A further case of ocular parastrongyliasis has been seen in a patient from Sri Lanka. As it is a juvenile female worm it could not be identified to the species, but it is probably Parastrongylus (= Angiostrongylus) cantonensis which is the commonly reported species in the country. This is the third authentic case of such an infection in Sri Lanka in recent years.  相似文献

Ribonucleotides are frequently incorporated into DNA during replication. They are recognized and processed by several cellular enzymes, and their continued presence in the yeast nuclear genome results in replicative stress and genome instability. Thus, it is important to understand the effects of isolated ribonucleotide incorporation on DNA structure. With this goal in mind, we describe the nuclear magnetic resonance structure of the self-complementary Dickerson dodecamer sequence [d(CGC)rGd(AATTCGCG)](2) containing two symmetrically positioned riboguanosines. The absence of an observable H(1)-H(2) scalar coupling interaction indicates a C3'-endo conformation for the ribose. Longer-range structural perturbations resulting from the presence of the ribonucleotide are limited to the adjacent and transhelical nucleotides, while the global B-form DNA structure is maintained. Because crystallographic studies have indicated that isolated ribonucleotides promote global B → A transitions, we also performed molecular modeling analyses to evaluate the structural consequences of higher ribonucleotide substitution levels. Increasing the ribonucleotide content increased the minor groove width toward values more similar to that of A-DNA, but even 50% ribonucleotide substitution did not fully convert the B-DNA to A-DNA. Comparing our structure with the structure of an RNase H2-bound DNA supports the conclusion that, as with other DNA-protein complexes, the DNA conformation is strongly influenced by the interaction with the protein.  相似文献

Background and Aims

Studies on the effects of sub- and/or supraoptimal temperatures on growth and phosphorus (P) nutrition of perennial herbaceous species at growth-limiting P availability are few, and the impacts of temperature on rhizosphere carboxylate dynamics are not known for any species.

Methods

The effect of three day/night temperature regimes (low, 20/13 °C; medium, 27/20 °C; and high, 32/25 °C) on growth and P nutrition of Cullen cinereum, Kennedia nigricans and Lotus australis was determined.

Key Results

The highest temperature was optimal for growth of C. cinereum, while the lowest temperature was optimal for K. nigricans and L. australis. At optimum temperatures, the relative growth rate (RGR), root length, root length per leaf area, total P content, P productivity and water-use efficiency were higher for all species, and rhizosphere carboxylate content was higher for K. nigricans and L. australis. Cullen cinereum, with a slower RGR, had long (higher root length per leaf area) and thin roots to enhance P uptake by exploring a greater volume of soil at its optimum temperature, while K. nigricans and L. australis, with faster RGRs, had only long roots (higher root length per leaf area) as a morphological adaptation, but had a higher content of carboxylates in their rhizospheres at the optimum temperature. Irrespective of the species, the amount of P taken up by a plant was mainly determined by root length, rather than by P uptake rate per unit root surface area. Phosphorus productivity was correlated with RGR and plant biomass.

Conclusions

All three species exhibited adaptive shoot and root traits to enhance growth at their optimum temperatures at growth-limiting P supply. The species with a slower RGR (i.e. C. cinereum) showed only morphological root adaptations, while K. nigricans and L. australis, with faster RGRs, had both morphological and physiological (i.e. root carboxylate dynamics) root adaptations.  相似文献

Heparan sulfate (HS) is an abundant polysaccharide in the animal kingdom with essential physiological functions. HS is composed of sulfated saccharides that are biosynthesized through a complex pathway involving multiple enzymes. In vivo regulation of this process remains unclear. HS 2-O-sulfotransferase (2OST) is a key enzyme in this pathway. Here, we report the crystal structure of the ternary complex of 2OST, 3′-phosphoadenosine 5′-phosphate, and a heptasaccharide substrate. Utilizing site-directed mutagenesis and specific oligosaccharide substrate sequences, we probed the molecular basis of specificity and 2OST position in the ordered HS biosynthesis pathway. These studies revealed that Arg-80, Lys-350, and Arg-190 of 2OST interact with the N-sulfo groups near the modification site, consistent with the dependence of 2OST on N-sulfation. In contrast, 6-O-sulfo groups on HS are likely excluded by steric and electrostatic repulsion within the active site supporting the hypothesis that 2-O-sulfation occurs prior to 6-O-sulfation. Our results provide the structural evidence for understanding the sequence of enzymatic events in this pathway.  相似文献

Background and aims

Australian herbaceous native species have evolved in phosphorus (P) impoverished soils. Our objective was to explore shoot and root adaptations of two of these species with potential to be developed as pasture plants, at low, moderate and high P supply after 4 and 7?weeks of growth.

