Abstract The vascular patterns of the ventral side of the kidneys in Bufo bufo and Rana ridibunda is similar. Strong venulae renales revehentes dominate. In Bombina variegata and Xenopus laevis, however, also many superficially located glomeruli are found at the ventral side. In Xenopus further branching of the renal arteries into afferent arterioles attracts attention. Bundles of delicate afferent arterioles originate within circumscribed areas of the renal artery. The glomerular layer of the kidney is tightest in Bombina, and has its maximal extension in Rana. It covers up to 2/3 of the thickness of the kidney while in the other species studied the glomeruli are restricted to the ventral third of the kidney. Glomeruli with double afferent or efferent arterioles were rarely found in Xenopus. The vascularization of the dorsal side of the kidneys is characterized by the presence of large (Bufo, Rana, Xenopus) or small (Bombina) venulae renales advehentes.  相似文献

The angioarchitecture of the toad's mesencephalon is studied by scanning electron microscopy of vascular corrosion casts. The arterial supply is performed by the superior mesencephalic artery and by an artery arising from the ramus posterior of the arteria carotis cerebralis just in front of the retroin-fundibular communicating artery. The venous drainage is exerted by the vena diencephalica, the vena lateralis mesencephalis and by anterior and posterior branches of the encephaloposthypophysial portal vein. Characteristics of the mesencephalic angioarchitecture are the centrifugal direction of the blood flow, the intensive capillarization, and the formation of vascular meshworks in the region of the stratum griseum periventriculare tecti and of the stratum griseum superficiale tecti.  相似文献

The ubiquitin ligase Cbl-b is an established regulator of T cell immune response thresholds. We recently showed that adoptive cell transfer (ACT) of cblb(-/-) CD8(+) T cells enhances dendritic cell (DC) immunization-mediated anti-tumor effects in immune-competent recipients. However, translation of cblb targeting to clinically applicable concepts requires that inhibition of cblb activity be transient and reversible. Here we provide experimental evidence that inhibition of cblb using chemically synthesized siRNA has such potential. Silencing cblb expression by ex vivo siRNA transfection of polyclonal CD8(+) T cells prior to ACT increased T cell tumor infiltration, significantly delayed tumor outgrowth, and increased survival rates of tumor-bearing mice. As shown by ex vivo recall assays, cblb silencing resulted in significant augmentation of intratumoral T cell cytokine response. ACT of cblb-silenced polyclonal CD8(+) T cells combined with DC-based tumor vaccines predominantly mediated anti-tumor immune responses, whereas no signs of autoimmunity could be detected. Importantly, CBLB silencing in human CD8(+) T cells mirrored the effects observed for cblb-silenced and cblb-deficient murine T cells. Our data validate the concept of enhanced anti-tumor immunity by repetitive ACT of ex vivo cblb siRNA-silenced hyper-reactive CD8(+) T cells as add-on adjuvant therapy to augment the efficacy of existing cancer immunotherapy regimens in clinical practice.  相似文献

The vascularization of the diencephalon (with choroid plexus of the third ventricle, epithalamus and pineal region, thalamus and hypothalamus) of the toad, Bufo bufo (L.) has been studied by means of scanning electron microscopy of vascular corrosion casts. To localize angioarchitectonic patterns of distinct diencephalic regions the authors refer to critical point dried specimens and to histological sections. In the choroid plexus a supply via one choroid artery, which arises from the posterior telencephalic artery, was found. Its strict dichotomous branching is pointed out. In generally a similar vascular pattern like that in the choroid plexus of the fourth ventricle has to be reported. Furthermore the epithalamic region with the epiphysial area was under investigation. No special angioarchitecture of the epiphysis was found. There are also no prominent vascular connections with thalamic or hypothalamic regions. The thalamic region is supplied by branches of the posterior telencephalic artery as well as by branches of the preoptic artery. Epithalamic and thalamic regions are drained via the posterior diencephalic vein. Special attention was also paid to the preoptic, the chiasmatic and the retrochiasmatic area. CCL18 is a human chemokine secreted by monocytes and dendritic cells. The receptor for CCL18 is not yet known and the functions of this chemokine on immune cells are not fully elucidated. In this study, we describe that CCL18 is present in skin biopsies of atopic dermatitis (AD) patients but not in normal or psoriatic skin. CCL18 was specifically expressed by APCs in the dermis and by Langerhans and inflammatory dendritic epidermal cells in the epidermis. In addition, the serum levels of CCL18 and the percentages of CCL18-producing monocyte/macrophages and dendritic cells were significantly increased in AD patients compared with healthy controls. Furthermore, we demonstrate that CCL18 binds to CLA(+) T cells in peripheral blood of AD patients and healthy individuals and induces migration of AD-derived memory T cells in vitro and in human skin-transplanted SCID mice. These findings highlight a unique role of CCL18 in AD and reveal a novel function of this chemokine mediating skin homing of a subpopulation of human memory T cells.  相似文献

