LPS、TNFα刺激PBL诱导TRAIL表达的初步研究

近来的实验显示从 EST来源的 TRAIL在体外能特异性地诱导肿瘤细胞凋亡。我们选择人PBL作为人 TRAIL基因的来源 ,抽提细胞在 LPS和 rh- TNFα刺激 2、1 0、1 8h的总 RNA,通过RT- PCR观察 TRAIL基因转录 m RNA的水平 ,然后用 PCR扩增 TRAIL基因 ,并用免疫印迹法分析 TRAIL m RNA翻译蛋白的水平。结果发现 :在 LPS和 rh- TNFα的刺激下可以从 1 0 6 ～ 1 0 7个人 PBL中扩增出 TRAIL基因。提示 LPS和 rh- TNFα刺激细胞并进一步诱导细胞凋亡可能与诱导表达 TRAIL有关。本实验得到了 TRAIL基因 ,为重组表达 TRAIL打下了基础。

The design and synthesis of novel, phosphonate-containing transient receptor potential melastatin 8 (TRPM8) antagonists

A series of benzothiophene-based phosphonates was synthesized and many analogs within the series were shown to be potent antagonists of the TRPM8 channel. The compounds were obtained as a racemic mixture in 5 synthetic steps, and were tested for TRPM8 antagonist activity in a recombinant, canine TRPM8-expressing cell line using a fluorometric imaging plate reader (FLIPR) assay. Structure-activity relationships were developed initially by modification of the core structure and subsequently by variation of the aromatic substituents and the phosphonate ester. Compound 9l was administered intraperitoneally to rats and demonstrated engagement of the TRPM8 target in both prevention and reversal-modes in an icilin-induced 'wet-dog' shake model.     

新疆醉马草内生菌群落结构

【目的】揭示醉马草不同组织内生细菌和内生真菌种群组成和分布。【方法】采用液氮研磨法分别提取醉马草种子、叶、茎、根组织总DNA,采用通用引物扩增内生细菌16S rDNA和真菌ITS,通过限制性内切酶HhaⅠ和RsaⅠ对16S rDNA PCR产物酶切,HhaeⅢ和HinfⅠ对真菌rDNA ITS PCR产物酶切得到不同的TRFs片段。TRFs经T-RFLP分析程序结合基因文库比对后,分析醉马草不同组织中内生细菌和内生真菌的群落组成及内生菌群落相似性。【结果】研究表明醉马草根部内生细菌多样性最高,而种子内生真菌多样性最为丰富。醉马草各组织内生细菌优势菌属均为Bacillus(29%以上),种子、叶、茎、根内生真菌优势菌属分别为Mycosphaerella(6.5%),Teratosphaeria(4.5%),Fragum(1.1%),Sebacina(11.3%)。聚类分析表明茎和叶内生细菌群落结构相似,种子和其他组织内生细菌群落结构相似性较远,而茎和种子内生真菌群落结构相似,叶和其他组织内生真菌群落结构相似性较远。醉马草内生菌多样性丰富且存在尚未认识的新类群。

By expression of the TPS genes, significant achievements have been made on metabolic engineering to Increase terpenoid production. This review mainly summarizes the recent research progress In elucidating the ecological role of terpenoids and characterization of the enzymes Involved in the terpenold biosynthesis. Spatial and temporal regulations of terpenoids metabolism are also discussed.     

Von Hippel-Lindau disease: identification of deletion mutations by pulsed-field gel electrophoresis

Von Hippel-Lindau disease (VHL) is an inherited multisystem neoplastic disorder. We prepared a 2.5-megabase (Mb) restriction map of the region surrounding the VHL gene and identified and characterized overlapping deletions in three unrelated patients affected with VHL. The smallest nested deletion (100 kb) was located within a 510-kb NruI fragment detected by 19–63. The rearrangements detected will be useful in isolating and evaluating candidate cDNAs for the VHL gene. The detailed physical map will be useful in studying the organization and structure of genes in the VHL region.
