A series of 16 new chiral nonracemic polyhydroxylated piperidines was synthesized utilizing several chiral beta-amino-alcohols. They act as a nitrogen source, chirality inducer and iminium stabilizer, in the desymmetrization of meso-trihydroxylated glutaraldehyde. The biological activity of these compounds towards several glycosidases (alpha-D-glucosidase, alpha-D-mannosidase, alpha-L-fucosidase) has been evaluated.  相似文献

Summary. The effect of different taurine doses (0.050, 0.125, 0.250, 0.500 and 1.000g/kg) administered intraperitoneally to Wistar rats was studied in both the plasma and the hippocampal microdialysate content.The samples were analyzed by reverse phased HPLC for the microdialysate samples and by HPLC with ion-exchange post-column derivatization (ninhydrin) for the plasma samples.In both plasma and microdialysate, we observed a dose dependent increase of taurine concentration. The AUC curves obtained from both microdialysate and plasma samples showed that the increase of taurine concentrations were linear. The mean ratio between AUCs microdialysate and plasma was 1.63±0.21 showing thus an unbalance between plasma and brain taurine content; a mechanism which enhance taurine transfer from the plasma to the brain was assumed.  相似文献

A statistical analysis of the nucleotide sequence variability in 14 published hepatitis B virus (HBV) genomes was carried out using parametric and nonparametric methods. A parametric statistical model revealed that the different regions of the genome differed significantly in their variability. The conclusion was supported by a nonparametric kernel-density model of the HBV genome. Genes S, C, and P, region X, the precore region, and the pre-S2/pre-S1 regions were ranked in order of increasing variability. In many instances, conserved regions of the genome identified with sequences of known function in HBV biology. However, other characterized regions (such as pre-S) showed much variability despite the involvement of their encoded peptides in specific functions. Point mutations that may result in the formation of stop codons and amino acid changes may affect the clinical picture of HBV infection and may be reflected in atypical serological patterns.   相似文献

The stoichiometric reaction between d-TpGpGpCpCpA (d(T-G-G-C-C-A)) and cis-[Pt(NH3)2(H2O)2](NO3)2 (8.4 × 10?6 to 1.3 × 10?4M in water at pH 5.5–6) gives a single complex. High pressure gel permeation chromatography and pH-dependent 1H NMR analyses of the nonexchangeable base protons, show that it is a platinum chelate with the cis-PtII(NH3)2 moiety bound to the two N7 atoms of the adjacent guanines. A 3 × 10?3M reaction gives the same platinum chelate, via the formation of intermediate complexes, together with unsoluble adducts.  相似文献

The bonding of the bound-phenolic acids present in three lignin preparations isolated from wheat straw where determined. p-Coumaric acid was mainly ester-linked whereas 35–75% of the recovered ferulic acid was ether-linked to milled straw lignin or enzyme lignin. Ferulic acid ethers accounted for 1.1% dry wt of alkali extracted lignin and might explain the high solubility of Gramineae lignins in soda. Isolated lignins were associated to hemicelluloses, principally arabinoglucuronoxylans. The possible existence of ferulic acid cross-links between lignin and arabinoglucuronoxylans is discussed.  相似文献

The rat Chromosome (Chr) 2 harbors several genes controlling tumor growth or development, blood pressure, and non-insulin-dependent diabetes mellitus. We report that the region (2q1) containing the mammary susceptibility cancer gene Mcs1 also harbors the genes encoding cyclin B1, interleukin 6 signal transducer (gp130), and proprotein convertase 1. We also generated 13 new anonymous microsatellite markers from Chr 2-sorted DNA. These markers, as well as a microsatellite marker in the cyclin B1 gene, were genetically mapped in combination with known markers. In aqueous solution DNA intercalating ellipticine derivatives aggregate in n-mers. The self-association constants K are higher than those of 2-nicthoxy-6-chloro-9-[3-diniethylaminopropyl-amino]-acridine and ethidium bromide. They are of the same order as that of actinomycin D but inferior to that of acridine orange. The increase of the 9-hydroxy-ellipticine constant by addition of sodium chloride shows the importance of anion participation in the mechanism of stacking in accordance with the high energy of self-association. In the stacked n-mets the ellipticine rings aie inverted. The geometry shows the importance of the orientation of the quadiupole axis in the intermotecular association of the intercalating drugs.  相似文献

Background: We hypothesized that due to the absence of a dietary source of omega-3 fatty acids, the essential fatty acid (EFA) deficiency model leads to an overestimate of linoleic acid (LA) requirements. Methods: over 7 wk, young rats consumed an EFA diet containing either 0 en% linoleate (0LA) and 0 en% α-linolenate (0LNA) or a diet containing 0.5 en% LNA plus one of seven levels of added LA (0.12–4.0 en%; n=6/group).Results: Rats consuming the 0LA–0LNA diet had the lowest final body weight, 34–68% lower LA and arachidonate in plasma and liver, 87% lower LA in epididymal fat, and an 8–20 fold higher eicosatrienoate in plasma, liver and muscle lipids. 0.5LNA completely prevented the lower growth and partly prevented the rise in eicosatrienoate seen in the 0LA–0LNA group.Conclusion: Providing dietary LNA at 0.5 en% reduces the rat's physiological requirement for LA by an estimated factor of at least four (0.5 en% instead of 2 en%). Since LA requirements in humans are also based on the same flawed model of EFA deficiency, it is plausible that they too have been overestimated and should therefore be reinvestigated.  相似文献

A 2D NMR NOESY spectrum of integral CaM in water(148 residues) reveals a series of downfield-shifted crosspeaks stemming from the NH protons of the Ca2(+)-binding loops III and IV. Their attribution, with the help of already assigned proton resonances of isolated tryptic fragments, was complemented by means of energy-minimizations on the Ca2+ complexes of loops III and IV. From these calculations, a set of two alternative, related conformations was obtained for each loop. The first type of conformation provides a coordination pattern for Ca2+ that is similar to that found in loop EF of parvalbumin. The computed interproton distances in both loops are fully compatible with the inferences from the sets of NOESY cross-peaks. Evidence is also provided for interloop interactions.  相似文献
