N-ras is one of the transforming genes in human hepatic cancer cells.It has been found that N-ras was overexpressed at the mRNA and protein level in hepatoma cells.In order to explore the biological roles of N-ras in human hepatic carcinogenesis and the potential application in control of cancer cell growth,a preudotype retrovirus containing antisense sequence of human N-ras was constructed and packaged.A recombinant retrovirus vector containing antisense or sense sequences of N-ras cDNA was constructed by pZIP-NeoSV(X)1.The pseudotype virus was packaged ang rescued by transfection and infection in PA317 and ψ 2 helper cells.It has been demonstrated that the pseudotype retrovirus containing antisense N-ras sequence did inhibit the growth of human PLC/PRF/5 hepatoma cells accompanied with inhibition of p21 expression,while the retrovirus containing sense sequence had none.The pseudotype virus had no effect on human diploid fibroblasts.  相似文献

CFTR is a chloride channel that is required for fluid secretion and salt absorption in many exocrine epithelia. Mutations in CFTR cause cystic fibrosis. CFTR expression influences some ion channels, but the range of channels influenced, the mechanism of the interaction and the significance for cystic fibrosis are not known. Possible interactions between CFTR and other ion channels were studied in C127 mouse mammary epithelial cell lines stably transfected with CFTR, ΔF508-CFTR, or vector. Cell lines were compared quantitatively using an 125I efflux assay and qualitatively using whole-cell patch-clamp recording. As expected, 125I efflux was significantly increased by forskolin only in the CFTR line, and forskolin-stimulated whole-cell currents were time- and voltage independent. All three lines responded to hypotonic challenge with large 125I efflux responses of equivalent magnitude, and whole-cell currents were outwardly rectified and inactivated at positive voltages. Unexpectedly, basal 125I efflux was significantly smaller in the ΔF508-CFTR cell line than in either the CFTR or control cell lines (P < 0.0001), and the magnitude of the efflux response to ionomycin was largest in the vector cell line and smallest in the cell line expressing ΔF508-CFTR (P < 0.01). Whole-cell responses to ionomycin had a linear instantaneous I-V relation and activated at depolarizing voltages. Forskolin responses showed simple summation with responses to ionomycin or hypotonic challenge. Thus, we found no evidence for interactions between CFTR and the channels responsible for swelling-mediated responses. Differences were found in basal and ionomycin-stimulated efflux, but these may arise from variations in the clonally selected cell lines that are unrelated to CFTR expression. Efficient expression of herpes simplex virus genes requires the synthesis of functional ICP4, a nuclear phosphoprotein that contains a prominent serine-rich region between amino acids 142 and 210. Residues in this region not only are potential sites for phosphorylation but also are involved in the functions of ICP4. By comparing the growth of a virus in which this region is deleted (d8-10) with wild-type virus (KOS) in PC12 cells or PC12 cells that are deficient in cyclic AMP-dependent protein kinase (PKA), two observations were made: (i) the growth of wild-type virus was impaired by 1 to 2 orders of magnitude in the PKA-deficient cells, indicating the involvement of PKA in the growth cycle of herpes simplex virus type 1, and (ii) while the growth of d8-10 was impaired by almost 2 orders of magnitude in wild-type cells, it was not further impaired (as was that of wild-type virus) in PKA-deficient cells, implicating the region deleted in d8-10 as a possible target for cellular PKA. In trigeminal'ganglia of mice, the d8-10 mutant virus grew poorly; however, it established latency in nearly 90% of ganglia tested. Studies of the phosphorylation of wild-type and d8-10 ICP4 proteins revealed that the serine-rich region is a major determinant for phosphorylation of ICP4 in vivo and that the phosphorylation state could change as a function of the PKA activity. Consistent with this observation, the serine-rich region of ICP4 was shown to be a target for PKA in vitro. While intact ICP4 was readily phosphorylated by ICP4 in vitro, the d8-10 mutant ICP4 was not. Moreover, a synthethic peptide representing a sequence in the serine tract that is predicted to be a substrate for PKA was phosphorylated by PKA in vitro, having a Km within the physiological range. These data suggest that PKA plays a role in viral growth through phosphorylation of one or more sites on the ICP4 molecule.  相似文献

