The permanently eutrophic Sundays Estuary experiences recurrent harmful algal blooms (HABs) of Heterosigma akashiwo (Raphidophyceae). This study aimed to identify the environmental variables shaping phytoplankton community composition and succession patterns during a typical spring/summer harmful algal bloom (HAB) period. Monitoring of abiotic and phytoplankton variables was undertaken over the period of a month in 2016. Surface water salinity corresponding to mesohaline conditions (9 to 12) was a prerequisite for site selection. During the study, two HABs (>550 µg Chl a l?1) of H. akashiwo occurred, each lasting for approximately a week in duration. Analyses highlighted nutrient depletion (i.e. nitrate and phosphate concentrations) as the key constraint on bloom duration. When the density of H. akashiwo decreased, the community composition became more diverse with species belonging to Bacillariophyceae and Dinophyceae becoming more abundant; albeit to a lesser degree (<180 µg Chl a l?1). Dissolved oxygen shifted from super-saturated conditions (>14 mg l?1) during peak HAB conditions, to instances of bottom water oxygen depletion (2–4 mg l?1) during the decay phase. These findings highlight the potential severity of transforming a catchment from natural to one that is highly regulated by agricultural practices, while also emphasising the need for management intervention. 相似文献

The nonbiting midge subfamily Tanypodinae represents one of the most diverse lineages of Chironomidae. Despite the wide distribution and high diversity of tanypodine chironomids, the evolutionary history of the subfamily remains poorly understood. Here, we present the first phylogenetic analysis of the subfamily Tanypodinae based on morphological data. Cladistic analyses were conducted using 86 morphological characters from 115 species belonging to 54 tanypodine genera, including the eight currently recognised tribes: Anatopyniini, Clinotanypodini, Coelopyniini, Macropelopiini, Natarsiini, Pentaneurini, Procladiini and Tanypodini. We use characters from fourth‐instar larvae, pupae and adults of both sexes. We examine the effects of implied weighting by reanalysing the data with varying values of concavity constant (k). Our analysis supports the monophyly of Tanypodinae with Podonominae as its sister group. All previously proposed tribes are recovered as monophyletic assemblages under a wide range of weighting factors. Under these conditions, the genus Fittkauimyia is the sister group of the remaining Macropelopiini and is erected as a new monobasic tribe, Fittkauimyiini trib.n . The tribe Pentaneurini is recovered as monophyletic with some internal relationships resolved. The genus Paramerina, recovered as sister of Reomyia + Zavrelimyia, is formally synonymised with Zavrelimyia syn.n. , based on morphological similarity in all three life stages and treated as a subgenus of the latter. Finally, the recently suggested synonymies of Gressittius and Guassutanypus with Alotanypus and the establishment of the subgenera Conchapelopia (Helopelopia), Macropelopia (Bethbilbeckia), Monopelopia (Cantopelopia), Thienemannimyia (Hayesomyia) and Zavrelimyia (Reomyia and Schineriella) are investigated. Our results support all proposed changes, except for the subgenus‐level status of Helopelopia and Cantopelopia. We suggest re‐establishment of Helopelopia as a genus, but refrain from promoting genus‐level status of Cantopelopia at present because the apparent sister‐relationship between Monopelopia + Nilotanypus likely is due to wing vein reduction caused by miniaturisation. 【背景】芳樟醇具有特殊的香气和多种生物学活性,是食品、医药和化妆品行业的重要原料。随着合成生物学的高速发展,代谢改造微生物进行芳樟醇生物合成是当前研究的一大热点。然而在微生物的生物合成中,芳樟醇对底盘细胞的毒性是一大瓶颈问题,也是其他单萜物质生物合成的共性问题。【目的】建立合理的耐受性改造方法,以提高微生物宿主细胞对芳樟醇的耐受性。【方法】以酿酒酵母BY4741为研究对象,通过对ABC转运蛋白、活性氧调控相关酶及转录调控因子的过表达,考察它们对酿酒酵母芳樟醇耐受性的影响,并通过对酿酒酵母细胞进行定向驯化,筛选耐受性提高的酿酒酵母突变株。【结果】单独过表达ABC转运蛋白(Yor1、Snq2、Pdr5、Pdr15和Pdr18)、ROS调控相关酶(Gre2、Ctt1、Yhb1、Gpx2、Trr1、Trx2和Gsh2)及转录调控因子(Ino2、Yap1、Yap5和Stb5)并不能有效提高酿酒酵母的耐受性,但在传代适应性驯化过程中获得了两株耐受性提高的酿酒酵母突变株,将芳樟醇的致死浓度从430mg/L提高到了645mg/L以上。进一步通过基因组重测序分析揭示了驯化菌株突变位点。其中YBR074W... 相似文献

