Chronic inflammatory demyelinating polyneuropathy (CIDP) is a chronic disorder, manifesting with monophasic or relapsing course. Progressive course is rare in children. The article presents a boy with progressive generalized muscle weakness and areflexia since the age of two, developed after viral infection. Electromyoneurography showed severe neurogenic lesion, with myopathic pattern in proximal muscles. Increased serum ganglioside antibody titers (anti-GM1 and anti-GD1b) were registered. Sural nerve biopsy revealed demyelination and onion bulbs. Inflammatory perivascular CD3 positive infiltrates were present in muscle and nerve biopsies. Brain magnetic resonance imaging showed cortical atrophy, hyperintensities of the white matter and gray matter hypointensities. Improvement occurred on intravenous immune globulins and methylprednisolone treatment. Demyelination might develop in central and peripheral nervous system associated with inflammatory myopathy in patients with progressive course of CIDP.  相似文献

Centromere association of the chromosomal passenger complex (CPC; Borealin-Survivin-INCENP-Aurora B) and Sgo1 is crucial for chromosome biorientation, a process essential for error-free chromosome segregation. Phosphorylated histone H3 Thr3 (H3T3ph; directly recognized by Survivin) and histone H2A Thr120 (H2AT120ph; indirectly recognized via Sgo1), together with CPC’s intrinsic nucleosome-binding ability, facilitate CPC centromere recruitment. However, the molecular basis for CPC–Sgo1 binding and how their physical interaction influences CPC centromere localization are lacking. Here, using an integrative structure-function approach, we show that the “histone H3-like” Sgo1 N-terminal tail-Survivin BIR domain interaction acts as a hotspot essential for CPC–Sgo1 assembly, while downstream Sgo1 residues and Borealin contribute for high-affinity binding. Disrupting Sgo1–Survivin interaction abolished CPC–Sgo1 assembly and perturbed CPC centromere localization and function. Our findings reveal that Sgo1 and H3T3ph use the same surface on Survivin to bind CPC. Hence, it is likely that these interactions take place in a spatiotemporally restricted manner, providing a rationale for the Sgo1-mediated “kinetochore-proximal” CPC centromere pool.  相似文献

Exposure of murine leukemia cells in culture to bis-acetyl-diaminopentane (BADP) caused erythroid maturation as measured by the accumulation of hemoglobin in treated cells. The appearance of differentiated cells in cultures exposed to BADP occurred 18 to 20 hours earlier than in those treated with dimethylsulfoxide (DMSO), a standard inducer of differentiation in this system. Studies with [3H]BADP indicated the occurrence of relatively rapid association of the inducer with cells, and subsequent linear accumulation. Fractionation of cellular components and measurement of radioactivity from BADP therein demonstrated that this agent preferentially associates with a fraction enriched for plasma membrane. In addition, [3H]BADP was capable of binding to the plasma membrane-enriched fraction isolated from murine erythroleukemia cells as measured by gel filtration. These findings support the concept that interaction of inducers of murine erythroleukemia differentiation such as BADP with components of the surface membrane may be important in the cascade of events that lead to the erythroid maturation of these leukemic cells.  相似文献

Bacterial genomes are a huge reservoir of genes encoding J-domain protein co-chaperones that recruit the molecular chaperone DnaK to assist protein substrates involved in survival, adaptation, or fitness. The atc operon of the aquatic mesophilic bacterium Shewanella oneidensis encodes the proteins AtcJ, AtcA, AtcB, and AtcC, and all of them, except AtcA, are required for growth at low temperatures. AtcJ is a short J-domain protein that interacts with DnaK, but also with AtcC through its 21 amino acid C-terminal domain. This interaction network is critical for cold growth. Here, we show that AtcJ represents a subfamily of short J-domain proteins that (i) are found in several environmental, mostly aquatic, β- or ɣ-proteobacteria and (ii) contain a conserved PX7W motif in their C-terminal extension. Using a combination of NMR, biochemical and genetic approaches, we show that the hydrophobic nature of the tryptophan of the S. oneidensis AtcJ PX7W motif determines the strong AtcJ–AtcC interaction essential for cold growth. The AtcJ homologues are encoded by operons containing at least the S. oneidensis atcA, atcB, and atcC homologues. These findings suggest a conserved network of DnaK and Atc proteins necessary for low-temperature growth and, given the variation in the atc operons, possibly for other biological functions.  相似文献

