Summary Histological examination of subcutaneous implants of a C3H mammary carcinoma regressing after a period of growth in syngeneic female recipients showed that regression was concomitant with a local cellular reaction that covered the tumor without incursion by effector cells into the tumor. Regressing tumors and tumors in arrested growth appeared to be avascular. There was no evident tendency for responding host cells to make contact with tumor cells, and where close contact between lymphocytes and tumor cells existed at the periphery of the tumor, there were no signs of direct toxic effect on the tumor cells. When tumor pieces that included significant numbers of plasma cells were implanted into normal mice, the local cellular reaction was unusually rapid and abundant. This type of reaction did not develop against similar tumor pieces implanted into presensitized mice.  相似文献

In the present study, nonionic surfactant vesicles (niosomes) formulated with Span 20, cholesterol, and novel synthesized spermine-based cationic lipids with four hydrocarbon tails in a molar ratio of 2.5:2.5:1 were investigated as a gene carrier. The effects of the structure of the cationic lipids, such as differences in the acyl chain length (C14, C16, and C18) of the hydrophobic tails, as well as the weight ratio of niosomes to DNA on transfection efficiency and cell viability were evaluated in a human cervical carcinoma cell line (HeLa cells) using pDNA encoding green fluorescent protein (pEGFP-C2). The niosomes were characterized both in terms of morphology and of size and charge measurement. The formation of complexes between niosomes and DNA was verified with a gel retardation assay. The transfection efficiency of these cationic niosomes was in the following order: spermine-C18 > spermine-C16 > spermine-C14. The highest transfection efficiency was obtained for transfection with spermine-C18 niosomes at a weight ratio of 10. Additionally, no serum effect on transfection efficiency was observed. The results from a cytotoxicity and hemolytic study showed that the cationic niosomes were safe in vitro. In addition, the cationic niosomes showed good physical stability for at least 1 month at 4°C. Therefore, the cationic niosomes offer an excellent prospect as an alternative gene carrier.  相似文献

The administration of 80 μg of corticosterone/ml of drinking solution to adrenalectomized (ADX) rats resulted in a 24 hour serum corticosterone pattern similar to that of intact animals except that the magnitude of the afternoon-nocturnal surge was one third. Basal plasma prolactin levels and the estrogen-induced afternoon prolactin surge were similar for intact and for ADX animals receiving corticosterone in the drinking solution. Adrenalectomized animals receiving 0.9% NaCl to drink, however, had an afternoon prolactin surge that was significantly lower than that of intact animals while basal levels were similar.  相似文献

Pseudomonas aeruginosa is an opportunistic human pathogen that can cause serious infection in those with deficient or impaired phagocytes. We have developed the optically transparent and genetically tractable zebrafish embryo as a model for systemic P. aeruginosa infection. Despite lacking adaptive immunity at this developmental stage, zebrafish embryos were highly resistant to P. aeruginosa infection, but as in humans, phagocyte depletion dramatically increased their susceptibility. The virulence of an attenuated P. aeruginosa strain lacking a functional Type III secretion system was restored upon phagocyte depletion, suggesting that this system influences virulence through its effects on phagocytes. Intravital imaging revealed bacterial interactions with multiple blood cell types. Neutrophils and macrophages rapidly phagocytosed and killed P. aeruginosa , suggesting that both cell types play a role in protection against infection. Intravascular aggregation of erythrocytes and other blood cells with resultant circulatory blockage was observed immediately upon infection, which may be relevant to the pathogenesis of thrombotic complications of human P. aeruginosa infections. The real-time visualization capabilities and genetic tractability of the zebrafish infection model should enable elucidation of molecular and cellular details of P. aeruginosa pathogenesis in conditions associated with neutropenia or impaired phagocyte function.  相似文献

Ammonia volatilization from applied nitrogen in alkali soils   总被引:1,自引:0,他引:1  
Incubation studies in a highly alkali soil showed ammonia volatilization losses from applied nitrogen to be largely governed by pH/alkalinity of the system. Submergence of the soil decreased the pH value resulting in lower losses. The anion of ammonium did not influence the losses. Ammonia volatilization obeyed first order kinetics. The losses were considerably reduced by deep placement of urea but were unchanged with variation in temperature from 20° to 40°C. Urea or ammonium sulphate lost similar amounts of nitrogen. Losses from green manure were very low. The results are discussed for their implication in nitrogen fertilizer efficiency and management in alkali soils.  相似文献

Melanoma is the most aggressive skin cancer. Its aggressiveness is most commonly attributed to ERK pathway mutations leading to constitutive signaling. Though initial tumor regression results from targeting this pathway, resistance often emerges. Interestingly, interrogation of the NCI-60 database indicates high growth hormone receptor (GHR) expression in melanoma cell lines. To further characterize melanoma, we tested responsiveness to human growth hormone (GH). GH treatment resulted in GHR signaling and increased invasion and migration, which was inhibited by a GHR monoclonal antibody (mAb) antagonist in WM35, SK-MEL 5, SK-MEL 28 and SK-MEL 119 cell lines. We also detected GH in the conditioned medium (CM) of human melanoma cell lines. GHR, JAK2 and STAT5 were basally phosphorylated in these cell lines, consistent with autocrine/paracrine GH production. Together, our results suggest that melanomas are enriched in GHR and produce GH that acts in an autocrine/paracrine manner. We suggest that GHR may constitute a therapeutic target in melanoma.  相似文献
