Bacteria were recovered from loci within skeletal regions ofthe glomerate coral,Porites lobata. The origin of these bacteriais unknown, although areas of discoloration suggest invasionfrom the substratum in the region of basal attachment. Weakenedareas of internal corallum contained from 104 to 105 bacteriaper gram dry weight as determined by plate counts on a peptone-agarmedium. Some of the isolated bacteria were capable of digestingchitin in vitro. This rinding suggested that the mechanism forskeletal weakening might be bacterial breakdown of the organicmatrix. Absence of change from aragonitic to calcitic crystalsfrom a discolored region supported the contention that skeletalweakening was due to the breakdown of organic matrix ratherthan dissolution of carbonate. Results were obtained as part of a survey to determine the numberand distribution of bacteria in some coral reef environments.  相似文献

1. This paper is a synthesis of a special issue on groundwater biodiversity with a focus on obligate subterranean species, the stygobionts. The series of papers constitutes a great leap forward in assessing and understanding biodiversity patterns because of the use of large quantitative data sets obtained over a broad geographic scale. They also represent a conceptual shift, away from a purely taxonomic and phylogenetic focus to the analysis of whole groundwater assemblages.
2. The general patterns emerging for groundwater fauna are: very high levels of endemism, low local diversity relative to regional diversity, a limited number of lineages, occurrence of many relicts, and truncated food webs with very few predators.
3. β-Diversity is at least as important as α-diversity in determining total richness at different scales (aquifer, basin and region) and overall taxa richness increases across spatial scales.
4. Advances in understanding groundwater biodiversity patterns further include identification of several important factors related to geology and hydrology that determine the composition of European stygobiotic assemblages.
5. Important challenges for future research include improving sampling strategies, filling gaps in sampling coverage, intensifying research on theoretical and statistical models, and including functional and genetic diversity components in biodiversity assessments.
6. Strategies are proposed for protecting groundwater biodiversity and an argument is made to integrate biodiversity in groundwater management. Applying principles such as complementarity and flexibility for groundwater biodiversity conservation is a major step toward delineating a reserve network that maximise species representation at the European scale.  相似文献

Methods for the specific detection of Bacillus spores are needed in many situations such as the recognition of food poisoning. This study presents an experimental design in order to find the best combination of germination conditions leading to a rapid and detectable fluorescent in situ hybridization (FISH) signal from Bacillus cereus spores present in pure cultures and milk samples.
B. cereus ATCC 14579 and HER 1414 were incubated in 20 different growth media by using a combination of various germinants such as sugars, amino acids and dipicolinic acid. Also, three different germination factors were tested: incubation temperature, inoculum concentration and a heat shock treatment. Permeabilization procedure and hybridization time were optimized on the best germination condition found. B. cereus -specific FISH probes were validated under the optimized condition and in detection of spiked B. cereus spores in 1% ultra heat-treated milk samples. FISH-labeled cells were detected by using flow cytometry, and the results were confirmed by fluorescence microscopy. The optimal condition allows the detection of B. cereus spores in less than 2 h. Overall, a ninefold reduction in total time for detection was achieved when comparing with previous works. Therefore, the permeabilization and hybridization optimizations mentioned in this study are major improvements for the detection time of B. cereus spores.

PRACTICAL APPLICATIONS


By using the optimized conditions of germination/outgrowth, permeabilization and hybridization, the detection of 103 cfu/mL of Bacillus cereus spores using fluorescent in situ hybridization is possible within 2 h in milk sample.  相似文献

CO2 production in terrestrial ecosystems is generally assumed to be solely biologically driven while the role of abiotic processes has been largely overlooked. In addition to microbial decomposition, photodegradation – the direct breakdown of organic matter (OM) by solar irradiance – has been found to contribute to litter mass loss in dry ecosystems. Previous small‐scale studies have shown that litter degradation by irradiance is accompanied by emissions of CO2. However, the contribution of photodegradation to total CO2 losses at ecosystems scales is unknown. This study determined the proportion of the total CO2 losses caused by photodegradation in two ecosystems: a bare peatland in New Zealand and a seasonally dry grassland in California. The direct effect of solar irradiance on CO2 production was examined by comparing daytime CO2 fluxes measured using eddy covariance (EC) systems with simultaneous measurements made using an opaque chamber and the soil CO2 gradient technique, and with night‐time EC measurements under the same soil temperature and moisture conditions. In addition, a transparent chamber was used to directly measure CO2 fluxes from OM caused by solar irradiance. Photodegradation contributed 19% of the annual CO2 flux from the peatland and almost 60% of the dry season CO2 flux from the grassland, and up to 62% and 92% of the summer mid‐day CO2 fluxes, respectively. Our results suggest that photodegradation may be important in a wide range of ecosystems with exposed OM. Furthermore, the practice of partitioning daytime ecosystem CO2 exchange into its gross components by assuming that total daytime CO2 losses can be approximated using estimates of biological respiration alone may be in error. To obtain robust estimates of global ecosystem–atmosphere carbon transfers, the contribution of photodegradation to OM decomposition must be quantified for other ecosystems and the results incorporated into coupled carbon–climate models.  相似文献

