitantly deteriorating habitat quality, individuals are often forced to disperse to new habitats, rendering dispersal an ecologically important process. However, dispersal ability may differ within and among populations, and is further dependent on environmental conditions. We therefore studied sexual differences in and environmental effects on movement patterns in the sooty copper butterfly Lycaena tityrus. As predicted, males were more active and covered longer distances than females, presumably owing to mate location and territorial disputes. Males alighted more often on flowers than females, probably to fuel their high flight activity, while females showed a high affinity to host-plants for egg-laying. Our findings provide a striking example of sex-related differences in animal behavior, as revealed by the use of customary smartphones, which apparently can comprise suitable means to reveal biologically significant behavioural patterns. More problematic than the technical device used seems to be the challenge of following individual butterflies for long enough in the field, such that any extrapolations to dispersal seem difficult.  相似文献

The density of a defined volume of the human lung can be measured in vivo by a new noninvasive technique. A beam of gamma-rays is directed at the lung and, by measuring the scattered gamma-rays, lung density is calculated. The density in the lower lobe of the right lung in normal man during quiet breathing in the sitting position ranged from 0.25 to 0.37 g.cm-3. Subnormal values were found in patients with emphsema. In patients with pulmonary congestion and edema, lung density values ranged from 0.33 to 0.93 g.cm-3. The lung density measurement correlated well with the findings in chest radiographs but the lung density values were more sensitive indices. This was particularly evident in serial observations of individual patients.  相似文献

Fluxes of D-xylose-1-C14 (xylose) across the wall of the isolated intestine of the bullfrog were studied. When sodium was the principal cation in the mucosal bathing fluid, the transport rate of xylose from the mucosa to the serosa was about 5 times greater than the transport rate from the serosa to the mucosa, indicating an active intestinal transport for this sugar. With potassium as the principal cation on the mucosal side, the transport rate of xylose from the mucosal to the serosal compartment is reduced about 5 to 6 times without appreciable change in the serosal to mucosal transport. The asymmetry was also considerably reduced when ouabain was added to the mucosal and serosal compartments. The data confirm the in vitro and in vivo observations indicating active transport of xylose and are also in accord with the earlier findings that active transport of sugars in the intestine is dependent upon the presence of sodium ions in the mucosal compartment and is inhibited by cardioactive steroids. Since the chemical constitution of xylose does not meet the requirements which were hitherto considered necessary for active transport of sugars in the intestine, this structural requirement has to be revised.  相似文献

The cryopreservation protocols are species‐specific owing to variable sperm sensitivity towards temperature reduction and contact with cryoprotectant solutions. However, little is known about spermatic pathologies, especially after the cryopreservation process. Thus, the objective was to evaluate the effect of cryopreservation on morphological changes in semen of jundiá (Rhamdia quelen). Sperm pool of five males, with >80% motility, as collected, diluted in a cryoprotectant solution and frozen in liquid nitrogen (?196°C). There was a reduction in the percentage of normal cells and sperm motility, accompanied by an increase in the percentage of sperm abnormalities after cryopreservation of R. quelen spermatozoa, indicating a substantial fragility of the spermatozoa towards the cryopreservation process. The most frequent types of morphological changes in the cryopreserved semen were macrocephaly, folded tail, strongly curled tail and distally curled tail. This is the first study to evaluate the spermatic morphology of R. quelen before and after cryopreservation, paving way for further investigations on morphological alterations and for a new classification of these changes in fish semen due to cryopreservation.  相似文献

Background

MUC7 12-mer (RKSYKCLHKRCR), a cationic antimicrobial peptide derived from the human low-molecular-weight salivary mucin MUC7, possesses potent antimicrobial activity in vitro. In order to evaluate the potential therapeutic application of the MUC7 12-mer, we examined the effects of mono- and divalent cations, EDTA, pH, and temperature on its antimicrobial activity.

Methods

Minimal Inhibitory Concentrations (MICs) were determined using a liquid growth inhibition assay in 96-well microtiter plates. MUC7 12-mer was added at concentrations of 1.56–50 μM. MICs were determined at three endpoints: MIC-0, MIC-1, and MIC-2 (the lowest drug concentration showing 10%, 25% and 50% of growth, respectively). To examine the effect of salts or EDTA, a checkerboard microdilution technique was used. Fractional inhibitory concentration index (FICi) was calculated on the basis of MIC-0. The viability of microbial cells treated with MUC7 12-mer in the presence of sodium or potassium was also determined by killing assay or flow cytometry.

Results

The MICs of MUC7 12-mer against organisms tested ranged from 6.25–50 μM. For C. albicans, antagonism (FICi 4.5) was observed for the combination of MUC7 12-mer and calcium; however, there was synergism (FICi 0.22) between MUC7 12-mer and EDTA, and the synergism was retained in the presence of calcium at its physiological concentration (1–2 mM). No antagonism but additivity or indifference (FICi 0.55–2.5) was observed for the combination of MUC7 12-mer and each K+, Na+, Mg2+, or Zn2+. MUC7 12-mer peptide (at 25 μM) also exerted killing activity in the presence of NaCl, (up to 25 mM for C. albicans and up to 150 mM for E. coli, a physiological concentration of sodium in the oral cavity and serum, respectively) and retained candidacidal activity in the presence of KCl (up to 40 mM). The peptide exhibited higher inhibitory activity against C. albicans at pH 7, 8, and 9 than at pH 5 and 6, and temperature up to 60°C did not affect the activity.

Conclusion

MUC7 12-mer peptide is effective anticandidal agent at physiological concentrations of variety of ions in the oral cavity. These results suggest that, especially in combination with EDTA, it could potentially be applied as an alternative therapeutic agent for the treatment of human oral candidiasis.  相似文献
