Gluten exclusion (protein complex present in many cereals) has been proposed as an option for the prevention of diseases other than coeliac disease. However, the effects of gluten-free diets on obesity and its mechanisms of action have not been studied. Thus, our objective was to assess whether gluten exclusion can prevent adipose tissue expansion and its consequences. C57BL/6 mice were fed a high-fat diet containing 4.5% gluten (Control) or no gluten (GF). Body weight and adiposity gains, leukocyte rolling and adhesion, macrophage infiltration and cytokine production in adipose tissue were assessed. Blood lipid profiles, glycaemia, insulin resistance and adipokines were measured. Expression of the PPAR-α and γ, lipoprotein lipase (LPL), hormone sensitive lipase (HSL), carnitine palmitoyl acyltransferase-1 (CPT-1), insulin receptor, GLUT-4 and adipokines were assessed in epidydimal fat. Gluten-free animals showed a reduction in body weight gain and adiposity, without changes in food intake or lipid excretion. These results were associated with up-regulation of PPAR-α, LPL, HSL and CPT-1, which are related to lipolysis and fatty acid oxidation. There was an improvement in glucose homeostasis and pro-inflammatory profile-related overexpression of PPAR-γ. Moreover, intravital microscopy showed a lower number of adhered cells in the adipose tissue microvasculature. The overexpression of PPAR-γ is related to the increase of adiponectin and GLUT-4. Our data support the beneficial effects of gluten-free diets in reducing adiposity gain, inflammation and insulin resistance. The data suggests that diet gluten exclusion should be tested as a new dietary approach to prevent the development of obesity and metabolic disorders.  相似文献

While many bacteria have been used as probiotics by industries, only two yeasts, Saccharomyces cerevisiae var. boulardii and Kluyveromyces fragilis (B0399), have been used for this purpose. In the present work, a total of 116 yeasts isolated from Brazilian indigenous fermented food, cocoa fermentation, and kefir were in vitro characterized for probiotic attributes. From 116 isolates, 36 were tolerant to gastrointestinal conditions evaluated by tolerance to pH 2.0, bile salts (0.3% w/v), and 37 °C temperature. From those, 15 isolates showed a similar or higher percentage (P < 0.05) of hydrophobicity, autoaggregation, and coaggregation with E. coli than the reference strain S. boulardii. All these strains showed a high percentage of adhesion to Caco-2 cells (> 63%) and antioxidant activity (ranging from 18 to 62%). Phytate hydrolysis was evaluated for these yeasts and 13 strains showed positive results, which is important for nutrient availability in plant-based foods. These results are important insights for characterization of novel probiotic yeast strains as well as to aggregate functional value to these food products.

The effects of supplemental Ca2+ on membrane integrity and lipid composition of cowpea plants submitted to salt stress (75 mM NaCl) were evaluated. The experimental design was factorial (2 × 6 + 1) corresponding to six saline treatments supplemented with CaCl2 and six saline treatments supplemented with CaSO4, both at 0.5, 1.25, 2.5, 5.0, 7.5 and 10.0 mM, plus control treatment (plants grown in half-strength Hoagland’s nutrient solution without supplemental calcium addition). Samples of leaves and root tips were analyzed for total lipid, glycolipid and fatty acid contents and membrane damage symptoms. Salt stress greatly reduced total lipid content in leaves and roots and caused great damage to membrane structures. In leaves, the glycolipid content was differently influenced by calcium treatments. Moreover, salinity increased the saturated/unsaturated fatty acid ratio in leaves and an increase in the concentration of calcium intensified this response. In roots, only saturated fatty acids were detected and their content was strongly influenced by salinity and very little by calcium treatments. Supplemental Ca2+ was unable to ameliorate the negative effects of salinity on the structural integrity and fluidity of plant membranes in cowpea.  相似文献

Brachiaria brizantha (syn. Urochloa brizantha) is an important tropical forage grass widely cultivated in Brazil. In order to optimize tissue culture conditions for B. brizantha, in vitro culture of mature seeds, basal segments and leaf segments from in vitro plants of an apomictic and a sexual genotype of B. brizantha was performed. When cultured on different media, leaf segments yielded non-embryogenic calluses which formed several roots. Friable calluses from mature seeds and basal segments explants incubated on Murashige and Skoog medium supplemented with 2,4-dichlorophenoxyacetic acid and 6-benzyladenine yielded 80% compact and nodular embryogenic structures. Calluses with such compact embryogenic structures were highly regenerable upon transfer to medium supplemented with kinetin and naphthalene acetic acid. They produced isolated somatic embryos, multiple fused scutelli or isolated scutellum with polyembryos that germinated into isolated or multiple shoots. Green and morphologically normal plants were obtained for the two genotypes. Changing the media from pH 5.8 to pH 4.0 increased the number of explants that formed calluses as well as the number of shoots per explant. When embryogenic calluses from mature seeds were successively sub-cultured for 4 months, aiming at repetitive somatic embryogenesis, all the regenerated plants were albinos. The embryogenic nature of the compact structure was confirmed by scanning electron microscopy.  相似文献