The vegetative anatomy and morphology of 142 species of the angraecoid orchids (Angraecinae + Aerangidinae) and 18 species of Aeridinae were examined using light and scanning electron microscopy. Leafless members of Vandeae were of particular interest because of their unique growth habit. Leafy and leafless members of Angraecinae and Aerangidinae were examined and compared with specimens of Aeridinae. Vandeae were homogeneous in both leaf and root anatomy. A foliar hypodermis and fibre bundles were generally absent. Stegmata with spherical silica bodies were found associated with sclerenchyma and restricted to leaves in almost all specimens examined. Distinct inner tangential wall thickenings of the endovelamen occurred in several vandaceous genera. Exodermal proliferations and aeration units commonly occurred in both leafy and leafless Vandeae. Cladistic analyses of Angraecinae and Aerangidinae with members of Aeridinae and Polystachyinae as outgroups using 26 structural characters resulted in 20 000+ equally parsimonious trees. Dispersal ability is a key determinant of the propensity of an organism to cope with habitat fragmentation and climate change. Here we quantify queen dispersal in two common bumblebee species in an arable landscape. Dispersal was measured by taking DNA samples from workers in the spring and summer, and from queens in the following spring, at 14 sites across a landscape. The queens captured in the spring must be full sisters of workers that were foraging in the previous year. A range of sibship reconstruction methods were compared using simulated data sets including or no genotyping errors. The program Colony gave the most accurate reconstruction and was used for our analysis of queen dispersal. Comparison of queen dispersion with worker foraging distances was used to take into account an expected low level of false identification of sister pairs which might otherwise lead to overestimates of dispersal. Our data show that Bombus pascuorum and B. lapidarius queens can disperse by at least 3 and 5 km, respectively. These estimates are consistent with inferences drawn from studies of population structuring in common and rare bumblebee species, and suggest that regular gene flow over several kilometres due to queen dispersal are likely to be sufficient to maintain genetic cohesion of ubiquitous species over large spatial scales whereas rare bumblebee species appear unable to regularly disperse over distances greater than 10 km. Our results have clear implications for conservation strategies for this important pollinator group, particularly when attempting to conserve fragmented populations.  相似文献

Habitat fragmentation may strongly reduce individuals’ dispersal among resource patches and hence influence population distribution and persistence. We studied the impact of landscape heterogeneity on the dispersal of the golden‐crowned sifaka (Propithecus tattersalli), an endangered social lemur species living in a restricted and highly fragmented landscape. We combined spatial analysis and population genetics methods to describe population units and identify the environmental factors which best predict the rates and patterns of genetic differentiation within and between populations. We used non‐invasive methods to genotype 230 individuals at 13 microsatellites in all the main forest fragments of its entire distribution area. Our analyses suggest that the Manankolana River and geographical distance are the primary structuring factors, while a national road crossing the region does not seem to impede gene flow. Altogether, our results are in agreement with a limited influence of forest habitat connectivity on gene flow patterns (except for North of the species’ range), suggesting that dispersal is still possible today among most forest patches for this species. Within forest patches, we find that dispersal is mainly among neighbouring social groups, hence confirming previous behavioural observations.  相似文献

The Monogenea, which is divided into two clades, namely the Monopisthocotylea and Polyopisthocotylea, is a highly diversified group of platyhelminth parasites that infest mainly actinopterygian and chondrichthyan fishes but also, to a lesser extent, freshwater sarcopterygian hosts. Euzetrema knoepffleri Combes, 1965 (Monogenea: Iagotrematidae), which is specific to the salamander Euproctus montanus Savi, 1838 is among the rare monopisthocotylean parasites infesting tetrapod hosts. We sequenced the complete 18S rRNA gene of this parasite to infer its phylogenetic position within the Monopisthocotylea. Our results provide a new insight for coevolutionary scenarios between monopisthocotyleans and gnathostomatan hosts. Indeed, the basal position of E. knoepffleri within a subgroup of the Monopisthocotylea which comprises two clusters that both include parasites of the Actinopterygii and Chondrichthyes, suggests a very old association between the Iagotrematidae and tetrapods. Furthermore, if we take into account a recent view of Gnathostomata evolution where bony and cartilaginous fishes are regarded as a monophyletic group, it could be argued that the Iagotrematidae arose very early, during the fish–tetrapod transition, as did the Polystomatidae, the only monogenean family of the Polyopisthocotylea that infests sarcopterygian hosts.  © 2003 The Linnean Society of London, Biological Journal of the Linnean Society , 2003, 80 , 727–734.  相似文献
