Abstract. Aim: Patterns of plant functional traits related to clonality (clonal growth modes; CGM) in plant communities were studied and hypotheses on the importance of the selected traits in plant communities supported by soils differing in moisture and nutrient status were tested. Material and Methods: Selected plant functional traits, such as the position of the mother‐daughter plants connections, length of spacers, frequency of multiplication, persistency of ramets connections, presence of storage organs and bud protection were studied in two contrasting plant communities (xeric and mesic abandoned pastures) typical of central Apennines, Italy. Results and Discussion: Clonality was shown to be of great importance in both mesic and xeric grasslands. The major differences between the two communities were due to the dominant CGMs: turf graminoids (having effective protection of growth meristems in dense tussocks) dominated xeric grasslands, while rhizomatous graminoids (typical of competitive resource‐rich environments) dominated mesic grasslands. Below‐ground CGOs (clonal growth organs), shorter spacers, higher multiplication potential, permanent ramet connection, large bud bank and increased importance of bud protection were found to be of importance in water stressed xeric grassland. Contrary to our expectations, the mesic (less stressed) grasslands have the higher number of clonal plants possessing storage organs.  相似文献

In Salicornia, morphology does not provide reliable diagnostic characters supporting the true extent of evolutionary divergence in the genus, and species concepts have been challenged by molecular analyses. Here, we report the results of an analysis of 91 accessions of the S. meyeriana complex from South Africa and Namibia using the measurements of 38 morphological traits and external transcribed spacer (ETS) sequence data. Morphological data were analysed using discriminant analysis, principal coordinate analysis and nonmetric multidimensional scaling. Phylogenetic divergence was compared with the geographical and ecological diversity of the sampled populations. Tree topology corresponds to geography and ecology, but not to morphology. Most clades have distinct distribution areas and ecological profiles related to tidal, supratidal or inland saline habitats. Salicornia probably diversified in habitats that have experienced regular fragmentation by marine transgression/regression cycles during the Pleistocene. We suggest that this radiation produced young, but genetically, geographically and ecologically well‐defined lineages. The lack of morphological signal reveals the existence of cryptic species in Salicornia and demonstrates the necessity of using molecular data to define taxa in this genus. We propose the recognition of two subspecies in the S. meyeriana complex: S. meyeriana subsp. meyeriana and S. meyeriana subsp. knysnaensis. © 2013 The Linnean Society of London, Botanical Journal of the Linnean Society, 2013, 172 , 175–186.  相似文献

Refugia are key environments in biogeography and conservation. Because of their unique eco‐evolutionary formation and functioning, they should display distinct functional trait signatures. However, comparative trait‐based studies of plants in refugia and non‐refugia are lacking. Here, we provide a comparison between resource‐rich (putative microrefugia for species preferring mesic habitats under increasing aridity) and resource‐impoverished woodlands (non‐refugia) around two granite outcrops in south‐western Australia. We measured and compared six functional traits (bark thickness, foliar δ13C, foliar C:N, leaf dry matter content, plant height, specific leaf area) in four woody species. We performed multiple‐trait, multiple‐species and single‐trait, within‐species analyses to test whether plants in resource‐rich habitats were functionally distinct and more diverse than those in the surrounding resource‐impoverished woodlands. We found that species in resource‐rich woodlands occupied larger and distinct multiple‐trait functional spaces and showed distinct single‐trait values (for specific leaf area and bark thickness). This suggests that plants in resource‐rich woodlands can deploy unique and more diverse ecological strategies, potentially making these putative microrefugia more resilient to environmental changes. These findings suggest that species in microrefugia may be characterised by unique functional signatures, illustrating the utility of comparative trait‐based approaches to improve understanding of the functioning of refugia.  相似文献

Human topoisomerase I (Top1) relaxes supercoiled DNA during cell division. Camptothecin stabilizes Top1/dsDNA covalent complexes which ultimately results in cell death, and this makes Top1 an anti-cancer target. There are two current models for how camptothecin and derivatives bind to Top1/dsDNA covalent complexes (Staker, et al., 2002, Proc Natl Acad Sci USA 99: 15387-15392; and Laco, et al., 2004, Bioorg Med Chem 12: 5225-5235). The interaction energies between bound camptothecin, and derivatives, and Top1/dsDNA in the two models were calculated. The published structure-activity-relationships for camptothecin and derivatives correlated with the interaction energies for camptothecin and derivatives in the Laco et al. model, however, this was not the case for several camptothecin derivatives in the Stacker et al. model. By defining the binding orientation of camptothecin and derivatives in the Top1/dsDNA active-site these results allow for the rational design of potentially more efficacious camptothecin derivatives.  相似文献

Sarcocornia decussata, S. freitagii and S. tegetaria from South Africa are described as new taxa. Sarcocornia decussata and S. freitagii are narrow endemics of the West Coast of South Africa where they are found in inland saline habitats (quartz patches, salt pans and saline alluvia) while S. tegetaria is an endemic of southern African coasts (spanning Namibia, South Africa and Mozambique) where it is confined to low-lying intertidal habitats of estuaries.  相似文献

