Chemical and surface analyses are carried out using Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM–EDS), atomic force microscopy (AFM), confocal laser scanning microscopy (CLSM), glow discharge spectroscopy (GDS) and extracellular surface protein quantification to thoroughly investigate the effect of supplementary As(V) during biooxidation of arsenopyrite by Acidithiobacillus thiooxidans. It is revealed that arsenic can enhance bacterial reactions during bioleaching, which can strongly influence its mobility. Biofilms occur as compact-flattened microcolonies, being progressively covered by a significant amount of secondary compounds (S n 2- , S0, pyrite-like). Biooxidation mechanism is modified in the presence of supplementary As(V), as indicated by spectroscopic and microscopic studies. GDS confirms significant variations between abiotic control and biooxidized arsenopyrite in terms of surface reactivity and amount of secondary compounds with and without As(V) (i.e. 6 μm depth). CLSM and protein analyses indicate a rapid modification in biofilm from hydrophilic to hydrophobic character (i.e. 1–12 h), in spite of the decrease in extracellular surface proteins in the presence of supplementary As(V) (i.e. stressed biofilms).  相似文献

In vivo phosphorylation of PEPC in Egeria densa was studied using plants at high temperature and in light, and plants kept at low temperature and in light. The isoform induced by high temperature and light was more phosphorylated in the light. Insect disease vectors show diminished fecundity when infected with Plasmodium. This phenomenon has already been demonstrated in laboratory models such as Aedes aegypti, Anopheles gambiae and Anopheles stephensi. This study demonstrates several changes in physiological processes of A. aegypti occurring upon infection with Plasmodium gallinaceum, such as reduced ecdysteroid levels in hemolymph as well as altered expression patterns for genes involved in vitellogenesis, lipid transport and immune response. Furthermore, we could show that P. gallinaceum infected A. aegypti presented a reduction in reproductive fitness, accompanied by an activated innate immune response and increase in lipophorin expression, with the latter possibly representing a nutritional resource for Plasmodium sporozoites.  相似文献

We studied the mechanical properties of living starfish oocytes belonging to two species, Astropecten Auranciacus and Asterina pectinifera, over a wide range of timescales. We monitored the Brownian motion of microspheres injected in the cytoplasm using laser particle‐tracking (LPT) and video multiple‐particle‐tracking (MPT) techniques, to explore high‐ and low‐frequency response ranges, respectively. The analysis of the mean‐square‐displacements (MSD) allowed us to characterize the samples on different timescales. The MSD behavior is explained by three power‐law exponents: for short times (τ < 1 ms) it reflects the semiflexible behavior of the actin network; for intermediate timescales (1 ms < τ < 1 s) it is similar to that of a soft‐glass material; finally for long times (τ > 1 s) it behaves mainly like a viscous medium. We computed and compared the viscoelastic moduli using a recently proposed model describing the frequency response of the cell material. The variety AF-28 of Sorghum bicolor Pers and the hybrid R-1090 were compared in relation to the sorghum midge, Contarinia sorghicola (Coquillet), (Diptera: Cecidomyidae) in two counties of the State of São Paulo. They were compared in seven successive monthly trials beginning in September 1972. Averages of 252 and 137 adult flies emerged per head of the hybrid R-1090 in Campinas and Jaboticabal respectively, while 2.9 and 0.8 emerged from AF-28. The damage was graded with a visual scale from 0 to 4. Average damage grades of 3.5 and 3.3 were attributed to the hybrid R-1090 in Campinas and Jaboticabal respectively, while the variety AF-28 had averages of 0.5 and 0.3, respectively.
Résumé La variété AF-28 et l'hybride R-1090 de Sorghum bicolor Pers. ont été comparés à différentes époques de l'année et en deux localités différentes de l'Etat de Sâo Paulo, Brésil.On n'a pas constaté d'interaction variété x époque ou variété x lieu.AF-28 a prouvé sa résistance à toutes les époques, aussi bien dans un lieu que dans l'autre; l'hybride par contre s'est montré grandement susceptible.Des moyennes de 252,2 et 136,7 mouches par panicule ont été observées pour l'hybride R-1090 à Campinas et à Jaboticabal contre 2,9 et 0,8 seulement par panicule de AF-28. Le préjudice a été estimé visuellement selon une échelle de 0 à 4. Des préjudices d'une valeur moyenne de 3,5 et 3,3 ont été attribués à l'hybride R-1090 à Campinas et à Jaboticabal respectivement, alors que simultanément la variété AF-28 ne subissait que des préjudices évalués à 0,5 et 0,3 respectivement.Quatre espèces de microhyménoptères parasites de Contarinia sorghicola ont été récoltées au cours de ce travail: Eupelmus popa et trois espèces différentes du genre Tetrastichus.

Clinical characteristics associated with isolation of Borrelia burgdorferi sensu lato from skin have not been fully evaluated. To gain insight into predictors for a positive EM skin culture, we compared basic demographic, epidemiologic, and clinical data in 608 culture-proven and 501 culture-negative adult patients with solitary EM. A positive Borrelia spp. skin culture was associated with older age, a time interval of >2 days between tick bite and onset of the skin lesion, EM ≥5 cm in diameter, and location of the lesion on the extremities, whereas several other characteristics used as clinical case definition criteria for the diagnosis of EM (such as tick bite at the site of later EM, information on expansion of the skin lesion, central clearing) were not. A patient with a 15-cm EM lesion had almost 3-fold greater odds for a positive skin culture than patients with a 5-cm lesion. Patients with a free time interval between the tick bite and onset of EM had the same probability of a positive skin culture as those who did not recall a tick bite (OR=1.02); however, the two groups had >3-fold greater odds for EM positivity than patients who reported a tick bite with no interval between the bite and onset of the lesion. In conclusion, several yet not all clinical characteristics used in EM case definitions were associated with positive Borrelia spp. skin culture. The findings are limited to European patients with solitary EM caused predominantly by B. afzelii but may not be valid for other situations.  相似文献

Knowledge on the distribution and abundance of species is plagued by significant taxonomic and geographic biases that influence the analyses on biodiversity patterns. Due to this, standard, easy-to-use methods are needed to design efficient field campaigns that minimize data deficiencies. We evaluate the applicability, usefulness and effectiveness of a survey design protocol based on the Environmental Diversity (ED) criterion under different scenarios, with examples of varying extent of environmental niche, range of spatial distribution and level of previous knowledge. We planned surveys for epiphytic bryophytes growing in three types of forests at NW Iberian Peninsula (dominated by Quercus ilex, Q. faginea and Q. pyrenaica). Knowledge on the distribution and abundance of epiphytic bryophytes in this region presents large gaps and strong geographic biases. Besides, the three forest types differ in their environmental requirements, spatial distribution and level of previous knowledge, providing three working scenarios to test the response of the protocol under different situations. The protocol was implemented as a set of sequential selection rules, starting by an ED-based criterion aiming at maximizing the coverage of climatic and geographic variability; further criteria include an iterative set of qualitative properties: maximizing forest area, conservation status and accessibility. The protocol performed efficiently at different ranges of spatial distribution levels of environmental variability, and degree of previous knowledge and generated an even distribution of sampling points that efficiently covered the diversity of epiphytic bryophytes. The results show that ED protocols are a proficient and time-saving approach to select sampling sites by objective criteria also for groups with high dispersal ability and fragmented landscapes.  相似文献
