Abstract. The clonal sedge Carex humilis forms rings of densely aggregated ramets in a dry grassland community in Central Europe. We describe the small‐scale spatial variation, both in abiotic factors and vegetation, in relation to these rings. Compared to the surrounding vegetation the cover of plants, other than C. humilis, was significantly lower both in the central area of rings and within the rings themselves. The vegetation structure was also different. The soil was more fertile in the central area and within the ring than in the surroundings, measured both directly and by the abiotic response values of the vascular plants. We conclude that neither resource depletion nor competition from other plants were likely to be responsible for the low ramet density in the central area of C. humilis rings. Instead, we suggest that the ring form is caused either by the deposition of growth inhibiting substances or by intrinsic morphological rules. 相似文献

The development of an effective vaccine is urgently needed to fight tuberculosis (TB) which is still the leading cause of death from a single infectious agent worldwide. One of the promising vaccine candidates M72/AS01E consists of two proteins subunits PepA and PPE18 coded by Rv0125 and Rv1196. However, preliminary data indicate a high level of sequence variability among clinical Mycobacterium tuberculosis complex (MTBC) strains that might have an impact on the vaccine efficacy. To further investigate this finding, we determined ppE18 sequence variability in a well-characterized reference collection of 71 MTBC strains from 23 phylogenetic lineages representing the global MTBC diversity. In total, 100 sequence variations consisting of 96 single nucleotide polymorphisms (SNPs), three insertions and one deletion were detected resulting in 141 variable positions distributed over the entire gene. The majority of SNPs detected were non-synonymous (n = 68 vs. n = 28 synonymous). Strains from animal adapted lineages, e.g., M. bovis, showed a significant higher diversity than the human pathogens such as M. tuberculosis Haarlem. SNP patterns specific for different lineages as well as for deeper branches in the phylogeny could be identified. The results of our study demonstrate a high variability of the ppE18 gene even in the N-terminal domains that is normally highly conserved in ppe genes. As the N-terminal region interacts with TLR2 receptor inducing a protective anti-inflammatory immune response, genetic heterogeneity has a potential impact on the vaccine efficiency, however, this has to be investigated in future studies. 相似文献

Nest defence is a fundamental aspect of parental care in secondary cavity‐nesting birds, and predation or competition for nesting sites can involve different defensive behaviours. Because habitat quality determines breeding success, we were interested in whether breeding pairs of the Eurasian nuthatch, Sitta europaea, established in more favourable environment also manifest higher probability of cooperative behaviour during their nest‐site defence. To explore this relationship, we quantified behavioural displays of both parents and analysed activity budget ethogram data from simulated territorial intrusions performed in the chick‐feeding phase with one conspecific and two different heterospecific stimuli (dummies of nuthatch, starling and woodpecker). We found that paired individuals shared their roles during nest‐site defence to a considerable extent. Males had a significantly higher number of attacks on intruders than females, and females performed more threat displays and controls of the brood than males. Multinomial analysis of the cooperative behaviour suggested that pairs in a high‐quality territory had higher probability of reciprocal substitution of different roles towards a balance between attacks, threat displays and nest controls. Contrary to this, pairs in a low‐quality territory had less likely pairwise combinations of simultaneous behavioural states that are associated with effective nest‐site defence. The difference in response probability according to territory quality was, however, highly variable in view of the stimulus that was used in simulated territorial intrusion. Because individual roles and the complex behavioural repertoire of pairs altered in response to territory quality and potential nest‐site competitor or brood predator, our results suggest that the cooperative nest‐defence behaviour could be linked to the breeding success of this year‐round territorial species living in a heterogeneous forest habitat. 相似文献

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a cytokine that can trigger apoptosis in many types of human cancer cells via engagement of its two pro-apoptotic receptors TRAIL-R1 (DR4) and TRAIL-R2 (DR5). TRAIL can also activate several other signaling pathways such as activation of stress kinases, canonical NF-κB signaling and necroptosis. Though both receptors are ubiquitously expressed, their relative participation in TRAIL-induced signaling is still largely unknown. To analyze TRAIL receptor-specific signaling, we prepared Strep-tagged, trimerized variants of recombinant human TRAIL with high affinity for either DR4 or DR5 receptor. Using these receptor-specific ligands, we examined the contribution of individual pro-apoptotic receptors to TRAIL-induced signaling pathways. We found that in TRAIL-resistant colorectal HT-29 cells but not in pancreatic PANC-1 cancer cells, DISC formation and initial caspase-8 processing proceeds comparably via both DR4- and DR5-activated signaling. TRAIL-induced apoptosis, enhanced by the inhibitor of the Bcl-2 family ABT-737, or by the translation inhibitor homoharringtonine, proceeded in both cell lines predominantly via the DR5 receptor. ShRNA-mediated downregulation of DR4 or DR5 receptors in HT-29 cells also pointed to a stronger contribution of DR5 in TRAIL-induced apoptosis. In contrast to apoptosis, necroptotic signaling was activated similarly by both DR4- or DR5-specific ligands. Activation of auxiliary signaling pathways involving NF-κB or stress kinases proceeded under apoptotic conditions mainly in a DR5-dependent manner, while these signaling pathways were during necroptosis similarly activated by either of these ligands. A syntaxonomic revision of vegetation of anthropogenic metalliferous habitats (mine spoils of lead, zinc and copper mining) in the Eastern Alps (Austria, Germany, Italy and Slovenia) was made. The communities studied belong to theThlaspietea rotundifolii (Linaria alpina-Cerastium uniflorum comm.,Papaveri kerneri-Thlaspietum kerneri, Violetum dubyanae, Thlaspietum cepaeifolii, Thlaspietum rotundifolii, Scrophulario juratensis-Erysimetum sylvestris, Minuartia gerardii-Silene glareosa comm.,Epipactido atrorubentis-Silenetum glareosae andSileno alpestris-Moehringietum muscosae) and to theAsplenietea trichomanis (Sileno rupestris-Asplenietum). There is neither floristic support nor syntaxonomic justification for the concepts of theGalio anisophylli-Minuartion vernae, theVioletalia calaminariae and theVioletea calaminariae in the Eastern Alps. A revision was undertaken of the high-rank syntaxa of the vegetation occurring in rock fissures, ledges and screes of mainland Greece and Crete. All published phytosociological relevés available were collected and subjected to numerical classification and ordination. Four orders (Androsacetalia vandelii, Onosmetalia frutescentis, Potentilletalia speciosae andPetromaruletalia pinnatae) comprising 8 alliances (one of them new) were distinguished within theAsplenietea trichomanis (rock fissures, clefts, and ledges). The scree vegetation was classified partly within theThlaspietea rotundifolii (Drypidetalia spinosae with 2 alliances) and partly within theDaphno-Festucetea (Saturejo-Scutellarietalia—a new order, with 2 alliances). Ordination revealed clear differences in floristic composition of the alliances distinguished. The studied chasmophytic vegetation of Greece contains a very high proportion of endemics which serve as diagnostic species of the syntaxa. Further, an analysis of chorological species spectra suggested that the chorological homogeneity of a syntaxon should be considered an important diagnostic feature at high-syntaxon levels. 相似文献