The vascular patterns of the epidermally derived slime glands of the Atlantic hagfish, Myxine glutinosa, and of the Pacific hagfish, Eptatretus stouti, have been studied by scanning electron microscopy of vascular corrosion casts. In Myxine a simple two-dimensional vascular network sheaths the slime glands, while in Eptatretus there is also a great number of capillary loops of different lengths arising from the sheathing network and extending into the interior of the glands. These basic differences in slime glands vascular patterns are thought to reflect substantially different physiological behaviour of the slime glands in Myxine and Eptatretus.  相似文献

Larval to adult microvascular anatomy of the esophagus was studied in the South African Clawed Toad, Xenopus laevis (Daudin) by scanning electron microscopy of vascular corrosion casts and correlative light microscopy of paraplast embedded stained tissue sections. Analyses of vascular corrosion casts of tadpole esophaguses at premetamorphosis revealed a wide‐meshed, but mature looking capillary bed which during following prometamorphosis increased in density and gained the adult‐like pattern during late metamorphic climax by sprouting and nonsprouting angiogenesis. In adult Xenopus, the esophageal mucosa possessed a dense subepithelial capillary bed fed by one or two esophageal arteries that originated from right and/or left thoracic aorta just distal to the origin of the subclavian arteries. In the adult undistended esophagus, esophageal arteries revealed an undulating course, a pattern that guarantees a continuous blood supply when the esophagus is extremely wide expanded as it is the case when adult Xenopus swallows large prey.  相似文献

Systemic knockout of adipose triglyceride lipase (ATGL), the pivotal enzyme of triglyceride lipolysis, results in a murine phenotype that is characterized by progredient cardiac steatosis and severe heart failure. Since cardiac and vascular dysfunction have been closely related in numerous studies we investigated endothelium-dependent and -independent vessel function of ATGL knockout mice. Aortic relaxation studies and Langendorff perfusion experiments of isolated hearts showed that ATGL knockout mice suffer from pronounced micro- and macrovascular endothelial dysfunction. Experiments with agonists directly targeting vascular smooth muscle cells revealed the functional integrity of the smooth muscle cell layer. Loss of vascular reactivity was restored ~ 50% upon treatment of ATGL knockout mice with the PPARα agonist Wy14,643, indicating that this phenomenon is partly a consequence of impaired cardiac contractility. Biochemical analysis revealed that aortic endothelial NO synthase expression and activity were significantly reduced in ATGL deficiency. Enzyme activity was fully restored in ATGL mice treated with the PPARα agonist. Biochemical analysis of perivascular adipose tissue demonstrated that ATGL knockout mice suffer from perivascular inflammatory oxidative stress which occurs independent of cardiac dysfunction and might contribute to vascular defects. Our results reveal a hitherto unrecognized link between disturbed lipid metabolism, obesity and cardiovascular disease.  相似文献

The three-dimensional microvascular anatomy of the testes of adult Xenopus laevis was studied by scanning electron microscopy of vascular corrosion casts. Results showed that testicular arteries branch off from urogenital arteries and approach the testes via the mesorchium. At the dorsal surface of testes, arteries course towards the medial and lateral surfaces of the ovoid testes, pierce the tunica albuginea and run in the subalbugineal space and then penetrate the testicular parenchyma. In the parenchyma, they branch in all directions. At sites where larger branches arise, sphincters are present. Intimal cushions are found at the origin of smaller branches. Terminal arterioles capillarize and form a wide-meshed capillary network around convoluted seminiferous tubules. Inter- and peritubular capillaries cannot be differentiated reliably. Arterio-venous transition distances are short. Arterio-arterial anastomoses and veno-venous anastomoses are present. Testicular venules empty into a venous subalbugineal plexus which shows veno-venous anastomoses of different lengths and calibres. Signs of the ongoing non-sprouting angiogenesis, particularly intussusceptive branch remodelling, reflect the dynamics of the testicular parenchyma. The present study is the first that demonstrates the testicular microvascular anatomy in an anuran species and shows that X. laevis and men share the intricate convoluted seminiferous tubules with associated vascular patterns as a feature in common.  相似文献
