In a clear and a turbid freshwater lake the biomasses of phytoplankton, periphytic algae and periphytonassociated macrograzers were followed in enclosures with and without fish (Rutilus rutilus) and four light levels (100%, 55%, 7% and < 1% of incoming light), respectively. Fish and light affected the biomass of primary producers and the benthic grazers in both lakes. The biomass of primary producers was generally higher in the turbid than the clear lake, and in both lakes fish positively affected the biomass, while shading reduced it. Total biomass of benthic grazing invertebrates was higher in the clear than in the turbid lake and the lakes were dominated by snails and chironomids + ostracods, respectively. While light had no effect on the biomass of grazers in the clear lake, snail breeding was delayed in the most shaded enclosures and presence of fish reduced the number of snails and the total biomass of grazers. In the turbid lake ostracod abundance was not influenced by light, but was higher in fish-free enclosures. Density of chironomids correlated positively with periphyton biomass in summer, while fish had no effect. Generally, light-mediated regulation of primary producers was stronger in the turbid than in the clear lake, but the regulation did not nambiguously influence the primary consumers. However, regulation by fish of the benthic grazer community was stronger in the clear than in the turbid lake, and in both lakes strong top-down effects on periphyton were seen. The results indicate that if present-day climate in Denmark in the future is found in coastal areas at higher latitudes, the effect of lower light during winter in such areas will be highest in clear lakes, with typically lower fish biomass and higher invertebrate grazer density.  相似文献

The kinetics of cellulose hydrolysis have long been described by an initial fast hydrolysis rate, tapering rapidly off, leading to a process that takes days rather than hours to complete. This behavior has been mainly attributed to the action of cellobiohydrolases and often linked to the processive mechanism of this exo-acting group of enzymes. The initial kinetics of endo-glucanases (EGs) is far less investigated, partly due to a limited availability of quantitative assay technologies. We have used isothermal calorimetry to monitor the early time course of the hydrolysis of insoluble cellulose by the three main EGs from Trichoderma reesei (Tr): TrCel7B (formerly EG I), TrCel5A (EG II), and TrCel12A (EG III). These endo-glucanases show a distinctive initial burst with a maximal rate that is about 5-fold higher than the rate after 5 min of hydrolysis. The burst is particularly conspicuous for TrCel7B, which reaches a maximal turnover of about 20 s(-1) at 30 °C and conducts about 1200 catalytic cycles per enzyme molecule in the initial fast phase. For TrCel5A and TrCel12A the extent of the burst is 2-300 cycles per enzyme molecule. The availability of continuous data on EG activity allows an analysis of the mechanisms underlying the initial kinetics, and it is suggested that the slowdown is linked to transient inactivation of enzyme on the cellulose surface. We propose, therefore, that the frequency of structures on the substrate surface that cause transient inactivation determine the extent of the burst phase.  相似文献

To prevent and treat immune-mediated platelet disorders (e.g. neonatal allo-immune thrombocytopenia and platelet transfusion refractoriness) the causative idiotypic platelet-reactive antibodies have to be detected with high sensitivity and specificity. The "Monoclonal Antibody Immobilization Platelet Assay" (MAIPA) is the diagnostic gold standard for immunotyping sera with respect to alloantibodies against human platelet antigens (HPA). However, it is labor-intensive and time-consuming. In this work, an automated protein chip assay (enzyme-linked sandwich immunoassay) based on interdigitated gold microelectrodes in combination with an electrical read-out system was developed and optimized. For this purpose, specific capture antibodies were immobilized on the gold electrodes. The binding of the target is detected via an enzyme-labeled detection antibody by a redox-recycling process that corresponds to the amount of bound target molecule. With this electrical chip assay it is possible to detect antibodies against HPA-1a, HPA-5b and HLA with high sensitivity and specificity in less than half the duration of the MAIPA protocol with similar intra- and interassay variance.  相似文献

Summary Giving the foundation of his doctrine of the Umwelten v.Uexküll considersKants idealism as the best starting point. The present essay, to the contrary, tries to demonstrate, that the peculiarity of his problems rather corresponds to the logical and metaphysical position ofLeibniz's monadology and so shares its philosophical profundity as well as its ontological difficulties. Cardinal points of this correspondence are the following: 1) There is a plurality of subjective worlds=Umwelten=monads. 2) They are completely isolated one from another. 3) The subject bilds up its reality quite autonomically according to an ideal and specific law=plan=conception. 4) The individual vital laws harmonize according to an optimal general plan. 5) The objective space is denied and regarded as the formalized system of living subjective centres=points of view. 6) Therefrom results the difficulty of explaining a causality of inconscious nature, which is independent from the subject. 7) From this follows the necessity of speculatively amplifying the conception of subject or monad as well as finally displacing the problem of reality into the metaphysical-religious sphere of a supreme (divine) monad or subject.
Résumé En donnant les raisonnements de sa doctrine des Umwelten, M. v.Uexküll considère l'idéalisme deKant comme le meilleur point de départ. Par contre le présent essai cherche à démontrer que la disposition du problème effectivement en question le met plutÔt en parallèle, du point de vue du criticisme et de la métaphysique, avec la monadologie deLeibniz, à l'ampleur philosophique de laquelle elle participe aussi bien qu'à ses difficultés ontologiques. Les points cardinaux de cette concordance sont les suivants: 1) la pluralité des mondes subjectifs=Umwelten =monades; 2) leur intégral isolement réciproque; 3) l'autonomie du sujet quant à la construction de sa réalité, suivant une loi (=plan=conception) idéelle et spécifique; 4) l'harmonie de toutes les lois vitales et individuelles suivant un plan d'ensemble tendant à un optimum; s) le reniement de la réalité absolue de l'espace objectif et son interprétation comme système formalisé de centres subjectifs vivants (= points de vues); 6) la difficulté qui en résulte pour l'admissibilité d'une causalité de la nature inconsciente, indépendante du sujet; 7) la nécessité, en découlant, d'une amplification spéculative de l'idée du sujet ou de la monade, ainsi que, finalement, le renvoi du problème de la réalité dans la sphère métaphysico-religieuse d'une monade suprÊme (divine) ou d'un suprÊme sujet.

