This study investigated neurotoxicity of chronic fluorosis in the rat hippocampus. Newly weaning, male, Sprague-Dawley (SD) rats were administered 15, 30, and 60 mg/L sodium fluoride (NaF) solution (fluorine ion concentration 8.25, 16.50, and 33.00 mg/L, respectively), and tap water, for 18 months. The neurotoxicological mechanism was examined with a focus on intracellular calcium overload. Results showed that as the fluoride concentration increased, calcium ion concentration [Ca2+], the expression of calcium/calmodulin-dependent protein kinase II α (CaMKIIα), and the expression of catus proto-oncogene protein c-fos (c-fos) all tend to increase. Compared to the control group, Ca2+, CaMKIIα, and c-fos significantly increased (P < 0.05) in the moderate-fluoride and the high-fluoride groups. These results indicate that Ca2+/CaMKIIα/c-fos channel signal may be the molecular mechanism of central nervous system damage caused by chronic fluoride intoxication. Moreover, elevated Ca2+ concentration in the hippocampus may be the initiating factor of neuronal apoptosis induced by fluoride. 相似文献

Grafts made from human amniotic membrane are used to prevent recurrence of pterygium after excision. The success of the procedure can be affected by the quality of preparation and preservation of the grafts. We prospectively evaluated the safety and efficacy of cryopreserved amniotic membrane prepared at the research tissue bank of the Biotechnology Research Center in Tripoli, Libya, and used as adjunct therapy in primary pterygium excision. Twenty-six patients (15 males and 11 females) aged 21–78 years and indicated for primary pterygium excision were transplanted at the Tripoli Eye Hospital with the amniotic membrane grafts. Sixteen patients (62 %) were available for all three follow-up visits scheduled at 1, 3 and 6 months post-surgery. By the third visit, two patients (12.5 %) developed granuloma and three (18.8 %) had pterygium recurrence. The grafts were used after cryopreservation for ≤180 days or >180 days, but statistical analysis showed that the complications were not associated with the length of storage. Moreover, the high rate of complications in this study was not caused by use of cryopreserved AM. In conclusion, locally produced cryopreserved AM is safe as an adjunct therapy for treatment of primary pterygium excision. 相似文献

Impact of exogenous calcium and ethylene glycol tetraacetic acid (EGTA) supplement on chickpea (Cicer arietinum L.) germinating seeds exposed to cadmium stress for 6 days was studied. Ca and EGTA late treatment (3 days) alleviated growth inhibition and decreased Cd accumulation as well as lipid peroxidation and protein carbonylation in both root and shoot cells. Exogenous effector application relieved Cd-induced cell death which was associated with a constant level of ATP, which was considered as an apoptotic-like process. Redox balance was examined through the study of the redox state of pyridine nucleotide couples NAD+/NADH and NADP+/NADPH as well as their related oxidative [NAD(P)H-oxidase] and dehydrogenase (glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and malate dehydrogenase) enzyme activities. The present research illustrated an ameliorative effect of Ca and EGTA on growth of Cd-exposed chickpea seedlings that occurs through the protection of sensitive cell sites from Cd-induced oxidation, namely membrane lipids and proteins, rather than the improvement of recycling capabilities of the cellular reducing power. 相似文献

In Tunisia, Tipuana tipu (Benth .) Kuntze is an exotic tree, which was introduced many years ago and planted as ornamental street, garden, and park tree. The present work reported, for the first time, the chemical composition and evaluates the allelopathic effect of the hydrodistilled essential oils of the different parts of this tree, viz., roots, stems, leaves, flowers, and pods gathered in the area of Sousse, a coastal region, in the East of Tunisia. In total, 86 compounds representing 89.9 – 94.9% of the whole oil composition, were identified in these oils by GC‐FID and GC/MS analyses. The root essential oil was clearly distinguished for its high content in sesquiterpene hydrocarbons (β‐caryophyllene, 1 (44); 24.1% and germacrene D, 2 (53); 20.0%), while those obtained from pods, leaves, stems, and flowers were dominated by non‐terpene hydrocarbons. The most important ones were n‐tetradecane (41, 16.3%, pod oil), 1,7‐dimethylnaphthalene (43, 15.6%, leaf oil), and n‐octadecane (77, 13.1%, stem oil). The leaf oil was rich in the apocarotene (E)‐β‐ionone ( 4 (54); 33.8%), and the oil obtained from flowers was characterized by hexahydrofarnesylacetone ( 5 (81); 19.9%) and methyl hexadecanoate (83, 10.2%). Principal component and hierarchical cluster analyses separated the five essential oils into three groups and two subgroups, each characterized by the major oil constituents. Contact tests showed that the germination of lettuce seeds was totally inhibited by the root essential oil tested at 1 mg/ml. The inhibitory effect on the shoot and root elongation varied from ?1.6% to ?32.4%, and from ?2.5% to ?64.4%, respectively. 相似文献

The decolourisation of fresh and stored olive mill wastewaters (OMW) and the biodegradation of three groups (F1, F2 and F3) of phenolic compounds by Geotrichum candidum were investigated. Separated phenolic compounds derived from natural OMW ultrafiltration using membranes with a cutoff 2and 100 kDa. G. candidum growth on fresh OMW decreased pH and reduced COD and colour of 75% and 65%, respectively. However, on the stored-black OMW a failure of COD and colour removal were observed. G. candidum activity on this later substrate was enhanced by the addition of a carbon source easily metabolised, misleading an improvement of the COD reduction and decolourization that reached 58% and 48%, respectively. Growth of G. candidum in the presence of F2 or F3 polyphenolic fractions induced high decolourisation and depolymerisation of phenolic compounds. Whereas, very week decolourisation and biodegradation were observed with F1 fraction. Moreover, the highest levels of lignin peroxidase (LiP) and manganese peroxidase (MnP) activities were obtained in the presence of F2 fraction. These results showed that increasing of molecular-mass of aromatics led to an increase in levels of depolymerisation, decolourisation and COD removal by G. candidum culture. 相似文献