Question: How can vegetation gradients be described in riparian zones located in a species‐rich mountain range and how do these gradients explain the variation found in the vegetation? Location: Hottentots Holland Mountains, Western Cape, South Africa. Methods: Three gradients (geographic, longitudinal, lateral) were defined to describe the complex vegetation patterns found here. The gradients are related as follows: (1) the geographic gradient: related to the climatic and topographical changes across the entire mountain range; (2) the longitudinal gradient related to the changes along the different river reaches; (3) the lateral gradient related to the processes along the profile of the riverbed. These three gradients operated on three different hierarchical levels. Partial Canonical Correspondence Analysis (pCCA) was used to determine the amount of variation that is explained on each of the hierarchical levels. Results: The geographical gradient explained the highest fraction (more than 50%) of the total variation explained. This can be ascribed to the high species turnover across landscapes in the Fynbos Biome; this is most likely an outcome that is specific for this region. The second most important gradient was the lateral gradient, which reflects stream power and inundation frequencies of the river. This gradient is represented by ca. 48% of the explained variation and this gradient explains the major disturbances occurring in a riverine ecosystem. The longitudinal gradient was the least important of the gradients and shows overlap with the geographical gradient. Conclusions: In the species‐rich environment of the Fynbos Biome geographical factors do not only account for variation in zonal vegetation but also for variation within azonal vegetation, like riparian corridors.  相似文献

Dendritic cells (DC) represent a unique set of APCs that initiate immune responses through priming of naive T cells. Maturation of DC is a crucial step during Ag presentation and can be induced by triggering a broad spectrum of DC surface receptors. Although human DC express several receptors for the Fc portion of IgG which were described to play an important role in Ag internalization, little is known about the effects of IgG or immune complexes on DC maturation. In this study, we show that cross-linking of FcgammaR-type II (CD32) with immobilized IgG (imIgG) can induce maturation of human monocyte-derived DC via the NF-kappaB signaling pathway. IgG-mediated maturation was accompanied by a moderate increase of IL-10 secretion, whereas no IL-12 production was observed. Involvement of CD32 was further supported by experiments with the anti-CD32 mAb, which blocked IgG-triggered DC maturation and cytokine secretion significantly. Furthermore, DC cultivated in the presence of imIgG induced allogeneic T cell proliferation. Because this imIgG-induced maturation was considerably impaired in monocyte-derived DC from systemic lupus erythematosus patients, we suggest that DC, which matured in the presence of immune complexes, may contribute to prevention of pathological immune responses.  相似文献

Human topoisomerase I (top1) is an important target for anti-cancer drugs, which include camptothecin (CPT) and its derivatives. To elucidate top1 inhibition in vitro, we made a series of duplex DNA substrates containing a deoxyadenosine stereospecifically modified by a covalent adduct of benzo[a]pyrene (BaP) diol epoxide [Pommier, Y., et al. (2000) Proc. Natl. Acad. Sci. U.S.A. 97, 10739-10744]. The known orientation of the hydrocarbon adduct in the DNA duplex relative to the top1 cleavage site, in combination with a top1/DNA crystal structure [Redinbo, M. R., et al. (1998) Science 279, 1504-1513], was used to construct a structure-based model to explain the in vitro top1 inhibition results obtained with adducted DNA duplexes. Here we experimentally determined that the lactone form of CPT was stabilized by an irreversible top1/DNA covalent complex. We removed the BaP moiety from the DNA in the published model, and docked the lactone forms of CPT and derivatives into the top1/DNA active site cavity. The docked ligands were minimized, and interaction energy scores between the ligands and the top1/DNA complex were determined. CPT docks perpendicular to the DNA backbone, projects outward from the major groove, and makes a network of potential H-bonds with the active site DNA and top1 residues, including Arg364, Lys532, and Asn722. The results are consistent with the known structure-activity relationships of CPT and derivatives. In addition, the model proposed a novel top1/N352A "resistance" mutation for 10-OH derivatives of CPT. The in vitro biochemical characterization of the top1/N352A mutant supported the model.  相似文献

Complex processes related to biotic and abiotic forces can impose limitations to assembly and composition of plant communities. Quantifying the effects of these constraints on plant functional traits across environmental gradients, and among communities, remains challenging. We define ecological constraint (Ci) as the combined, limiting effect of biotic interactions and environmental filtering on trait expression (i.e., the mean value and range of functional traits). Here, we propose a set of novel parameters to quantify this constraint by extending the trait‐gradient analysis (TGA) methodology. The key parameter is ecological constraint, which is dimensionless and can be measured at various scales, for example, on population and community levels. It facilitates comparing the effects of ecological constraints on trait expressions across environmental gradients, as well as within and among communities. We illustrate the implementation of the proposed parameters using the bark thickness of 14 woody species along an aridity gradient on granite outcrops in southwestern Australia. We found a positive correlation between increasing environmental stress and strength of ecological constraint on bark thickness expression. Also, plants from more stressful habitats (shrublands on shallow soils and in sun‐exposed locations) displayed higher ecological constraint for bark thickness than plants in more benign habitats (woodlands on deep soils and in sheltered locations). The relative ease of calculation and dimensionless nature of Ci allow it to be readily implemented at various scales and make it widely applicable. It therefore has the potential to advance the mechanistic understanding of the ecological processes shaping trait expression. Some future applications of the new parameters could be investigating the patterns of ecological constraints (1) among communities from different regions, (2) on different traits across similar environmental gradients, and (3) for the same trait across different gradient types.  相似文献