Zusammenfassung Im Gegensatz zur Volkeltschen, auf situationsbedingte Komplexqualitäten zurückgreifenden Deutung des Verhaltens der Spinne gegenüber der Beute innerhalb und außerhalb des Netzes wird gezeigt, daß die Spinne in weitestem Maße durch den isolierten Vibrationsreiz zur Fanghandlung bestimmt wird.Analog wird ein ebenso isolierbarer Einzelreiz aufgewiesen, der das Feindverhalten bewirkt, nämlich der durch die Luft übertragene Schwingungsreiz, der biologisch sinnvoll nur als Warnungssignal vor Wespen gedeutet werden kann.Ein direktes, nicht erst durch Netzschwingungen vermitteltes Wahrnehmen der Luftschwingungen wird trotz bisher nicht mit Sicherheit festgestellter Schallrezeptoren wahrscheinlich gemacht.Während in den Funktionskreisen des Feindes und der Beute dem situativen Moment gegenüber den scharfumrissenen Einzelreizen somit eine sehr geringe Bedeutung zukommt, gestaltet sich die Lage für die Volketsche Auffassung günstiger im Funktionsbereich des Netzbaues. Die Bedeutung bestimmter Merkmale hängt hier wesentlich ab von der Situation im Sinne der jeweils erreichten Phase der Bauhandlung.Allerdings genügt es nicht, unter der Situation nur die Gesamtheit der äußeren Gegebenheiten zu verstehen. Wesentlich ist auch die innere Situation, in der das Tier sich befindet und auf Grund deren objektiv gleiche Bedingungen eine Umstimmung erfahren. Dies wird durch zwei Experimente gezeigt, bei denen sowohl in der Phase des Radienspinnens als auch in der Phase des Fangfädenspinnens alle gesponnenen Fäden sofort wieder mit Hilfe glühender Nadeln zerstört werden. Trotz des hierdurch hervorgerufenen Fehlens der normalen äußeren Abschlußbedingungen für die einzelnen Phasen nimmt die Bauhandlung, wenngleich in verzögerter Weise, schließlich doch ihren normalen Verlauf.  相似文献

We formalise and present a new generic multifaceted complex system approach for modelling complex business enterprises. Our method has a strong focus on integrating the various data types available in an enterprise which represent the diverse perspectives of various stakeholders. We explain the challenges faced and define a novel approach to converting diverse data types into usable Bayesian probability forms. The data types that can be integrated include historic data, survey data, and management planning data, expert knowledge and incomplete data. The structural complexities of the complex system modelling process, based on various decision contexts, are also explained along with a solution. This new application of complex system models as a management tool for decision making is demonstrated using a railway transport case study. The case study demonstrates how the new approach can be utilised to develop a customised decision support model for a specific enterprise. Various decision scenarios are also provided to illustrate the versatility of the decision model at different phases of enterprise operations such as planning and control.  相似文献

BackgroundConflicting data exist for anti-cancer effects of anti-placental growth factor (anti-PlGF) in combination with anti-VEGF. Still, this treatment combination has not been evaluated in intracranial glioblastoma (GBM) xenografts. In clinical studies, position emission tomography (PET) using the radiolabeled amino acid O-(2-18F-fluoroethyl)-L-tyrosine (18F-FET) and magnetic resonance imaging (MRI) add complementary but distinct information about glioma growth; however, the value of 18F-FET MicroPET combined with MicroMRI has not been investigated preclinically. Here we examined the use of 18F-FET MicroPET and MicroMRI for evaluation of anti-VEGF and anti-PlGF treatment response in GBM xenografts.MethodsMice with intracranial GBM were treated with anti-VEGF, anti-PlGF + anti-VEGF or saline. Bioluminescence imaging (BLI), 18F-FET MicroPET and T2-weighted (T2w)-MRI were used to follow tumour development. Primary end-point was survival, and tumours were subsequently analysed for Ki67 proliferation index and micro-vessel density (MVD). Further, PlGF and VEGFR-1 expression were examined in a subset of the xenograft tumours and in 13 GBM patient tumours.ResultsAnti-VEGF monotherapy increased survival and decreased 18F-FET uptake, BLI and MVD, while no additive effect of anti-PlGF was observed. 18F-FET SUVmax tumour-to-brain (T/B) ratio was significantly lower after one week (114±6%, n = 11 vs. 143±8%, n = 13; p = 0.02) and two weeks of treatment (116±12%, n = 8 vs. 190±24%, n = 5; p = 0.02) in the anti-VEGF group as compared with the control group. In contrast, T2w-MRI volume was unaffected by anti-VEGF. Gene expression of PlGF and VEGFR-1 in xenografts was significantly lower than in patient tumours.Conclusion18F-FET PET was feasible for anti-angiogenic response evaluation and superior to T2w-MRI; however, no additive anti-cancer effect of anti-PlGF and anti-VEGF was observed. Thus, this study supports use of 18F-FET PET for response evaluation in future studies.  相似文献
