Five-membered-ring heterocyclic urea mimics have been found to be potent and selective antagonists of the P2Y1 receptor. ADP receptors, P2Y1 and P2Y12 have been recognized as potential targets for antithrombotic drugs. A series of P2Y1 antagonists that contain 2-aminothiazoles as urea surrogates were discovered. Extensive SAR of the thiazole ring is described. The most potent compound 7j showed good P2Y1 binding (Ki = 12 nM), moderate antagonism of platelet aggregation (PA IC50 = 5.2 μM) and acceptable PK in rats.  相似文献

Purpose

In response to the increasing concerns on the environmental conservation and energy saving, manufacturers are more aware of proving the ‘green’ performance of their products. Some qualitative eco design tools are used to support the development of greener products; however, most of these tools require subjective judgement during the evaluation processes. This paper is therefore to propose an alternative approach that is objective, systematic and efficient, by integrating the ant colony optimization (ACO) and life cycle assessment (LCA), to facilitate the decision-making process.

Methods

The proposed integrative LCA-ACO approach aims to support the simultaneous thorough evaluations of multiple design options. A sequence of options of the lowest corresponding environmental impact value can be obtained. A case application example of various design combinations is presented to demonstrate the applicability of the proposed approach.

Results and discussion

The proposed approach offers decision makers a preliminary fast-track approach for screening decisions without lengthy processes of LCA studies. This approach helps the decision makers, especially during the early design selection stages, identifying the most appropriate design combination from the environmental perspective. The proposed approach is proved a significant contribution to the field of LCA and green product design.

Conclusions

Since full-scale LCA studies require significant effort in data collection processes and experts for result interpretations, it would be time consuming and costly to conduct a full-scale LCA during early product development processes. The proposed approach offers a more convenient way for decision makers to assess multiple design options regarding the environmental considerations. The case example presented in this paper proves the practicality of the proposed approach.

Protein degradation by aminopeptidases is involved in bacterial responses to stress. Escherichia coli produces two metal‐dependent M17 family leucine aminopeptidases (LAPs), aminopeptidase A (PepA) and aminopeptidase B (PepB). Several structures have been solved for PepA as well as other bacterial M17 peptidases. Herein, we report the first structures of a PepB M17 peptidase. The E. coli PepB protein structure was determined at a resolution of 2.05 and 2.6 Å. One structure has both Zn2+ and Mn2+, while the second structure has two Zn2+ ions bound to the active site. A 2.75 Å apo structure is also reported for PepB from Yersinia pestis. Both proteins form homohexamers, similar to the overall arrangement of PepA and other M17 peptidases. However, the divergent N‐terminal domain in PepB is much larger resulting in a tertiary structure that is more expanded. Modeling of a dipeptide substrate into the C‐terminal LAP domain reveals contacts that account for PepB to uniquely cleave after aspartate.  相似文献

Cyanobacteria require efficient protein-quality-control mechanisms to survive under dynamic, often stressful, environmental conditions. It was reported that three serine proteases, HtrA (high temperature requirement A), HhoA (HtrA homologue A) and HhoB (HtrA homologue B), are important for survival of Synechocystis sp. PCC 6803 under high light and temperature stresses and might have redundant physiological functions. In the present paper, we show that all three proteases can degrade unfolded model substrates, but differ with respect to cleavage sites, temperature and pH optima. For recombinant HhoA, and to a lesser extent for HtrA, we observed an interesting shift in the pH optimum from slightly acidic to alkaline in the presence of Mg2+ and Ca2+ ions. All three proteases formed different homo-oligomeric complexes with and without substrate, implying mechanistic differences in comparison with each other and with the well-studied Escherichia coli orthologues DegP (degradation of periplasmic proteins P) and DegS. Deletion of the PDZ domain decreased, but did not abolish, the proteolytic activity of all three proteases, and prevented substrate-induced formation of complexes higher than trimers by HtrA and HhoA. In summary, biochemical characterization of HtrA, HhoA and HhoB lays the foundation for a better understanding of their overlapping, but not completely redundant, stress-resistance functions in Synechocystis sp. Palm oil industry is one of the leading agricultural industries in Malaysia with average crude palm oil production of more than 13 million tonne per year. However, production of such huge amount of crude palm oil has consequently resulted to even larger amount of palm oil mill effluent (POME). POME is a highly polluting wastewater with high chemical oxygen demand (COD) and biochemical oxygen demand (BOD) in which can caused severe pollution to the environment, typically pollution to water resources. On the other hand, POME was identified as a potential source to generate renewable bioenergies such as biomethane and biohydrogen through anaerobic digestion. In other words, a combination of wastewater treatment and renewable bioenergies production would be an added advantage to the palm oil industry. In line with the world's focus on sustainability concept, such strategy should be implemented immediately to ensure palm oil is produced in an environmental friendly and sustainable manner. This review aims to discuss various technologies to convert POME to biomethane and biohydrogen in a commercial scale. Furthermore, discussion on using POME to culture microalgae for biodiesel and bioethanol production was included in the present paper as a new remedy to utilize POME with a greater beneficial return.  相似文献

