Petioles of the semi-aquatic fern Regnellidium diphyllum donot show acid growth but low wall pH is a necessary conditionfor maximum rates of IAA-induced cell expansion. Measurementsof wall pH by two indirect methods indicate an unusually lowvalue, in the range pH 4 to 5. This is one to two pH units belowthat estimated for petioles of the semi-aquatic dicotyledonNymphoides peltata, a species in which IAA aand ethylene causegrowth responses very similar to those in Regnellidium but whereacid growth occurs. Having shown previously that fusicoccinenhances proton secretion in both Regnellidium and Nymphoides,we now show that although it causes a reduction in the estimatedapoplast pH to below 4·0 in Regnellidium, cell expansionis not promoted. The FC-induced reduction in pH in Nymphoidesis less and occurs more slowly, but growth is promoted significantly;when IAA and fusicoccin are present together, growth promotionis approximately additive for Nymphoides A model is proposed for Regnellidium in which equilibrium wallpH is maintained at a low value that is optimal for acid growth,the availability of acid-labile sites in the wall being thechief limitation to cell extension. We suggest that this controlmechanism may be widespread for organs without a cuticle, includingroots and the gametophytes of lower plants growing in acidicconditions. Differences in premature leaf abscission and in visible steminjury in genetic lines of poplar followed continuous fumigationswith air pollutant levels of SO2 (90–100 nl l–1)and O3 (70–80 nl l–1) either separately or together.The clones used were: Populus deltoides var. missiouriensisMarsh., P. nigra cv. ‘italicd’ L., and the hybridsP. nigra cv. ‘italica’ xP. deltoides (He-X/3) andP. nigra cv.‘italica’ x P. nigra cv. ‘Serres’(He-K/7). While most leaf abscission occurred within 20 d fromthe start of fumigation, stem lesions (intumescences), appearedonly after 72 d. Their anatomical characteristics include theformation of lysigenous aerenchyma in the lower parts of theintumescence, the sloughing of superficial cells from the injuredarea, and the development of crystalline formations on the surfaceof the lesions. P. deltoides exhibited the least morphologicalresponse to the gases. Ethylene released from fumigated leaves was determined at thesame gas concentration of SO2 (100 nl l–1), O3 (75 nll–1) and their mixture. Leaves of P. deltoides consistentlyshowed the lowest ethylene production after the gas treatments.P. ‘italica’ production was higher but was littlealtered by the treatments. The two hybrids He-X/3 and He-K/7showed the greatest increases in ethylene evolution with time.With He-K/7 exposed to the gas mixture the production of ethylenedecreased after the initial sharp rise during days 1–2,and reflected the considerable leaf damage observed after day3. The results suggest that sensitivity to air pollution, (as shownby leaf abscission and the formation of stem intumescences)can be correlated with the level of pollutant-induced ethyleneevolution from leaves. Initially high levels could induce abscission,whilst prolonged production could be responsible for intumescenceinitiation. The discussion proposes a series of events fromSO2 and/or O3 entry into the leaf and the physiological reasonsfor the clonal differences. Evidence of the source of carcass contamination of pigs at slaughter was obtained by determining presumptive coliform counts on faeces and on carcass surfaces, and comparing the O-serotypes and antibiotic sensitivity patterns of Escherichia coli from both sites. All of the 16 pig carcasses from the slaughter line of a commercial abattoir were contaminated with presumptive coliform bacilli on most sites examined; the carcasses of six out of eight pigs slaughtered at the Meat Research Institute (MRI) abattoir were also contaminated, but only small numbers of coliforms could be detected on a few of the sites. The proportion of O-serotypes of E. coli present in faeces which were also detected on carcass surfaces, indicating faecal contamination, varied between 0 and 8.6% in MRI slaughtered pigs but reached 66.6% in one group of commercially slaughtered pigs. O-serotypes found on carcass surfaces but not in the faeces of the pigs, were used as an indication of environmental contamination and this was very evident in the commercially slaughtered pigs. A high proportion of E. coli O-serotypes in the gut were resistant to antibiotics and these were also often found on the carcass surface and, since the range of O-serotypes in the pig is similar to that reported in man, the pig must be considered to be a potential reservoir of antibiotic resistant E. coli for man.  相似文献

Chloroplasts, Kinetin and Protein Synthesis   总被引:1,自引:0,他引:1  
The effect of kinetin on protein synthesis of isolated chloroplasts was investigated by following the incorporation of 14C-leucine into isolated chloroplasts from Nicotiana tabacum. The incorporation activity varied greatly during the year, being largest in the winter and smallest in the summer. Conversely, the relative effect of kinetin on the incorporation of 14C-leucine, whether applied as a pretreatment to the leaves or directly in the incubation medium, was largest in the summer and smallest or absent altogether in the winter. Kinetin did not prolong the net incorporation period, which lasted about 40 min, but only enhanced the initial rate of the reaction. Chloroplasts extracted from leaves that had been detached for 24 or 48 h displayed very little of their original, pre-aged incorporation activity and treating the leaves with kinetin did not, essentially, prevent this loss. It was concluded that the major effect of kinetin upon chloroplasts may be related primarily to an effect upon hydration and permeability of the chloroplast and its membranes, and not to an effect directly upon its machinery for protein synthesis.  相似文献

Astorga and Williams1 have demonstrated that leucocytes from some patients with rheumatoid arthritis (RA) have an impaired one-way mixed leucocyte reaction2 (MLR) when stimulated with leucocytes from other such patients. No definite explanation of this phenomenon is given. We have investigated other diseases in which auto-immune aetiology is suspected to see whether similar phenomena are associated with them.
