This study was designed to investigate whether thermotolerant roots exhibit respiratory acclimation to elevated temperatures. Root respiratory acclimation traits in response to increasing temperatures were compared between two Agrostis species contrasting in heat tolerance: thermal A. scabra and heat-sensitive A. stolonifera. Roots of both species were exposed to 17, 27, or 37 degrees C. Root RGR declined with increasing temperatures from 17 degrees C to 37 degrees C in both species; however, root growth of A. scabra maintained a significantly higher RGR than A. stolonifera at 27 degrees C or 37 degrees C. A. scabra exhibited a significantly higher respiration acclimation potential to elevated temperatures, both in the short term (60 min) and in the long term (7-28 d) as compared with A. stolonifera, when temperatures increased from 17 degrees C to 27 degrees C or from 27 degrees C to 37 degrees C. Thermal A. scabra also maintained a significantly lower maintenance cost than A. stolonifera as temperatures increased to 27 degrees C or 37 degrees C. The results suggested that root thermotolerance of thermal A. scabra was associated with both short-term and long-term respiratory acclimation to changes in temperatures. The superior ability of adjusting the rate of root respiration to compensate for increases in carbon demand during short- or long-term temperature increases in the heat-tolerant A. scabra may result in the reduction in carbon expenditure or costs for maintenance, leading to extended root survivability in high temperature soils.  相似文献

Even though the growth‐promoting effects of gibberellins (GAs) in plants are well established, little is known about GA action on carbon metabolism and the available reports seem contradictory. We studied the effects of GA deficiency in mutants of tomato (Solanum lycopersicum L.) on rates of carbon acquisition and the allocation of acquired carbon to growth and respiration of leaves, stems and roots. Carbon budgets were calculated from 24 h measurements of photosynthesis and respiration. The partitioning of nitrogen compounds to leaves, stems and roots, which strongly influences carbon budgets, was also studied. The GA‐deficient mutants acquired less carbon per unit plant mass per day than did the wild type and used a larger fraction of it for root growth and root respiration. To find out to what extent these changes were just consequences of restriction of growth, the experiment was repeated at a low exponential nitrate addition rate, which forced all genotypes to grow at the same rate. Under these conditions, the low‐GA mutants still photosynthesized and respired faster and partitioned more carbon to root growth than the wild type did. The reasons for the observed differences in carbon economies between the wild type and the low‐GA mutants are discussed.  相似文献

The light subunit of mushroom, Agaricus bisporus, tyrosinase (LSMT), has been identified as an extrinsic component of the enzyme. Its function is unknown, but it can cross an epithelial cell layer, which suggests that it can be absorbed by the intestine. A similar capability has been demonstrated for the HA-33 component of the progenitor toxin from Clostridium botulinum, which is the closest structural homolog of LSMT. Unlike HA-33, LSMT appears to be non-immunogenic as shown by preliminary tests in Swiss Webster mice. We investigated the immunogenicity and histopathology of LSMT in mice to determine its safety in vivo. LSMT did not evoke generation of antibodies after prolonged periods of intraperitoneal administration. Histopathological observations confirmed the absence of responses in organs after twelve weekly administrations of LSMT. We found that LSMT is not toxic and is less immunogenic than the C. botulinum HA-33 protein, which supports further research and development for pharmaceutical application.  相似文献

Aims

Rytidosperma species are native Australian grasses which have different growth rates and phosphorus (P) requirements. This study examined the role of root morphology traits in response to P supply.

Methods

Nine Rytidosperma species ranging from slow- to fast-growth were examined along with Lolium perenne and Bromus hordeaceus. Plants were grown in a glasshouse for 47 days in soil supplied with six levels of P between 0 and 60 mg P per pot. Root mass, length and diameter, root hair length and density, and extent of mycorrhizal colonisation were measured.

Results

Across all species there was a positive correlation (P < 0.001) between P uptake and root mass, length and root hair cylinder volume (RHCV; estimated using root diameter, root hair length and root length) at all levels of P supply. An exception was the RHCV of B. hordeaceus, where expected P uptake was not achieved due to a markedly reduced root length at low-P supply. For the Rytidosperma species, morphological plasticity for specific root length, root mass fraction and root hair length ranged from 1.5-fold to 2.7-fold between high- and low-P supply. However, across all species and P levels no single root morphological trait was identified for universally increasing the size of the root system and P uptake.

Conclusions

Fast-growing species took up more P as a result of an overall larger root mass, greater root length and larger RHCV.

Aims

Trifolium subterraneum L. is the predominant annual pasture legume in southern Australia. Cultivars with improved phosphorus (P) foraging ability would improve the P-use efficiency of agricultural systems. We therefore investigated variation in root traits related to P-uptake among six cultivars.

Methods

Micro-swards were grown at six levels of P in field soil with indigenous arbuscular mycorrhizal (AM) fungi for six weeks. Dry matter yield, tissue P concentration, rhizosphere carboxylates, AM fungal colonisation and root morphological traits were measured.

Results

The cultivars showed similar shoot and root yield responses to P supply. Average root diameter did not change, specific root length (SRL) increased and root tissue density (RTD) decreased with increased P supply. Amounts of total rhizosphere carboxylates were low (<1.2 nmol cm?1 root). The percentage of root length colonised by AM fungi was greatest (29–43 %) at an intermediate level (8 mg kg?1 dry soil) of P supply.

Conclusions

Most differences among cultivars were reasonably consistent across P supply levels, indicating greater numbers of lines could be screened reliably at a single P level. Low colonisation by AM fungi at low P supply deserves consideration when selecting soils for cultivar comparisons. Increased SRL and decreased RTD at high P supply likely result from self-shading within the micro-swards and warrant further investigation.

Aims Epiphytes and hemiparasites do not have direct access to soil nutrients. Epiphytes acquire nutrients through symbiosis, foliar leachates and throughfall, whilst hemiparasites have specialized structures (haustoria) to acquire nutrients from their host. Irrespective of the green leaf nutrient concentrations of epiphytes, hemiparasites and their hosts, nutrient-resorption efficiency and proficiency are expected to be the greater in epiphytes than in their hosts and in hemiparasites. The role of gibberellin (GA) in leaf elongation has long been known, however, its involvement in whole shoot growth and biomass allocation is much less clear. We studied the effects of exogenously supplied GA3 and paclobutrazol, an inhibitor of GA biosynthesis, on these processes in Aegilops caudata and Aegilops tauschii , species with contrasting leaf growth characteristics. In both species, addition of GA3 increased leaf elongation rate (LER) through its promoting effect on both cell size and cell number, while paclobutrazol decreased it. Similarly, GA3 increased biomass allocation to the leaves, mainly leaf sheaths, at the cost of allocation to the roots, whereas paclobutrazol had the opposite effect in both species. Despite the increase in LER and biomass allocation to the shoot upon GA3 application, the relative growth rate (RGR) remained constant. Specific leaf area (SLA) was only temporarily affected by GA3 addition. Our results show that the inherent differences in LER and biomass allocation between the slow-elongating A. caudata and the fast-elongating A. tauschii are considerably reduced by the exogenous supply of GA3 to the slow-elongating species, or paclobutrazol to the fast-elongating one. This suggests a role for gibberellins in explaining inherent differences in leaf area expansion and biomass allocation between the two species in this study.  相似文献
