In the field of therapeutic protein production, process intensification strategies entailing higher starting cell seeding densities, can potentially increase culture productivity, lower cost of goods and improve facility utilization. However, increased cell densities often trigger apoptotic cell death at the end of the cell culture process and thus reduce total viable cell count. Apoptosis-resistant Chinese hamster ovary cell lines may offer the possibility to diminish this undesired outcome of the intensified production process. In this study, we have generated and tested Bax/Bak double-knock-out (DKO) apoptosis resistant hosts to express standard and bispecific antibodies, as well as complex molecules in intensified production processes both as pools and single cell clones, and at different scales. In all cases, therapeutic proteins expressed from clones or pools generated from the Bax/Bak DKO hosts showed not only better viability but also enabled extended productivity in the later stages of the 14-day intensified production process. The product qualities of the produced molecules were comparable between Bax/Bak DKO and wild type cells. Overall, we showed that Bax/Bak DKO apoptosis-resistant host cell lines significantly improve viability and volumetric productivity of the intensified production cultures without altering product qualities.  相似文献

Light miscroscopy of the retina in the Atlantic stingray, Dasyatis sabina, reveal two distinct photoreceptor types. Histological criteria establish the two cell types as morphological rods and cones. The retina was studied through a sequential series of vertical sections in a protocol designed to evaluate the entire retina. By performing differential counts in multiple regions and expressing cones as a percent of the total photoreceptors in a domain, it was possible to determine relative cone density and distribution. Cone distribution varies throughout the retina in two patterns. First, relative cone density gradually decreases peripherally. Second, a cone-rich band occurs along the horizontal axis of the eye, extending from ora serrata to ora serrata. This structure appears to be a visual streak. Both distribution patterns are statistically significant and are consistent among animals regardless of age. © 1994 Wiley-Liss, Inc.  相似文献

Freeze branding using liquid nitrogen as a coolant was carried out on salmon parr and a sequential histopathological study was carried out for 16 weeks. The initial darkening of the brand area was due to destruction of melanophore control but the later, more diffuse, pattern was related to the invasion of the area of the stratum spongiosum and hypodermis by melanin containing cells such as are commonly found in healing teleost wounds. The traumatic damage was completely resolved well before the end of the experiment, suggesting that freeze branding is a particularly suitable short term batch marking method.  相似文献

Over the past few years, association analysis has become the primary tool for finding genes that underlie complex traits. Both population-based and family-based designs are commonly used designs in genetic association studies. Recent technological advances in exome and whole genome sequencing afford the next generation of sequence-based association studies.
