Background  

There are some early clinical indicators of cardiac ischemia, most notably a change in a person's electrocardiogram. Less well understood, but potentially just as dangerous, is ischemia that develops in the gastrointestinal system. Such ischemia is difficult to diagnose without angiography (an invasive and time-consuming procedure) mainly due to the highly unspecific nature of the disease.  相似文献

In this paper we address the question why slow-growing grass species appear to take up nitrate with greater respiratory costs than do fast-growing grasses when all plants are grown with free access to nutrients. Specific costs for nitrate transport, expressed as moles of ATP per net mole of nitrate taken up, were 1.5 to 4 times higher in slow-growing grasses than in fast-growing ones (Scheurwater et al., 1998, Plant, Cell & Environ. 21, 995–1005). The net rate of nitrate uptake is determined by two opposing nitrate fluxes across the plasma membrane: influx and efflux. To test whether differences in specific costs for nitrate transport are due to differences in the ratio of nitrate influx to net rate of nitrate uptake, nitrate influx and the net rate of nitrate uptake were measured in the roots of two fast-growing ( Dactylis glomerata L. and Holcus lanatus L.) and two slow-growing (Deschampsia flexuosa L. and Festuca ovina L.) grass species at four points during the diurnal cycle, using 15NO3 -. Efflux was calculated by subtraction of net uptake from influx; it was assumed that efflux of nitrogen represents the flux of nitrate. Transfer of the plants to the solution containing the labelled nitrate did not significantly affect nitrate uptake in the present grass species. The net rate of nitrate uptake was highest during the middle of the light period in all species. Diurnal variation in the net rate of nitrate uptake was mostly due to variation in nitrate influx. Variation in nitrate efflux did not occur in all species, but efflux per net mole of nitrate taken up was higher during darkness than in the light in the slow-growing grasses. The two fast-growing species, however, did not show diurnal variation in the ratio of efflux to net nitrate uptake. Integrated over 24 hours, the slow-growing grasses clearly exhibited higher ratios of influx to net uptake than the fast-growing grass species. Our results indicate that the higher ratio of nitrate influx to net nitrate uptake can account for higher specific costs for nitrate transport in slow-growing grass species compared with those in their fast-growing counterparts, possibly in combination with greater activity of the non-phosphorylating alternative respiratory path. Therefore, under our experimental conditions with plants grown at a non-limiting nitrate supply, nitrate uptake is less efficient (from the point of ATP consumption) in slow-growing grasses than in fast-growing grass species. Polar-desert plants experience low average air temperatures during their short growing season (4–8 °C mean July temperature). In addition, low availability of inorganic nitrogen in the soil may also limit plant growth. Our goals were to elucidate which N sources can be acquired by polar-desert plants, and how growth and N-uptake are affected by low growth temperatures. We compared rates of N-uptake and increases in mass and leaf area of two polar-desert species (Cerastium alpinum L. and Saxifraga caespitosa L.) over a period of 3 weeks when grown at two temperatures (6 °C vs. 15 °C) and supplied with either glycine, NH4 + or NO3 . At 15 °C, plants at least doubled their leaf area, whereas there was no change in leaf area at 6 °C. Measured mean N-uptake rates varied between 0.5 nmol g−1 root DM s−1 on glycine at 15 °C and 7.5 nmol g−1 root DM s−1 on NH4 + at 15 °C. Uptake rates based upon increases in mass and tissue N concentrations showed that plants had a lower N-uptake rate at 6 °C, regardless of N source or species. We conclude that these polar-desert plants can use all three N sources to increase their leaf area and support flowering when grown at 15 °C. Based upon short-term (8 h) uptake experiments, we also conclude that the short-term capacity to take up inorganic or organic N is not reduced by low temperature (6 °C). Nitrate influx, efflux and net nitrate uptake were measured for the slow-growing Quercus suber L. (cork-oak) to estimate the N-uptake efficiency of its seedlings when grown with free access to nitrate. We hypothesise that nitrate influx, an energetically costly process, is not very efficiently controlled so as to avoid losses through efflux, because Q. suber has relatively high respiratory costs for ion uptake. Q. suber seedlings were grown in a growth room in hydroponics with 1 mM NO3 -. Seedlings were labelled with 15NO3 - in nutrient solution for 5 min to measure influx and for 2 h for net uptake. Efflux was calculated as the difference between influx and net uptake. Measurements were made in the morning, afternoon and night. The site of nitrate reduction was estimated from the ratio of NO3 - to amino acids in the xylem sap; the observed ratio indicated that nitrate reduction occurred predominantly in the roots. Nitrate influx was always much higher than net acquisition and both tended to be lower at night. High efflux occurred both during the day and at night, although the proportion of 15NO3 - taken up that was loss through efflux was proportionally higher during the night. Efflux was a significant fraction of influx. We concluded that the acquisition system is energetically inefficient under the conditions tested. The effect of an elevated atmospheric CO2 concentration on growth, photosynthesis and root respiration of Plantago major L. ssp. major L. was investigated. Plants were grown in a nutrient solution in growth chambers at 350 and 700 μl I−1 CO2 during 7 weeks. The total dry weight of the Co2-enriched plants at the end of this period was 50% higher than that of control plants. However, the relative growth rate (RGR) was stimulated only during the first half of the growing period. The transient nature of the stimulation of the RGR was not likely to be due to end-product inhibition of photosynthesis. It is suggested that in P. major , a rosette plant, self-shading causes a decline in photosynthesis and results in an increase in the shoot: root ratio and a decrease in RGR. CO2-enriched plants grow faster and cosequently suffer more from self-shading. Corrected for this ontogenetic drift, high CO2 concentrations stimulated the RGR of P. major throughout the entire experiment.  相似文献