Under salinity stress, plants commonly accumulate carbohydrates for osmotic adjustment to balance the excess accumulated ions and to protect biomolecules. We selected two cowpea cultivars with contrasting response to salinity, Pitiúba (salt-tolerant) and TVu (salt-sensitive), to investigate whether the salt tolerance could be associated with changes in carbohydrate accumulation and metabolism in leaves and roots during a long-term experiment. Two salt treatments (0 and 75 mM NaCl) were applied to 10-day-old plants grown in nutrient solution for 24 days. Despite some changes in carbohydrate accumulation and carbohydrate metabolism enzymes induced by salt stress, no consistent alterations in carbohydrates could be found in leaves or roots in this study. Therefore, we suggest that tolerance to salt stress is largely unrelated to carbohydrate accumulation in cowpea.  相似文献

Despite not being a criterion for severe malaria, thrombocytopenia is one of the most common complications of both Plasmodium vivax and Plasmodium falciparum malaria. In a systematic review of the literature, platelet counts under 150,000/mm3 ranged from 24-94% in patients with acute malaria and this frequency was not different between the two major species that affected humans. Minor bleeding is mentioned in case reports of patients with P. vivax infection and may be explained by medullary compensation with the release of mega platelets in the peripheral circulation by megakaryocytes, thus maintaining a good primary haemostasis. The speculated mechanisms leading to thrombocytopenia are: coagulation disturbances, splenomegaly, bone marrow alterations, antibody-mediated platelet destruction, oxidative stress and the role of platelets as cofactors in triggering severe malaria. Data from experimental models are presented and, despite not being rare, there is no clear recommendation on the adequate management of this haematological complication. In most cases, a conservative approach is adopted and platelet counts usually revert to normal ranges a few days after efficacious antimalarial treatment. More studies are needed to specifically clarify if thrombocytopenia is the cause or consequence of the clinical disease spectrum.  相似文献

It is generally accepted that Plasmodium vivax, the most widely distributed human malaria parasite, causes mild disease and that this species does not sequester in the deep capillaries of internal organs. Recent evidence, however, has demonstrated that there is severe disease, sometimes resulting in death, exclusively associated with P. vivax and that P. vivax-infected reticulocytes are able to cytoadhere in vitro to different endothelial cells and placental cryosections. Here, we review the scarce and preliminary data on cytoadherence in P. vivax, reinforcing the importance of this phenomenon in this species and highlighting the avenues that it opens for our understanding of the pathology of this neglected human malaria parasite.  相似文献

Volatile and semi-volatile components of maté (Ilex paraguariensis St. Hil.) were analysed by headspace solid-phase microextraction (HS-SPME) coupled to gas chromatography and mass spectrometry. Five SPME fibres coated separately with 100 microm poly(dimethylsiloxane) (PDMS), 65 microm PDMS-divinylbenzene (DVB), 70 microm carbowax (CW)-DVB, 85 microm carboxen (CAR)-PMDS or 50/30 microm DVB-CAR, were tested. Seventy compounds were identified in the sample headspace, including propanal, (E)-2-pentenal, hexanal, (E)-2-hexenal, 6-methyl-5-hepten-2-one, (E,Z)-2,4-heptadienal, (E,E)-2,4-heptadienal, (E,Z)-3,5-octadien-2-one, beta-cyclocitral, 3-ethyl 4-methyl-(1H)-pyrrole-2,5-dione, alpha-ionone, geranylacetone, beta-ionone, dihydroactinidiolide and caffeine. Extraction parameters such as temperature, time and sample mass were studied and optimized. The best conditions for trapping volatile and semi-volatile compounds were obtained using a DVB-CAR fibre at 80 degrees C for 60 min with a sample mass starting from 100 mg in a vial of 4 mL.  相似文献

In the 1950s, the strategy of adding chloroquine to food salt as a prophylaxis against malaria was considered to be a successful tool. However, with the development of Plasmodium resistance in the Brazilian Amazon, this control strategy was abandoned. More than 50 years later, asexual stage resistance can be avoided by screening for antimalarial drugs that have a selective action against gametocytes, thus old prophylactic measures can be revisited. The efficacy of the old methods should be tested as complementary tools for the elimination of malaria.  相似文献
