Recent research demonstrated the utility of fecal progestagens (P4) for detecting pregnancy in elk (Cervus elaphus) during mid- to late gestation. Several factors, however, may influence fecal P4 excretion and limit its use in free-ranging animals. We investigated the effects of nutrition and body condition (percent ingesta-free body fat) on fecal P4 concentrations and incidence of abortion. During mid-gestation (late December 1997 through early March 1998), 40 gravid cow elk varying in body condition were placed on three diets (high, medium, and low) in which the amount of food offered varied. Feces were collected periodically and analyzed for P4 via radioimmunoassay. We found no significant effect of dietary treatment on P4 concentrations, but as body condition declined, P4 concentrations declined significantly. This decline did not impede the ability to detect pregnancy based on previously reported criteria, even for elk in such poor condition that they aborted. However, fecal P4 concentrations in 10% (4/39) of samples collected from 13 non-pregnant animals maintained on a high plane of nutrition were false-positive for pregnancy. We suggest alternate criteria for determining pregnancy in elk using fecal P4 values: > 1.25 micrograms/g feces as pregnant, < 1.0 microgram/g feces as non-pregnant, and 1.0-1.25 micrograms/g feces as inconclusive. Finally, two cows that aborted did not abort until weeks after being classified as emaciated and near death, suggesting that nutrition-associated abortion in elk may not occur during mid-gestation except under extremely harsh conditions. 相似文献

When (22S, 23S)-homobrassinolide (SSHB) was added at 1 g l–1 to hairy root cultures of Artemisia annua, the production of artemisinin reached to 14 mg l–1, an increment of 57% over the control. SSHB treatment led concomitantly to an increased biomass production of 15 g l–1. Chemical signals originating from the ovaries of gravid females of Musca domestica (Diptera: Cyclorrhapha: Muscidae) attract ovipositing females to common egg-laying sites. Behavioral experiments indicated that females preferred to oviposit in fermented wheat bran containing ovaries from reproductively mature houseflies. Females preferred to oviposit in fermented wheat bran than wet wheat bran. This effect was additive with the attraction to housefly ovaries. Solvent extracts from housefly ovaries were attractive to gravid females. Extracts obtained with hexane were most attractive to gravid females for egg laying, and extracts obtained with ethyl acetate attracted more egg laying than extracts obtained by dichloromethane. Gas chromatography (GC) and gas chromatography coupled with mass spectrometry (GC-MS) analysis showed that tricosane and (Z)-9-tricosene were the main components of the hexane extracts. Both tricosane and (Z)-9-tricosene were shown to elicit dose-dependent aggregation of gravid females in oviposition bioassays, but high doses of either chemical were not attractive. 相似文献

The integrative nuclear FGFR1 signaling (INFS) pathway functions in association with cellular growth, differentiation, and regulation of gene expression, and is activated by diverse extracellular signals. Here we show that stimulation of angiotensin II (AII) receptors, depolarization, or activation protein kinase C (PKC) or adenylate cyclase all lead to nuclear accumulation of fibroblast growth factor 2 (FGF-2) and FGFR1, association of FGFR1 with splicing factor-rich domains, and activation of the tyrosine hydroxylase (TH) gene promoter in bovine adrenal medullary cells (BAMC). The up-regulation of endogenous TH protein or a transfected TH promoter-luciferase construct by AII, veratridine, or PMA (but not by forskolin) is abolished by transfection with a dominant negative FGFR1TK-mutant which localizes to the nucleus and plasma membrane, but not by extracellularly acting FGFR1 antagonists suramin and inositolhexakisphosphate (IP6). Mechanism of TH gene activation by FGF-2 and FGFR1 was further investigated in BAMC and human TE671 cultures. TH promoter was activated by co-transfected HMW FGF-2 (which is exclusively nuclear) but not by cytoplasmic FGF-1 or extracellular FGFs. Promoter transactivation by HMWFGF-2 was accompanied by an up-regulation of FGFR1 specifically in the cell nucleus and was prevented FGFR1(TK-) but not by IP6 or suramin. The TH promoter was also transactivated by co-transfected wild-type FGFR1, which localizes to both to the nucleus and the plasma membrane, and by an exclusively nuclear, soluble FGFR1(SP-/NLS) mutant with an inserted nuclear localization signal. Activation of the TH promoter by nuclear FGFR1 and FGF-2 was mediated through the cAMP-responsive element (CRE) and was associated with induction of CREB- and CBP/P-300-containing CRE complexes. We propose a new model for gene regulation in which nuclear FGFR1 acts as a mediator of CRE transactivation by AII, cell depolarization, and PKC. 相似文献

Two major types of arbucular mycorrhizal associations, the Arum-type and the Paris-type, have been identified based on morphological features. Although the Paris-type is the most common, it is the Arum-type that has been most intensively studied in terms of structure/function because of its prevalence in agronomically important plant species. In this study, the interface between the host cell cytoplasm and intracellular hyphae (extensive hyphal coils and arbusculate coils), which typify the Paris-type mycorrhiza, was studied. Using immunofluorescence techniques combined with laser scanning confocal microscopy, dramatic changes in the cytoskeleton in colonized cells were observed. Changes in the positioning of both host cell microtubules and actin filaments occurred in colonized plant cells. Both microtubules and actin filaments were associated with the hyphal coils and the arbusculate coils. An interfacial matrix, of host origin, was demonstrated between hyphal coils and arbusculate coils using various affinity techniques. It formed an apoplastic compartment consisting of cellulose and pectins between the fungus and host cell cytoplasm. There was less labelling adjacent to the fine branches of arbusculate coils compared to the hyphal coils. These observations show some similarities to those seen with Arum-type mycorrhizas. 相似文献

In most eukaryotes telomeres are extended by telomerase. Drosophila melanogaster, however, lacks telomerase, and telomere-specific non-LTR retrotransposons, HeT-A and TART, transpose specifically to chromosome ends. A Drosophila strain, Gaiano, that has long telomeres has been identified. We extracted the major Gaiano chromosomes into an Oregon-R genetic background and examined the resulting stocks after 60 generations. In situ hybridization using HeT-A and TART sequences showed that, in stocks carrying either the X or the second chromosome from Gaiano, only the Gaiano-derived chromosomes display long telomeres. However, in stocks carrying the Gaiano third chromosome, all telomeres are substantially elongated, indicating that the Gaiano chromosome 3 carries a factor that increases HeT-A and TART addition to the telomeres. We show that this factor, termed Telomere elongation (Tel), is dominant and localizes as a single unit to 69 on the genetic map. The long telomeres tend to associate with each other in both polytene and mitotic cells. These associations depend on telomere length rather than the presence of Tel. Associations between metaphase chromosomes are resolved during anaphase, suggesting that they are mediated by either proteinaceous links or DNA hydrogen bonding, rather than covalent DNA-DNA bonds. 相似文献

We present the elements of a mathematical computational model that reflects the experimental finding that the time-scale of a neuron is not fixed; but rather varies with the history of its stimulus. Unlike most physiological models, there are no pre-determined rates associated with transitions between states of the system nor are there pre-determined constants associated with adaptation rates; instead, the model is a kind of "modulating automata" where the rates emerge from the history of the system itself. We focus in this paper on the temporal dynamics of a neuron and show how a simple internal structure will give rise to complex temporal behavior. The internal structure modeled here is an abstraction of a reasonably well-understood physiological structure. We also suggest that this behavior can be used to transform a "rate" code into a "temporal one". 相似文献