Abstract: This review focuses on the biochemical regulation and function of the alternative oxidase in vivo. About 10 years ago, two activation mechanisms were discovered in isolated mitochondria, namely activation by reducing sulfur bonds in the protein and activation by an allosteric effect of pyruvate. It was proposed that plants would have a regulatory mechanism to modify alternative oxidase activity in vivo. However, more recent studies have shown that these two activation mechanisms may not play such an important role in regulation of alternative oxidase activity in vivo after all. Pyruvate and reduction of the sulfide bonds in the protein are definitely required for alternative oxidase activity, but they do not appear to be regulating the activity in vivo.
Despite the energy wasting nature of the alternative oxidase, there was no obvious physiological function for the pathway for many years. It is now more clear that the alternative oxidase can prevent the production of excess reactive oxygen species radicals by stabilizing the redox state of the mitochondrial ubiquinone pool, while allowing continued activity of the citric acid cycle. This may be important under conditions when the NADH supply is relatively high (reductant overflow), or when the cytochrome pathway is restrained. The cytochrome pathway might be inhibited by naturally occurring cyanide, nitric oxide, sulfide, high concentrations of CO2, low temperatures, or by limited phosphate supply.  相似文献

Are there intrinsic differences in the rates of photosynthesis, shoot- and root-respiration between inherently fast- and slow-growing monocotyledons at high and low nitrogen supply? To analyze this question we grew 5 monocotyledons, widely differing in their inherent relative growth rate at high and low nitrogen supply in a growth room. Nitrate was exponentially added to the plants, enabling us to compare inherent differences in plant characteristics, without any effect of species differences in the ability to take up nutrients. At high nitrogen supply, the fast-growing species from productive habitats had a higher photosynthetic nitrogen use efficiency and rate of root respiration than the slow-growing ones from unproductive habitats. Only minor differences were observed in their rates of photosynthesis and shoot respiration per unit leaf area. At low nitrogen supply, the rates of photosynthesis and shoot- and root respiration decreased for all species, even though there were no longer any differences in these processes between inherently fast- and slow-growing species. The photosynthetic nitrogen use efficiency increased for all species, and no differences were found among species. Differences in the photosynthetic nitrogen use efficiency among species and nitrogen treatments are discussed in terms of the utilization of the photosynthetic apparatus, whereas differences in respiration rate are discussed in terms of the energy demand for growth, maintenance and ion uptake and their related specific respiratory energy costs. It is concluded that the relatively high abundance of slow-growing species compared to fast-growing ones in unproductive habitats is unlikely to be explained by differences in rates of photosynthesis and respiration or in photosynthetic nitrogen use efficiency.  相似文献

BACKGROUND: Global phosphorus (P) reserves are being depleted, with half-depletion predicted to occur between 2040 and 2060. Most of the P applied in fertilizers may be sorbed by soil, and not be available for plants lacking specific adaptations. On the severely P-impoverished soils of south-western Australia and the Cape region in South Africa, non-mycorrhizal species exhibit highly effective adaptations to acquire P. A wide range of these non-mycorrhizal species, belonging to two monocotyledonous and eight dicotyledonous families, produce root clusters. Non-mycorrhizal species with root clusters appear to be particularly effective at accessing P when its availability is extremely low. SCOPE: There is a need to develop crops that are highly effective at acquiring inorganic P (Pi) from P-sorbing soils. Traits such as those found in non-mycorrhizal root-cluster-bearing species in Australia, South Africa and other P-impoverished environments are highly desirable for future crops. Root clusters combine a specialized structure with a specialized metabolism. Native species with such traits could be domesticated or crossed with existing crop species. An alternative approach would be to develop future crops with root clusters based on knowledge of the genes involved in development and functioning of root clusters. CONCLUSIONS: Root clusters offer enormous potential for future research of both a fundamental and a strategic nature. New discoveries of the development and functioning of root clusters in both monocotyledonous and dicotyledonous families are essential to produce new crops with superior P-acquisition traits.  相似文献

The species Urtica dioica L., Plantago major ssp. major L., Plantago lanceolata L., Hypochaeris radicata L. ssp. radicata and Hypochaeris radicata ssp. ericetorum Van Soest were grown under high and low nutrient conditions (1/4 Hoagland and 2% of 1/4 Hoagland further called the 100% and 2% treatment, containing 3.75 mM NO-3 and 0.075 mM NO-3, respectively). After a certain period half of the plants were transferred from low to high or high to low nutrients, yielding the 100%/2% and the 2%/100% treatments. The kinetics of nitrate uptake in the range of system I of the five species grown under the different nutrient conditions were measured during a three week experimental period. The nitrate uptake of all the species showed the characteristic features of Michaelis-Menten kinetics. Under low nutrient conditions the apparent Vmax of U. dioica expressed per g dry root was lower than under high nutrient conditions. For H. radicata ssp. radicata and for H. radicata ssp. ericetorum the reverse was found. The Vmax values of P. major ssp. major were almost the same for the two treatments. The apparent Vmax in young plants of P. lanceolata was higher in the 100% treatment than in 2%; whereas the reverse was found in mature plants. The results are explained in relation to the relative growth rate, the shoot to root ratio and the natural environment of the species. The apparent Km values were not influenced by the different treatments. Differences in Km between the species, if any, were very small. It is suggested that the Vmax is a more important parameter for the distribution of plant species in the field than the Km. The rate of nitrogen accumulation was calculated from growth data and the contents of nitrate and reduced nitrogen. It is concluded that the Vmax of system I for nitrate uptake in most cases was sufficient to explain the observed growth rates.  相似文献
