This study reports the purification and biochemical characterization of a raw starch-digesting α-amylase from Geobacillus thermoleovorans subsp. stromboliensis subsp. nov. (strain PizzoT). The molecular weight was estimated to be 58 kDa by SDS–PAGE. The enzyme was highly active over a wide range of pH from 4.0–10.0. The optimum temperature of the enzyme was 70°C. It showed extreme thermostability in the presence of Ca2+, retaining 50% of its initial activity after 90 h at 70°C. The enzyme efficiently hydrolyzed 20% (w/v) of raw starches, concentration normally used in starch industries. The α-amylase showed an high stability in presence of many organic solvents. In particular the residual activity was of 73% in presence of 15% (v/v) ethyl alcohol, which corresponds to ethanol yield in yeast fermentation process. By analyzing its complete amyA gene sequence (1,542 bp), the enzyme was proposed to be a new α-amylase.  相似文献

JCC76 is a derivative of cyclooxygenase-2(COX-2) selective inhibitor nimesulide and exhibits potent anti-breast cancer activity. It selectively induces apoptosis of Her2 positive breast cancer cells. However, the specific molecular targets of JCC76 still remain unclear, which significantly withdraw the further drug development of JCC76. To identify the molecular targets of JCC76, a six carbon linker and biotin conjugated JCC76 probe was designed and synthesized. The anti-proliferation activity of the probe and its analogs was evaluated.  相似文献

This contribution describes a new laboratory experience that improves medical students' learning of chronobiology by introducing them to basic chronobiology concepts as well as to methods and statistical analysis tools specific for circadian rhythms. We designed an autorhythmometry laboratory session where students simultaneously played the role of researchers and experimental subjects. During this session, which lasted 24 h, students recorded their own arterial pressure, heart rate, oral temperature, forced expiratory flow, glucose tolerance, muscular strength, reaction time, and sensorimotor coordination at regular intervals and also took the Horne and Ostberg test, after which they analyzed their own data. Furthermore, to gather information from subjects under normal sleep and eating schedules, some students acquired data at home. To guide and help students with their work, a dedicated web page was implemented with scientific references, cosinor analysis software, and other valuable information. All these "raw" data were combined into a single database that students could use to evaluate whatever aspect of the data they seemed fit. A number of suggestions were offered to them as guidance. Students were then instructed to write a scientific article on the subject they had chosen. The experience was highly rewarding for both instructors and students alike. In view of the high level of absenteeism in Spanish universities and the fact that 93% of the students attended the exam and 95% of these passed, the experience was considered a great success.  相似文献

Euglossa cyanochlora Moure, 1996 (Hymenoptera: Apidae: Euglossina) is known from only two specimens and is considered as one of the rarest bees. An intensive search for this species in its expected area of occurrence in eastern Brazil was carried out during three consecutive summers and involved over 1,000?h of active sampling in 50 sites distributed among 20 forest remnants. Almost 15,000 orchid bee males were attracted to scent baits and collected, but only 24 of them belonged to E. cyanochlora. Some degree of mitochondrial gene (COI, cytB, and 16S) variation was observed in this species. Although no intra-specific variation was identified at the 16S locus, three and six haplotypes were characterized at the COI and cytB genes, respectively. Combined data from field work and molecular analysis suggest that the species southernmost distribution limit is close to its type locality, near Parque Nacional do Monte Pascoal, in southern Bahia. The conservation status of E. cyanochlora is discussed and measures for its protection are here proposed.  相似文献

Introduction

Animal travel speed is an ecologically significant parameter, with implications for the study of energetics and animal behaviour. It is also necessary for the calculation of animal paths by dead-reckoning. Dead-reckoning uses heading and speed to calculate an animal’s path through its environment on a fine scale. It is often used in aquatic environments, where transmission telemetry is difficult. However, its adoption for tracking terrestrial animals is limited by our ability to measure speed accurately on a fine scale. Recently, tri-axial accelerometers have shown promise for estimating speed, but their accuracy appears affected by changes in substrate and surface gradients. The purpose of the present study was to evaluate four metrics of acceleration; Overall dynamic body acceleration (ODBA), vectorial dynamic body acceleration (VDBA), acceleration peak frequency and acceleration peak amplitude, as proxies for speed over hard, soft and inclined surfaces, using humans as a model species.

Results

A general linear model (GLM) showed a significant difference in the relationships between the metrics and speed depending on substrate or surface gradient. When the data from all surface types were considered together, VeDBA had the highest coefficient of determination.