The effects of light intensity and temperature on Arthrospira platensis growth and production of extracellular polymeric substances (EPS) in batch culture were evaluated using a three-level, full-factorial
design and response surface methodology. Three levels were tested for each parameter (temperature: 30, 35, 40°C; light intensity:
50, 115, 180 μmol photons m−2 s−1). Both growth and EPS production are influenced mainly by the temperature factor but the interaction term temperature*light
intensity also had a significant effect. In addition, conditions optimising EPS production are different from those optimising
growth. The highest growth rate (0.414 ± 0.003 day−1) was found at the lowest temperature (30°C) and highest light intensity (180 μmol photons m−2 s−1) tested, no optima were detectable within the given test range. Obviously, optima for growth must be at a temperature lower
than 30°C and a light intensity higher than 180 μmol photons m−2 s−1. For EPS production, light intensity had a positive linear effect (optimum obviously higher than 180 μmol photons m−2 s−1), but for the temperature parameter a maximum effect was detectable at 35°C. 相似文献

The impact of mapped microsatellites on the study of genetic diversity of Tunisian apricot accessions was assessed. The genetic
variability of 47 traditional apricot cultivars originating from several areas in Tunisia was investigated with 32 polymorphic
microsatellite loci selected for their location throughout the eight linkage groups of Prunus genome. The higher polymorphism and greater transportability of these markers among Prunus species were proved by the expected heterozygosity (He = 0.56) and Shannon’s index of diversity (I = 1.05), indicating that Tunisian apricot germplasm maintained a substantial level of genetic diversity. According to their
geographical origin, the genetic differentiation among groups (north, center, and south; Fst = 0.04) was lower, while the
gene flow among groups was consequent (Nm = 4.79), attesting a narrow genetic background of apricot in the country. Both unweighted
pair-group method with arithmetic mean dendrogram, based on Nei’s genetic distances and factorial correspondence analysis,
separated northern cultivars from central and southern cultivars, revealing the same molecular basis of apricot material in
the Center and the South of Tunisia. These results revealed the efficiency of mapped markers for genetic variability measurements
compared to randomly ones, however, no advantage was observed considering the genetic relationships among studied accessions. 相似文献

To understand the role of TGF-β signaling in cardiovascular development, we generated mice with conditional deletion of the
TGF-β type II receptor (TβRII) gene (Tgfbr2) in cells expressing the smooth muscle cell-specific protein SM22α. The SM22α promoter was active in tissues involved in cardiovascular development: vascular smooth muscle cells (VSMCs), epicardium and
myocardium. All SM22-Cre+/−/Tgfbr2
flox/flox embryos died during the last third of gestation. About half the mutant embryos exhibited heart defects (ventricular myocardium
hypoplasia and septal defects). All mutant embryos displayed profound vascular abnormalities in the descending thoracic aorta
(irregular outline and thickness, occasional aneurysms and elastic fiber disarray). Restriction of these defects to the descending
thoracic aorta occurred despite similar levels of Tgfbr2 invalidation in the other portions of the aorta, the ductus arteriosus and the pulmonary trunk. Immunocytochemistry identified
impairment of VSMC differentiation in the coronary vessels and the descending thoracic aorta as crucial for the defects. Ventricular
myocardial hypoplasia, when present, was associated to impaired α-SMA differentiation of the epicardium-derived coronary VSMCs.
Tgfbr2 deletion in the VSMCs of the descending thoracic aorta diminished the number of α-SMA-positive VSMC progenitors in the media
at E11.5 and drastically decreased tropoelastin (from E11.5) and fibulin-5 (from E.12.5) synthesis and/or deposition. Defective
elastogenesis observed in all mutant embryos and the resulting dilatation and probable rupture of the descending thoracic
aorta might explain the late embryonic lethality. To conclude, during mouse development, TGF-β plays an irreplaceable role
on the differentiation of the VSMCs in the coronary vessels and the descending thoracic aorta. 相似文献

Anguillicola crassus, parasite nematode of the European eel Anguilla anguilla, was recorded for the first time in Tunisia (1999) in the Ichkeul lagoon. Its distribution has since spread toward Bizerte and Ghar El Melh lagoons. The monthly epidemiological survey reveals that A. crassus exists throughout the year in the Ichkeul lagoon. In this lagoon, its prevalence is low in winter (12% in December), with a marked increases in the spring reaching a maximum in March (35%), before it starts to decrease in summer with a minimum in July (4.35%), which in turn is followed by a pronounced new rise in autumn (30% in November). However, mean intensity values do not show such a marked variation. The majority of the values are between 1 and 1.5 parasites per host. In the Bizerte and Ghar El Melh lagoons, the presence of this nematode is limited only to one to three months. Investigations in the Tunis lagoon did not reveal until now the presence of A. crassus. It has been observed that the length of the eel influences the prevalence values: A. crassus becomes less common if the length of the eel increases. Comparatively with the global epidemiological values of A. crassus signalled subsequently (1999) in the Ichkeul lagoon, we note that the present values record a clean increase. 相似文献