Danshen-Gegen (DG) decoction, an herbal formulation comprising Radix Salvia Miltiorrhiza and Radix Puerariae Lobatae, is prescribed for the treatment of coronary heart disease in Chinese medicine. Experimental and clinical studies have demonstrated that DG decoction can reduce the extent of atherosclerosis. In the present study, using an ex vivo rat model of myocardial ischemia/reperfusion (I/R) injury, we investigated the myocardial preconditioning effect of an aqueous DG extract prepared from an optimized weight-to-weight ratio of Danshen and Gegen. Short-term treatment with DG extract at a daily dose of 1 g/kg and 2 g/kg for 3 days protected against myocardial I/R injury in rats. The cardioprotection afforded by DG pretreatment was paralleled by enhancements in mitochondrial antioxidant status and membrane structural integrity, as well as a decrease in the sensitivity of mitochondria to Ca2+-stimulated permeability transition in vitro, particularly under I/R conditions. Short-term treatment with the DG extract also enhanced the translocation of PKC? from the cytosol to mitochondria in rat myocardium, and this translocation was inhibited by α-tocopherol co-treatment with DG extract in rats. Short-term DG treatment may precondition the myocardium via a redox-sensitive PKC?/mKATP pathway, with resultant inhibition of the mitochondrial permeability transition through the opening of mitochondrial KATP channels. Our results suggest that clinical studies examining the effectiveness of DG extract given prophylactically in affording protection against myocardial I/R injury would be warranted.  相似文献

The effects of pH and bulking agents on hydrolysis/acidogenesis of food waste were studied using leach bed reactor (LBR) coupled with methanogenic up-flow anaerobic sludge blanket (UASB) reactor. The hydrolysis rate under regulated pH (6.0) was studied and compared with unregulated one during initial experiment. Then, the efficacies of five different bulking agents, i.e. plastic full particles, plastic hollow sphere, bottom ash, wood chip and saw dust were experimented under the regulated pH condition. Leachate recirculation with 50% water replacement was practiced throughout the experiment. Results proved that the daily leachate recirculation with pH control (6.0) accelerated the hydrolysis rate (59% higher volatile fatty acids) and methane production (up to 88%) compared to that of control without pH control. Furthermore, bottom ash improved the reactor alkalinity, which internally buffered the system that improved the methane production rate (0.182 l CH4/g VSadded) than other bulking agents.  相似文献

How polytopic plasma membrane (PM) proteins reach their destination in plant cells remains elusive. Using transgenic tobacco BY-2 cells, we previously showed that the rice secretory carrier membrane protein 1 (SCAMP1), an integral membrane protein with four transmembrane domains (TMDs), is localized to the PM and trans-Golgi network (TGN). Here, we study the transport pathway and sorting signals of SCAMP1 by following its transient expression in tobacco BY-2 protoplasts and show that SCAMP1 reaches the PM via an endoplasmic reticulum (ER)-Golgi-TGN-PM pathway. Loss-of-function and gain-of-function analysis of various green fluorescent protein (GFP) fusions with SCAMP1 mutations further demonstrates that: (i) the cytosolic N-terminus of SCAMP1 contains an ER export signal; (ii) the transmembrane domain 2 (TMD2) and TMD3 of SCAMP1 are essential for Golgi export; (iii) SCAMP1 TMD1 is essential for TGN-to-PM targeting; (iv) the predicted topology of SCAMP1 and its various mutants remain identical as demonstrated by protease protection assay. Therefore, both the cytosolic N-terminus and TMD sequences of SCAMP1 play integral roles in mediating its transport to the PM via an ER-Golgi-TGN pathway.  相似文献
