The acute effects of aqueous solutions of As, Cd, Cu, Pb, F, and Zn ions at concentrations from 0.01 to 100 micrograms per milliliter and solutions adjusted to pH 2 to 6 with nitric or sulfuric acid were studied with respect to acetylene reduction, net photosynthesis, respiration rate, and chlorophyll content in Vernal alfalfa (Medicago sativa L. cv. Vernal). The effects of the various treatments on acetylene reduction varied from no demonstrable effect by any concentration of F and 42% inhibition by 100 micrograms Pb2+ per milliliter, to 100% inhibition by 10 micrograms Cd2+ per milliliter and 100 micrograms per milliliter As, Cu2+, and Zn2+ ions. Zn2+ showed statistically significant inhibition of activity at 0.1 micrograms per milliliter. Acid treatments were not inhibitory above pH 2, at which pH nitric acid inhibited acetylene reduction activity more than did sulfuric acid. The inhibition of acetylene reduction by these ions was Zn2+ > Cd2+ > Cu2+ > AsO3 > Pb2+ > F. The sensitivity of acetylene reduction to the ions was roughly equal to the sensitivity of photosynthesis, respiration, and chlorophyll content when Pb2+ was applied, but was 1,000 times more sensitive to Zn2+. The relationship of the data to field conditions and industrial pollution is discussed.  相似文献

Dipeptidase A (Dip-A), a new peptidase locus in Drosophila melanogaster, is located on the second chromosome at map position 55.2 and in the 41A-B; 42A2-3 interval in the salivary gland chromosomes. Three alleles are reported. In the Carpenter, North Carolina population the allele frequencies are: Dip-A 6 (fastest)=0.064; Dip-A 4 (intermediate)=0.920; and Dip-A 2 (slowest)=0.015.  相似文献

Gene frequency variation at eight polymorphic allozyme loci in Drosophila melanogaster populations in North Carollina and the east coast of the United States were analyzed utilizing the variance component estimation procedures suggested by Cockerham (1969, 1973). These variance components were used to estimate correlations of genes within small geographic regions. The average (over loci) correlation between genes in the same individual within subpopulations was estimated to be 0.033. That between genes in the same subpopulation in different individuals was estimated to be very small, although significantly different from zero. The macrogeographic variation measured by the correlation of genes sampled from the same local region was large for some loci and smaller for others. this variation was also analyzed by correlation with latitude and longitude. Several previously recognized clines were identified as were several new clines.--These results were interpreted as indicating either some degree of nonrandom mating and local breeding unit isolation or a low frequency of null alleles. The geographic and temporal variation has no simple interpretation.  相似文献

Observations of 8 groups each containing three adult spiny mice (Acomys cahirinus) revealed that instances of chasing and physical displacement were quite common, while fighting and biting were rarely observed. The relationships between the most frequent behavioral categories were presented in a correlation matrix. In a second, study, ♀♀ tested in their home cages were dominant over ♂♂ In the ♂♂ home cages, however, no differences in the frequency of ♀ vs. ♂ aggression were observed. A technique is reported for cryostat sectioning of lung tissue in an expanded state for use in viral immunofluorescence studies. A 1: 2 mixture of O.C.T. embedding compound and phosphate-buffered saline is injected intratracheally into fresh lung tissne. The lung tissue is frozen in liquid nitrogen and sectioned with a cryostat. Compared to other published reports of lung sectioning for immunofluorescence miscroscopy, this method has the advantages of bekg easy and quick, maintaining the lung sectiom in an expanded rather than collapsed state and avoiding contact with chemicals potentially capable of altering sensitive viral antigens.  相似文献

The effects of CO2 elevation on the dynamics of fine root (FR) mass and ectomycorrhizal (EM) mass and colonization were studied in situ in a Florida scrub oak system over four years of postfire regeneration. Soil cores were taken at five dates and sorted to assess the standing crop of ectomycorrhizal and fine roots. We used ingrowth bags to estimate the effects of elevated CO2 on production of EM roots and fine roots. Elevated CO2 tended to increase EM colonization frequency but did not affect EM mass nor FR mass in soil cores (standing mass). However, elevated CO2 strongly increased EM mass and FR mass in ingrowth bags (production), but it did not affect the EM colonization frequency therein. An increase in belowground production with unchanged biomass indicates that elevated CO2 may stimulate root turnover. The CO2-stimulated increase of belowground production was initially larger than that of aboveground production. The oaks may allocate a larger portion of resources to root/mycorrhizal production in this system in elevated rather than ambient CO2.  相似文献

Phosphatidylinositol 4-kinase has been purified 10,148-fold to a specific activity of 2.7 mumol/mg/min from bovine uteri. This purification was accomplished by detergent extraction of an acetone powder, ammonium sulfate precipitation, and chromatography on MonoQ, S-Sepharose, MonoP, and hydroxylapatite columns. The purified enzyme has a molecular mass of 55 kDa and appears to be monomeric. Kinetic analyses of the enzymatic activity demonstrated apparent Km values of 18 microM and 22 micrograms/ml (approximately 26 microM) for ATP and phosphatidylinositol, respectively, optimal activity in the pH range of 6.0-7.0, and a sigmoidal dependence of enzymatic activity on [Mg2+]. Ca2+ inhibited the enzyme at nonphysiological concentrations with 50% inhibition observed at a free [Ca2+] of approximately 300 microM. The purified enzyme efficiently utilized both ATP and 2'-deoxy-ATP as phosphoryl donors and specifically phosphorylated phosphatidylinositol on the fourth position. No phosphatidylinositol-4-phosphate 5-kinase activity was observed in the purified enzyme preparations. To our knowledge, this is the first reported purification of a phosphatidylinositol-specific phosphatidylinositol 4-kinase.  相似文献