Conclusions

All of the metrics showed some variation in their relationship with speed according to the surface type. This indicates that changes in the substrate or surface gradient during locomotion by animals would produce errors in speed estimates, and also in dead-reckoned tracks if they were calculated from speeds based entirely on a priori calibrations. However, we describe a method by which the relationship between acceleration metrics and speed can be corrected ad hoc, until tracks accord with periodic ground truthed positions, obtained via a secondary means (e.g. VHF or GPS telemetry). In this way, dead-reckoning provides a means to obtain fine scale movement data for terrestrial animals, without the need for additional data on substrate or gradient.  相似文献

Renin-angiotensin-aldosterone system (RAAS) activation mediates increases in reactive oxygen species (ROS) and impaired insulin signaling. The transgenic Ren2 rat manifests increased tissue renin-angiotensin system activity, elevated serum aldosterone, hypertension, and insulin resistance. To explore the role of aldosterone in the pathogenesis of insulin resistance, we investigated the impact of in vivo treatment with a mineralocorticoid receptor (MR) antagonist on insulin sensitivity in Ren2 and aged-matched Sprague-Dawley (SD) control rats. Both groups (age 6-8 wk) were implanted with subcutaneous time-release pellets containing spironolactone (0.24 mg/day) or placebo over 21 days. Systolic blood pressure (SBP) and intraperitoneal glucose tolerance test were determined. Soleus muscle insulin receptor substrate-1 (IRS-1), tyrosine phosphorylated IRS-1, protein kinase B (Akt) phosphorylation, GLUT4 levels, and insulin-stimulated 2-deoxyglucose uptake were evaluated in relation to NADPH subunit expression/oxidase activity and ROS production (chemiluminescence and 4-hydroxy-2-nonenal immunostaining). Along with increased soleus muscle NADPH oxidase activity and ROS, there was systemic insulin resistance and reduced muscle IRS-1 tyrosine phosphorylation, Akt phosphorylation/activation, and GLUT4 expression in the Ren2 group (each P < 0.05). Despite not decreasing blood pressure, low-dose spironolactone treatment improved soleus muscle insulin signaling parameters and systemic insulin sensitivity in concert with reductions in NADPH oxidase subunit expression/activity and ROS production (each P < 0.05). Our findings suggest that aldosterone contributes to insulin resistance in the transgenic Ren2, in part, by increasing NADPH oxidase activity in skeletal muscle tissue.  相似文献

Several marine thermophilic strains were analyzed for exopolysaccharide production. The screening process revealed that a significant number of thermophilic microorganisms were able to produce biopolymers, and some of them also revealed interesting chemical compositions. We have identified four new polysaccharides from thermophilic marine bacteria, with complex primary structures and with different repetitive units: a galacto-mannane type from strain number 4004 and mannane type for the other strains. The thermophilic Bacillus thermantarcticus produces two exocellular polysaccharides (EPS 1, EPS 2) that give the colonies a typical mucous character. The exopolysaccharide fraction was produced with all substrates assayed, although a higher yield 400 mg liter(-1) was obtained with mannose as carbon and energy source. NMR spectra confirmed that EPS 1 was a heteropolysaccharide of which the repeating unit was constituted by four different alpha-D-mannoses and three different beta-D-glucoses. It seems to be close to some xantan polymers. EPS 2 was a mannan. Four different alpha-D-mannoses were found as the repeating unit. Production and chemical studies of biopolymers produced by halophilic archaea, Haloarcula species were also reported.  相似文献

Thermoacidophilic and halotolerant microorganisms from the Antarctic continent were studied for their lipid modulation under stress growth conditions. Temperature-induced changes in complex lipids and fatty acids of four strains belonging to the genus Alicyclobacillus involved the relative proportions of different polar lipids and the synthesis of ω-cyclohexyl-acyl chains, which were favoured by high temperatures. Studies were carried out on the lipid composition of four strains of extremely halotolerant bacteria belonging to the genus Micrococcus grown at different salt concentrations from 0 up to 4.5 M NaCl. The main lipids found were two unidentified glycolipids and two phospholipids: 1,2 diacylglycero-3-phosphoryl-glycerol (PG) and cardiolipin (DPG). Among the strains analysed, the lipids of the Micrococcus strain Erebus were shown to be strongly influenced by salt concentrations, in that DPG and one glycolipid were absent at a low salt molarity while, under these conditions, PG was the main lipid found. The predominant fatty acids in all halotolerant strains were of the anteiso type; growth under increasing salinity gave rise to an increase in long chain fatty acids and of straight chain fatty acids, while a decrease in iso fatty acids occurred. Monodispersed silver (Ag) nanoparticles (NPs) were obtained by applying an electric field on unipolar-charged Ag NPs fabricated using a heterogeneous condensation technique in gas media. Well defined and charged Ag NPs were separated based on their sizes and were collected on quartz substrates. Thin films consisting of monodispersed Ag NPs with size ranging from 35 to 120 nm were prepared by varying an applied electric field during the fabrication process. Scanning electron microscope results showed that the samples have uniform size distribution. Coherent oscillations of conduction band electrons in gas medium induced by electromagnetic field and coupling of all similar plasmon resonances due to uniform Ag NPs size produced unique and interesting optical properties. Narrow extinction widths (~41 to ~69 nm) were observed compared to the width of polydispersed Ag sample. The ability to prepare samples in gas media and tune the plasmon resonance by merely varying an electric field during fabrication makes the method simple, fast, and highly economical.  相似文献