The species composition, relativeabundance, and seasonal dynamics of selectednatural enemies of cereal aphids were monitoredin spring wheat fields in Moscow, Idaho in 1997and 1998. Trials also examined the potentialimpact of resistance to Russian wheat aphid(RWA), Diuraphis noxia (Mordvilko)(Homoptera: Aphididae) in wheat, on aphidbiological control agents. Natural enemypopulations were monitored on two springwheats: D. noxia susceptible variety`Centennial' and resistant genotype `IDO488'. Field plots were artificially infested withadult D. noxia, and sampled for cerealaphids and parasitoids weekly. Coccinellidpredators were monitored once in 1997 and twicein 1998. The coccinellids Hippodamiaconvergens Guerin, Coccinellaseptempunctata L., C. transversoguttataBrown and C. trifasciata Mulsant weredetected. No significant differences in adultor immature coccinellid densities were observedbetween the D. noxia resistant andsusceptible genotypes. During both years, themost abundant primary hymenopteran parasitoidswere Diaeretiella rapae (M'Intosh), Aphidius ervi Haliday, A. avenaphis(Fitch), and Lysiphlebus testaceipes(Cresson), Aphelinus varipes (Foerster),Aphidius colemani Viereck, Aphidiuspicipes (Nees), Aphidius sp., Monoctonus washingtonensis Pike & Stary, Praon gallicum Stary, Praon occidentaleBaker, and Praon sp. were also detected. Numbers of both D. noxia and D.rapae were significantly greater on Centennialthan on IDO488 in both years. When all speciesof cereal aphids and parasitoids areconsidered, the total percentage parasitism wasnot significantly different between thegenotypes. There was no interaction betweenD. noxia resistance and the populationdensity of the predators or parasitoidsmonitored. These results suggest that the D. noxia resistant line had no adverse impacton natural enemies under the conditions ofthese field experiments.  相似文献

ThepK a values of His-38 and His-50 of the heparin-binding protein, bovine platelet factor 4, are 5.6 and 6.5, respectively, as determined by1H NMR spectroscopy. The1H NMR resonance of His-38 of bovine platelet factor 4 which exhibits the lowerpK a value is perturbed upon heparin binding to a greater degree than the resonance of His-50. Human platelet factor 4 contains the homologous residues His-23 and His-35. ThepK a values of the two histidine residues of human platelet factor 4 are 5.3 and 6.4. The1H NMR resonance of the histidine of human platelet factor 4 exhibiting the lowerpK a value also is perturbed upon heparin binding to a greater degree than the histidine resonance exhibiting the higherpK a , thereby suggesting comparable heparin-protein interactions in bovine and human platelet factor 4.  相似文献

A series of experiments examined possible strategies human subjectsmight use in tasks requiring rapid detection or recognitionof a taste quality. In a reaction time (RT) paradigm, subjectswere to decide whether each of a series of stimuli flowed overthe tongue contained a previously designated target taste. Severaltasks of varying difficulty were used. The simplest task requiredsubjects to decide whether the target taste or water was presented.The most difficult task required discrimination between twodifferent target tastes in a series of mixtures formed by orthogonallycombining the target taste with two different irrelevant tastes.The speed at which subjects could detect and/or recognize targettastes was related to the RT for the particular taste. However,it was also clear that other variables, including the specificstimuli in the mixtures and the cognitive demands placed onthe subjects, influenced performance. These results suggestthat differences in taste onset time, as indexed by RT, canserve as a cue which subjects use to aid identification of singletastes in a mixture. It is concluded that the ease with whichsubjects can identify single tastes in a mixture is relatedto, among other variables, the differences in taste onset timebetween the tastes.  相似文献

We tested herein the hypothesis that exposure time significantly contributes to the vertical distribution profile of nematodes during a tidal cycle as a function of distinct feeding and locomotion behaviors, conditioned by body morphology. We showed that the vertical distribution profile of the slender with filiform tail, numerically dominant Terschellingia longicaudata is in fact significantly correlated with sediment changes induced by tidal variation. Conversely, none of the other nematode species showed unequivocal evidence of vertical migration. Horizontal spatial heterogeneity also influenced the vertical distribution of nematode associations, probably as a response to varying temperature and desiccation levels at the sediment surface. The resulting vertical profiles for individual or species groups are a trade-off among locomotory and feeding strategies and concordant morphological adaptations.  相似文献
