In order to evaluate the intracellular function of glia maturation factor (GMF), we overexpressed GMF in C6 rat glioma cells using two methods: stable transfection using the pcDNA3 plasmid, and transient transfection using replication-defective human adenovirus. With both methods, C6 cells transfected with GMF and overexpressing the protein exhibit a lower saturation density in culture compared to non-transfected or vector alone controls. Transfected cells also exhibit morphological differentiation as shown by the outgrowth of cell processes. When inoculated into nude mice, transfected cells are less tumorigenic than controls, and express the mature astrocytic marker glial fibrillary acidic protein. In tissue culture, transfected cells show a 3.5-fold increase in CuZn-dependent superoxide dismutase (CuZnSOD) activity. Western blot analysis reveals a 3.5-fold increase in CuZnSOD protein, suggesting an induction of the enzyme. In view of recent findings that reactive oxygen species (ROS) and the antioxidant enzymes are intricately involved in key physiologic processes such as proliferation, differentiation and apoptosis, the study raises the possibility that CuZnSOD may be a mediator of GMF function.  相似文献

Duringdirect passive heating in young men, a dramatic increase in skin bloodflow is achieved by a rise in cardiac output (c) andredistribution of flow from the splanchnic and renal vascular beds. Toexamine the effect of age on these responses, seven young (Y; 23 ± 1 yr) and seven older (O; 70 ± 3 yr) men were passively heated withwater-perfused suits to their individual limit of thermal tolerance.Measurements included heart rate (HR), c (byacetylene rebreathing), central venous pressure (via peripherally inserted central catheter), blood pressures (by brachial auscultation), skin blood flow (from increases in forearm blood flow by venous occlusion plethysmography), splanchnic blood flow (by indocyanine green clearance), renal blood flow (byp-aminohippurateclearance), and esophageal and mean skin temperatures.c wassignificantly lower in the older than in the young men (11.1 ± 0.7 and 7.4 ± 0.2 l/min in Y and O, respectively, at the limit ofthermal tolerance; P < 0.05),despite similar increases in esophageal and mean skin temperatures andtime to reach the limit of thermal tolerance. A lower stroke volume (99 ± 7 and 68 ± 4 ml/beat in Y and O, respectively, P < 0.05), most likely due to anattenuated increase in inotropic function during heating, was theprimary factor for the lower c observed inthe older men. Increases in HR were similar in the young and older men;however, when expressed as a percentage of maximal HR, the older menrelied on a greater proportion of their chronotropic reserve to obtainthe same HR response (62 ± 3 and 75 ± 4% maximal HR in Y andO, respectively, P < 0.05). Furthermore, the older men redistributed less blood flow from thecombined splanchnic and renal circulations at the limit of thermaltolerance (960 ± 80 and 720 ± 100 ml/min in Y and O,respectively, P < 0.05). As a resultof these combined attenuated responses, the older men had asignificantly lower increase in total blood flow directed to the skin.

We cultured and sequenced newly collected material of Percursaria dawsonii Hollenberg et I. A. Abbott, a poorly known epibiont of limpets found along the west coast of North America. Zoospores, parthenogametes, and zygotes exhibited empty‐spore germination, which produced a prostrate system that expanded by stolon‐like development. A dense carpet of initially uniseriate filaments arose from the prostrate system. These upright filaments soon became biseriate proximally and pluriseriate distally, where they were flat, twisted and distromatic for approximately four‐fifths of their length and rarely became hollow in larger individuals. These fronds became fertile distally and released quadriflagellate zoospores and/or biflagellate isogametes. Individual gametophytic fronds were dioecious, and alternated with isomorphic sporophytic fronds. The gametes moved rapidly and often exhibited an unusual side‐to‐side vibration that made them appear like winged insects. Comparisons of 18S rRNA gene and ITS sequences indicated that P. dawsonii should be included in the genus Blidingia, family Kornmanniaceae.  相似文献

We have previously shown that treatment of mice bearing a large MOPC-315 plasmacytoma with a low dose of the anticancer drug melphalan (l-phenylalanine mustard;l-PAM) results in the acquisition of a potent CD8+ T-cell-mediated anti-MOPC-315 cytotoxic T lymphocyte (CTL) activity by the hitherto immunosuppressed tumor bearers, and this immunity contributes to complete tumor eradication. In the studies presented here, we sought to determine how the acquisition of this antitumor immunity following low-dose chemotherapy is possible, in light of the report that MOPC-315 tumor cells produce transforming growth factor- (TGF-), an immunosuppressive cytokine that can down-regulate the generation of CTL responses. We found that the acquisition of CTL activity following low-dosel-PAM therapy is not due to a chemotherapy-induced decrease in the sensitivity of MOPC-315 tumor bearer spleen cells to TGF--mediated inhibition of CTL-generation. Moreover, even spleen cells from MOPC-315 tumor-bearing mice, which had receivedl-PAM therapy 7 days earlier and had acquired CTL activity in vivo, were sensitive to the inhibitory activity of TGF- upon culture for as little as 1 day, with or without stimulator tumor cells. However, the production of TGF- by MOPC-315 tumors decreased drastically as a consequence of the low-dose chemotherapy. Summary The relative sampling efficiency of the Samplair (Allergenco Inc.) and Rotorod samplers (Sampling Technologies Inc.) were compared for different conifer pollen sizes and at different wind speeds. The wind tunnel experiments conducted with the two samplers are valid for the range of wind speeds and conifer pollen types which might be expected in seed orchards in Canada. A Quadratic relationship was established between the two samplers which revealed a decrease in relative efficiency on the part of the Samplair as compared to the Rotorod at wind speeds less than 5 m/s. An apparent increase in the efficiency of the samplair at higher wind speeds resulted from a decrease in the sampling ability of the rotorod.  相似文献

Photographs and maps of the floating canopy of the giant kelp, Macrocystis pyrifera, provide an important data source to monitor nearshore water quality in southern California. Declines in water quality related to turbidity from coastal development, ocean discharges, and non-point source runoff have caused reductions in the areal extent of these kelp beds. Historically the kelp beds have been monitored by a variety of methods including small format infrared and color photography. New digital remote sensing instruments combined with geographical information system (GIS) databases offer an efficient method for collecting and analyzing data on changes in kelp bed size and location. SPOT satellite imagery has been found to provide adequate resolution for mapping the larger beds of giant kelp along the California coast. Beds smaller than 10 ha are not resolved well with SPOT imagery and need to be mapped with a resolution greater than the 20 m pixel size provided by the SPOT multispectral imagery. Imagery from a prototype of the Positive Systems ADAR system, an airplane mounted multispectral video sensor, provided a spatial resolution of 2.3 m in 4 spectral bands. ADAR imagery taken on 2 October 1991 of the San Onofre Kelp Bed in northern San Diego County showed 39% more kelp than small format color infrared photography made during the same time period.  相似文献
