Summary Microinjections of dopamine (DA) were made into specific forebrain loci in goldfish (Carassius auratus: 40–85 g) to study the involvement of DA in behavioral thermoregulation. Injections of 25, 50, 100 and 250 ng DA into the anterior aspect of the nucleus preopticus periventricularis (NPP) led to consistent, dose-dependent decreases in selected temperature was observed following injections of 5 or 10 ng DA. Injections of the control solution were without effect.Injections of DA into other forebrain loci, including the posterior half of the NPP, either had no thermoregulatory effect or had minor thermoregulatory effects which, in comparison to injections into the most effective sites, were inconsistent and required larger doses to obtain. The decrease in selected temperature following injections of 100 ng DA into the anterior NPP was blocked by haloperidol, a dopaminergic antagonist, but not by phentolamine, a noradrenergic antagonist. Injections of haloperidol alone resulted in a minor, but statistically significant, increase in selected temperature.The most sensitive DA sites lie caudal to the sites most sensitive to norepinephrine within the anterior NPP. DA acts on the dopaminergic receptors of central thermoregulatory neurons in the anterior NPP of goldfish. Modified procedures for separating and determining adenine nucleotides are described. Separation was achieved by one-dimensional thin-layer chromatography on polyethyleneimine-cellulose plates in 1.4 LiCl after prewashing the spotted plate in deionized water. The fluorometric procedure for determining adenine and adenine derivatives using glyoxal dihydrate trimer was modified to reduce the heat step from 3 hr to 25 min without loss of sensitivity or stability. These modified procedures were used to determine the adenine nucleotide levels in rat liver.  相似文献

Human genomic DNAs for the eosinophil granule proteins, eosinophil-derived neurotoxin (EDN) and eosinophil cationic protein (ECP), were isolated from genomic libraries. Alignment of EDN (RNS2) and ECP (RNS3) gene sequences demonstrated remarkable nucleotide similarities in noncoding sequences, introns, and flanking regions, as well as in the previously known coding regions. Detailed examination of the 5'-noncoding regions yielded putative TATA and CAAT boxes, as well as similarities to promoter motifs from unrelated genes. A single intron of 230 bases was found in the 5' untranslated region and we suggest that a single intron in this region and an intronless coding region are features common to many members of the RNase gene superfamily. The RNS2 and RNS3 genes were localized to the q24-q31 region of human chromosome 14. It is likely that these two genes arose as a consequence of a gene duplication event that took place approximately 25-40 million years ago and that a subset of anthropoid primates possess both of these genes or closely related genes.  相似文献

Specialization in narrow ecological niches may not only help species to survive in competitive or unique environments but also contribute to their extermination over evolutionary time. Although the “evolutionary dead end” hypothesis has long been debated, empirical evidence from species with detailed information on niche specialization and evolutionary history remains rare. Here we use a group of four closely related Cnemaspis gecko species that depend highly on granite boulder caves in the Mekong Delta to investigate the potential impact of ecological specialization on their evolution and population dynamics. Isolated by unsuitable floodplain habitats, these boulder‐dwelling geckos are among the most narrowly distributed Squamata in the world. We applied several coalescence‐based approaches combined with the RAD‐seq technique to estimate their divergence times, gene flow and demographic fluctuations during the speciation and population differentiation processes. Our results reveal long‐term population shrinkage in the four geckos and limited gene flow during their divergence. The results suggest that the erosion and fragmentation of the granite boulder hills have greatly impacted population divergence and declines. The habitat specialization of these geckos has led to fine‐scaled speciation in these granite rocky hills; in contrast, specialization might also have pushed these species toward the edge of extinction. Our study also emphasizes the conservation urgency of these vulnerable, cave‐dependent geckos.  相似文献

Stressors are commonly encountered by organisms and often prove to be energetically costly. Certain stressors can simultaneously affect multiple components of an animal's energy budget and can either exacerbate energetic costs to the individual or offset one another. Here we used a commonly encountered stressor, the pesticide carbaryl, to examine the complex effects that acute environmental disturbances can have on energy expenditure, allocation, and acquisition, important processes that influence growth and reproduction. After exposing lizards (Sceloporus occidentalis) to carbaryl, we measured their metabolism over a 48 h period and assessed their food consumption over 96 h. We found no difference in total energy expenditure among treatment groups, but lizards exposed to the highest dose of carbaryl allocated energy differently than other groups. Compared to controls, these lizards exhibited a 16-30% increase in standard metabolic rate (SMR), which was offset by a 45-58% decrease in additional energy expenditures. Lizards in the highest dose group also exhibited a 30-34% decrease in energy acquisition compared to controls. The net result was a 1.83 kJ decrease in energy assimilation, equivalent to 5 times their daily SMR requirements. Our results indicate that energetic consequences of stressors may result in complex energetic trade-offs, and emphasize the need to simultaneously examine the effect of stressors on multiple portions of an animal's energy budget.  相似文献