Brown Shrikes Lanius cristatus are both transients and winter residents in Taiwan. Large numbers of birds crowd into the available habitat during the transient period, and the cost of maintaining territories changes greatly, depending on the number of shrikes present. This study examined how wintering Brown Shrikes adapt to the changing level of competition in territory establishment. About 24% of autumn transients arrived before any wintering birds, while 73% of wintering birds arrived during autumn transient time. The frequency of aggression recorded was significantly related to the number of birds seen along the transect line that day. Among ringed birds that returned during transient time, 80% returned directly to their previous territories, but some waited for the departure of transient birds before establishing territories. Territory compression occurred at high population densities. Departure from and arrival at the study area were frequent during the winter, but no birds moved territories within the study area. On average, 25% of ringed wintering shrikes returned to the study site in the subsequent years, while no transients returned. This low site tenacity may be. in part, a result of a high persecution rate along the migration route.  相似文献

抗IL-2受体α亚基的单克隆抗体不能阻断IL-2的中枢镇痛作用,以及丧失与IL-2受体β亚基结合能力的IL-2突变体仍具有提高大鼠痛阈的能力,这表明IL-2的中枢镇痛作用并不是通过IL-2受体所介导,亦表示IL-2的免疫和镇痛作用是通过不同的受体途径实现的。加之内源性阿片肽与IL-2分子有着共同的抗原决定基和结构相似性,提示IL-2可以与阿片受体直接结合产生中枢镇痛效应。从放射免疫法测定的IL-2侧脑室注射后不同时间大鼠脑内不同核团的内源性阿片肽含量,推测IL-2的中枢镇痛作用可能还与弓状核、室旁核、蓝斑等核团的β-EP和LEK有关。  相似文献

分别以猴头菇干粉(猴头菇Ⅰ组)和猴头菇浸出液(猴头菇Ⅱ组)饲喂小鼠,观察猴头菇对小鼠血清乳酸脱氢酶(LDH)活力、血乳酸、血清尿素氮(BUN)、肝糖原、肌糖原含量及运动耐力的影响。结果表明:实验60d后,猴头菇Ⅰ、Ⅱ组LDH活力、肝糖原及肌糖原含量明显高于对照组(P<0.05或P<0.01);运动后血乳酸的水平和BUN的增量明显低于对照组(P<0.05或P<0.01);运动后血乳酸消除速率显著高于对照组(P<0.05);在运动耐力测定时在水中淹死的时间比对照组长得多(P<0.05)。提示:猴头菇具有明显的增强运动能力和解除疲劳的作用。  相似文献

内皮素(endothelin,ET)是已知的体内活性最强的缩血管物质,其缩血管作用由G蛋白偶联受体所介导。但ET强大的促血管平滑肌细胞(VSMC)增生效应的机理尚未完全阐明。本研究选用培养的兔胸主动脉VSMC,探讨丝裂素活化蛋白激酶(MAPK)在ET促细胞增生中的作用。结果表明:ET-1呈时间和浓度依赖性地促进细胞摄取 ̄3H-TdR和激活MAPK,此作用可被蛋白激酶C(proteinkinaseC,PKC)抑制剂Staurosporine(STP),H-7和ET_A受体拮抗剂BQ123所抑制,但不被酪氨酸激酶抑制剂HerbimycinA(Herb)所抑制,用PKC激动剂PMA(Phorbolmyristateacetate)预处理VSMC,使其PKC活性下调,可显著减弱ET-1对MAPK的激活能力。本结果提示:(1)MAPK参与ET-1所致的VSMC增生;(2)ET-1促细胞增生与激活MAPK的作用是由ET_A受体和PKC介导的。  相似文献