Methods

A glasshouse experiment examined the effect of 5, 20 and 80?mg?P?kg?1 air-dry soil on growth, rhizosphere carboxylate content, and mineral nutrition of two Australian native perennials, Kennedia nigricans (Fabaceae) and Ptilotus polystachyus (Amaranthaceae), and the exotic Medicago sativa (Fabaceae).

Key results

Leaf P concentrations at P80 were 6, 14 and 52?mg?P?g?1 leaf dry weight for M. sativa, K. nigricans and P. polystachyus, respectively. As soil P concentration increased, rhizosphere carboxylate content decreased for M. sativa, increased and then decreased for K. nigricans and was unchanged for P. polystachyus. For all species, the contribution of malic acid declined at the second harvest. For all species and P treatments, the amount of rhizosphere carboxylates per unit root length decreased as root length of a plant increased. Plant P content was determined more by P uptake rate per unit root length and time than by root length. Uptake of Mo for all species, and uptake of K, Mg and Mn for P. polystachyus, increased with soil P concentration. Uptake of Fe and S was higher when the content of carboxylates in the rhizosphere was higher.

Conclusion

Root physiological adaptations (i.e. rhizosphere carboxylate content and P-uptake rate) are more important than morphological adaptations (i.e. root length and diameter) to enhance the uptake of P and cations.  相似文献

In an attempt to understand the mechanism of calcium accumulation in myopathies, changes in the major calcium-binding protein, calmodulin, was studied in genetically dystrophic chickens. Measurements by radioimmunoassay revealed an increase in the calmodulin concentration of dystrophic chicken muscles. Poly A-containing RNA(s) of fast and slow muscles from the normal and dystrophic chicks were hybridized with [32P]-labeled calmodulin cDNA probe by the dot-hybridization technique. Densitometric scan of the autoradiogram showed that the calmodulin mRNA levels of dystrophic fast muscles (pectoralis and posterior latissimus dorsi) were approximately two-fold higher than those of the corresponding normal muscles. No significant change in calmodulin and calmodulin messenger RNA of slow muscle (ALD) was found in dystrophic chickens. Our results suggest that increased calcium flux within the dystrophic muscle may be modulated by calmodulin.  相似文献

Two years of measurements are explored with a view to formulating an ecosystem model for Myall Lake. Stable physical characteristics, low catchment loads, low light attenuation, flat-bottomed hypsometry, and soft gyttja substrate allow stable charophyte biomass throughout the year in Myall Lake. Chara fibrosa dominates the total biomass and is abundant over the depth range 0.5–4 m. Nitella hyalina is found over the same depth range but has diminished biomass at depths greater than 2 m due to increased incidence of zero-biomass samples at depths greater than 1.5 m. Upper bounds for biomass densities were estimated for charophytes. Najas marina has great seasonal variability and meadows can have very high biomass in waters 1.5–2.7 m deep. High biomass of Najas marina is associated with low biomass of Nitella hyalina and may be a factor determining how the biomass of Nitella hyalina is distributed with respect to depth. Patchiness of Najas marina is particularly high. Temperature and light can support two growing seasons for Najas marina but mechanical disturbance is often high in spring and high biomass was only observed in late autumn during the present study. Less than 5% of the present-day production of submerged macrophytes would have been required to produce organic material of the gyttja over a 1,000-year period. The spatial distributions of gyttja and the dominant macrophytes are consistent with wind patterns. Down-lake limits on the distributions of charophytes and Najas marina are related to a gradient in the coefficient of light attenuation that is, in turn, related to proximity to the bulk of the catchment load.  相似文献
