The structure of the cloned fragment of wheat (Triticum aestivum L.) TADHN gene encoding dehydrin-like protein was examined. A comparative analysis of nucleotide and deduced amino acid sequences revealed a high homology of this fragment with sequences of the barley dhn8 gene and wheat wcor gene family. In deduced amino acid sequence of the TADHN fragment, a 15-residue region EKKGFLEKIKEKLPG was found, which corresponded to a highly conserved K-segment of dehydrins. Wheat seedling treatment with 3.7 μM ABA and 0.4 μM 24-epibrassinolide exerted similar stimulatory effects on expression of the TADHN gene, which indicates the involvement of dehydrins in the protective action of these phytohormones in wheat plants.  相似文献

Abscisic acid (ABA) and lectin content was immunoassayed in wheat cell cultures affected by temperature stress. The elevated temperature (40°C) resulted in a 7-fold increase in the level of ABA and a 10-fold increase in that of lectin. The increase in the lectin content in cells was preceded by ABA accumulation. It is suggested that this ABA increase induces the synthesis of lectin, which in addition to stress proteins, play an important role in controlling mechanisms of plant adaptation to unfavourable environments.Abbreviations ABA abscisic acid - WGA wheat germ agglutinin  相似文献

Current data on the diversity of plant lectins and their functional importance for plants, caused primarily by their capacity to link carbohydrate ligands specifically and convertibly, are reviewed. For instance, the role of plant lectins in the recognition of alien organisms and in the adaptation of plants to various stress-induced effects is discussed. In addition to centres of specific affinity to carbohydrates, plant lectins are characterized by the presence of sites responsible for hydrophobic interactions with non-carbohydrate molecules. These sites link to plant hormones, proteins, and other metabolites, thus participating in the regulation of metabolic processes controlling growth, development, and differentiation in plants. The structure and biological properties of ribosome-inactivating proteins having and not having lectin activity are discussed, as well as their role in plant protection from pests and pathogens. Current data on the assumed functions of the independent groups of plant lectins with specific endogenic role are given. These include chitin-specific lectins synthesized in phloem, which are capable of forming protein-protein and RNA-protein complexes and translocating via vessels, which thus play their specific intra- or intercellular interactions, processes of growth, development, and protection of plants. Other groups of plant lectins, induced by jasmonate, such as Nictaba (Nicotiana tabaccum agglutinin), and cereal lectins related to jacalin, which are localised in the cytoplasm and nucleus, probably play regulatory role in the formation of stress response in plants. The structure and currently discussed functions of wheat germ agglutinin, a typical representative of cereal lectins, are analysed in detail.  相似文献

The study of the electronograms of B. pertussis strains isolated in the foci of pertussis revealed the existence of the morphological variants of these cells, differing in the character of the cell wall, the state of the cytoplasm, the presence of amorphous inclusions of medium electron-optical density. The morphological variants did not correlate with the character of B. pertussis colonies isolated from blood-charcoal agar. The ultrastructure of the cells belonging to the second morphological variant was similar to that of the cells from the museum strain, altered by tetracycline treatment in the course of the experiment.  相似文献

Changes in the cell surface hydrophobicity (CSH) of probiotic bacteria Lactobacillus acidophilus La5 and Bifidobacterium lactis Bb12 and the survival of these cells were examined in response to varied cultivation conditions and adverse environmental conditions. An inverse linear relationship (P < 0.01) was detected between the CSH of intact L. acidophilus La5 and B. lactis Bb12 and survival of cells subjected to subsequent freezing/thawing, long-term storage or exposure to mineral and bile acids. The observed relationships were supported by significant correlations between the CSH and changes in composition of the cell envelopes (proteins, lipids and carbohydrates) of L. acidophilus La5 and B. lactis Bb12 examined using FT-IR spectroscopy and conventional biochemical analysis methods. The results also suggest that the estimates of hydrophobicity, being a generalized characteristic of cell surfaces, are important parameters to predict the ability of intact probiotic bacteria to endure extreme environments and therefore should be monitored during cultivation. A defined balance of cell components, which can be characterized by the reduced CSH values, apparently helps to ensure the resistance, improved viability and hence the overall probiotic properties of bacteria.  相似文献