Aedes aegypti (L.) (Diptera: Culicidae), the main vector of yellow fever and dengue viruses, was eradicated from Argentina between 1955 and 1963, but reinvaded the country in 1986. In Uruguay, the species was reintroduced in 1997. In this study we used highly polymorphic inter‐simple sequence repeats (ISSR) markers to analyse the genetic structure of Ae. aegypti populations from Uruguay and northeastern Argentina to identify possible colonization patterns of the vector. Overall genetic differentiation among populations was high (FST = 0.106) and showed no correlation with geographic distance, which is consistent with the short time since the reintroduction of the species in the area. Differentiation between pairs of Argentine populations (FST 0.072 to 0.221) was on average higher than between Uruguayan populations (FST?0.044 to 0.116). Bayesian estimation of population structure defined four genetic clusters and most populations were admixtures of two of them: Mercedes and Treinta y Tres (Uruguay) were mixtures of clusters 1 and 3; Salto (Uruguay) and Paraná (Argentina) of clusters 1 and 4; Fray Bentos (Uruguay) of clusters 2 and 3, and Gualeguaychú (Argentina) of clusters 2 and 3. Posadas and Buenos Aires in Argentina were fairly genetically homogeneous. 1. The life history of wood feeders was modelled in order to explain the multiseasonality of development and the great variability of adult size in this group. 2. The model was parameterised with experimental bioenergetic and reproductive data for the xylem feeder Aredolpona rubra (Coleoptera: Cerambycidae). 3. The length of the developmental period, which together with food quality directly determines adult size and indirectly determines the number of eggs laid, was optimised. 4. The results show that multi‐seasonal larval development maximises fitness under conditions of low food quality, relatively low predation pressure, and the presence of hostile periods during the year. 5. The variability of the number of seasons needed to complete development within a wood‐feeder population is a consequence of development time optimisation and the unavoidable extension of the egg‐laying period. These insects have an evolutionary dilemma: to eclose late in a given season at smaller size, bringing about low egg production and low offspring value, or to grow bigger to the next season, jeopardising their lives. 6. Abstract: The first mygalomorph spiders from the Lower Cretaceous Crato Lagerstätte of Cearà Province, north-east Brazil, are described, from adult males and females, in two new genera and species: Cretadiplura ceara Selden, gen. et sp. nov. and Dinodiplura ambulacra Selden, gen. et sp. nov. When [l-14C]-malonate was supplied to discs cut from matureleaves of Coffea arabica, 14CO2 was released (approximately12% of the total CO2 respired) and organic acids of the Krebscycle, uronic acids, sugars and amino acids became radioactive.There was no incorporation of MC into either lipids or phenoliccompounds. The formation of glucose from malonate has not beenobserved in other studies with plant tissues. The synthesisof labelled glucose together with an active pentose phosphatepathway that is stimulated by malonate explains the accumulationof radioactive phosphogluconate in the leaf discs. The crystal and molecular structure of potassium β-d-glucopyranose 6-sulphate has been determined by direct methods. The sugar ring has the expected 4C1 conformation although the sulphate group causes flattening of the ring. The potassium ion has octahedral co-ordination involving oxygen atoms of five independent β-d-glucopyranose 6-sulphate molecules. The n.m.r. spectrum of the 6-sulphate in the solid state is consistent with the occurrence of two molecules in the unit cell related by a 21 symmetry axis. 相似文献
