In the range of concentrations 12.5–100 mM glucose stimulated Cu-mediated free radical peroxidation of low density lipoproteins (LDL) from human blood plasma. Based on analysis of kinetic parameters of the LDL peroxidation it was found that intensification of this process is caused by formation of free radical intermediates of glucose autooxidation during generation of reactive oxygen species in the presence of transition metal ions. Normalization of blood glucose in patients with type 2 diabetes during therapy was accompanied by a significant decrease of LDL oxidation. Therapy with the sugar-lowering drug metformin, which utilizes methylglyoxal, caused much higher inhibition of the in vivo LDL peroxidation in blood of patients with diabetes mellitus probably due to the decrease of methylglyoxal-dependent generation of superoxide anion radicals shown by us earlier [Biochemistry (Moscow) 2009, vol. 74, pp. 568–574].  相似文献

Low-molecular-weight dicarbonyls formed during free radical peroxidation of polyene lipids (malondialdehyde) and autooxidation (glyoxal) or other oxidative transformations of glucose (methylglyoxal) are able to modify the structure of lipid-protein supramolecular complexes of cells. We investigated changes in the erythrocyte membrane structure after an 18-h exposure of human red blood cells in the presence of various natural dicarbonyls. The changes in the mechanical properties of the membrane after membrane modification by carbonyls were evaluated by the susceptibility of erythrocytes to hypoosmotic hemolysis. It has been shown that treatment of red blood cells with malondialdehyde increases the resistance of these cells to hypoosmotic hemolysis, whereas the malondialdehyde isomer, methylglyoxal, in contrast, makes red blood cells more sensitive to the action of hypoosmotic solutions. Paradoxically, a homologue of malondialdehyde, glyoxal, has no effect on hemolysis of red blood cells in hypoosmotic solutions. The findings point to the possibility of the multidirectional effect of low-molecular-weight dicarbonyls with similar structures on the structure and function of biological membranes.  相似文献

The maximal radioactivity of mouse liver cytosol was observed 18 hours after intraperitoneal injection of 5-CH3-[3H] alpha-tocopherol emulsion with Tween 80. Intraperitoneal injections of Tween 80 (500 mg/kg) without alpha-tocopherol significantly decreased the glutathione lipoperoxidase and superoxide dismutase activities of liver cytosol. Vitamin E effect on the glutathione lipoperoxidase activity was weakly pronounced, while the superoxide dismutase activities of cytosol and liver mitochondria were markedly increased thereby. It was assumed that natural free radical scavengers can induce enzymatic utilization of superoxide radical anions, while synthetic free radical scavengers can trigger on enzymatic systems of lipoperoxide utilization.  相似文献

Lipoproteins (very low, low and high density) purified from canine fresh plasma produced relaxation of isolated dog coronary artery and rabbit aortic ring segments precontracted with potassium, 30 mM or phenylephrine, 10(-6) M, respectively. No apparent species-specific effects of lipoproteins were revealed and they produced relaxation of other species vessels as well, which differed quantitatively. No apparent endothelium-dependent relaxation produced by animals' lipoproteins was evident. The interference of lipoproteins with some steps in electromechanical coupling is suggested which seemingly result to decrease in excitability of voltage and receptor operated calcium channels of smooth muscle cell membrane. The role of lipoproteins seems to be in maintenance of low level of smooth muscle cell membrane excitability.  相似文献

After 2 month of feeding vitamin E-supplemented diet (100.6 and 0 mg/kg; group I-control, II and III, respectively) the concentration of lipid peroxidation products (diene conjugates, malondialdehyde, Schiff's bases) and activity of antioxidant enzymes (superoxide dismutase, glutathione peroxidase) was estimated in rat heart and liver. Although the content of alpha-tocopherol in organs of group II was significantly decreased, the concentration of peroxidation products and enzyme activities was unchanged. Moreover, these parameters were constant in rat liver of group III. The heart was more sensitive because in group III to vitamin E deficiency (the alpha-tocopherol level was dropped fourfold) the concentration of diene conjugates and malondialdehyde was increased and superoxide dismutase activity was decreased. Thus insufficiency of vitamin E may result in selective alterations of myocardial functions. In addition, vitamin E may be useful instrument for correction of free radical oxidation and antioxidant system activity in the heart.  相似文献

The relationship between behavioral polymorphism and the variation for the function of the hypothalamic-pituitary-adrenocortical axis (HPAA) was studied in the meat-and-wool (n = 447) and Altaian fine-fleece (n = 45) sheep breeds under production farm conditions. The behavior studied included negatively associated reactions: feeding reactions and defensive reactions in relation to human. In sheep, the intrapopulation variation for the stress level of corticosteroids depended on the effect of behavior, the type of stimulus, and the interaction between these two factors and correlated with the behavior phenotype (r varied from -0.551 to -0.226). In the stress responses to either 24-h starvation or lamb weaning, hormonal reactions were similar in wild-type sheep that have clearly marked defensive reactions and in the sheep of the domestic phenotype that was contrasted to the wild type. In the stress responses to isolation from the herd, transportation, and competition for food, hormonal reactions were stronger in the wild type than in the domestic phenotype; alternatively, in the stress response to being kept in cages in pairs, hormonal reaction was stronger in the domestic phenotype than in the wild type. In the sheep that were placed in cages, a single playing of tape-recorded anxiety signals caused the motivational or emotional stress that was characterized by an unusually long latent period of the progressive HPAA reaction and its subsequent chronic course (up to 9 days). Animals of contrasted behavioral phenotypes differed in the course of the HPAA reaction. Heritable behavioral polymorphism and interaction between behavior and the type of stimulus are the regulation factors in the expression and variability of stimulus-specific reactions of HPAA. Behavior can serve as the genetic marker for individual features of HPAA functioning in the stress response of sheep.  相似文献
