Nine nuclear‐encoded microsatellites from an enriched genomic DNA library of the HAB (harmful algal bloom) dinoflagellate Karenia brevis were isolated and characterized. The microsatellites include five perfect (three dinucleotide and two trinucleotide) and four imperfect (two dinucleotide and two trinucleotide) repeat motifs. Gene (haplotype) diversity ranged from 0.153 to 0.750 among a sample of 13 isolates; the number of alleles among the isolates ranged from two to six and pairwise tests of genotypic disequilibria were nonsignificant. The microsatellites developed in this study will provide insight into the genetic diversity of this HAB species and tools that may prove useful in predicting source populations and physiological parameters of individual K. brevis blooms.  相似文献

Increasing rates of atmospheric nitrogen (N) deposition may reduce growth and accelerate decomposition of Sphagnum mosses in bogs. Sphagnum growth and rates of Sphagnum litter decomposition may also vary because of climate change as both processes are controlled by climatic factors. The initial purpose of this study was to assess if growth and litter decomposition of hummock and lawn Sphagnum species varied with increasing N input in a factorial mid‐term (2002–2005) experiment of N and phosphorus (P) addition, in a bog on the southern Alps of Italy. However, as the experimental period was characterized by an exceptional heat wave in summer 2003, we also explored the interacting effects of fertilization and strongly varying climate on growth and decomposition rates of Sphagnum. The heat wave implied strong dehydration of the upper Sphagnum layer even if precipitation in summer 2003 did not differ appreciably from the overall mean. Sphagnum production was somewhat depressed by high levels (3 g m−2 yr−1) of N addition without concomitant addition of P presumably because of nutrient imbalance in the tissues, but production rates were much lower than the overall means in 2003, when no effect of nutrient addition could be observed. Adding N at high level also increased the potential decay of Sphagnum litter. Higher CO2 emission from N‐fertilized litter was due to amelioration of litter chemistry showing lower C/N quotients in the N‐fertilized treatments. Rates of CO2 emission from incubated litter also were more strongly affected by water content than by nutrient status, with practically no CO2 emission detected when litter was dry. We conclude that higher rates of atmospheric N availability input may depress Sphagnum growth because of P, and presumably potassium, (co‐)limitation. Higher N availability is also expected to promote potential decay of Sphagnum litter by ameliorating litter chemistry. However, both effects are less pronounced if the growing Sphagnum apex and the underlying senescing tissues dry out.  相似文献

ABSTRACT As quality of forested habitat declines from altered fire regimes, gopher tortoises (Gopherus polyphemus) often move into ruderal areas to the detriment of the animal and land manager. We evaluated effects of a dormant-and-growing-season prescribed fire on habitat and gopher tortoise use of degraded longleaf pine (Pinus palustris) forests surrounding military training areas. We burned 4 of 8 sites in winter 2001–2002 and again in April 2003. Changes in vegetation measured during 2001–2004 indicated that burn treatments did not increase herbaceous vegetation. Similarly, movement patterns, burrow usage, and home range of tortoises radiotracked from 2002–2004 did not differ between treatments. Woody cover initially was reduced in the forests postburn, and we found more new burrows in burned forest sites. Once shrub cover was reduced, tortoises started using forested habitat that had become overgrown. However, shrub reduction may be temporary, as woody stem densities increased postburn. Thus, the one-time use of fire to manage tortoise habitat may not rapidly restore the open canopy, sparse woody midstory, and abundant herbaceous vegetation that this species requires. Repeated prescribed fires or additional management techniques may be needed for complete restoration.  相似文献

This commentary discusses the findings and conclusions of the paper “Drug resistant human Staphylococcus aureus findings in sanctuary apes and its threat to wild ape populations.” This paper confirms the zoonotic transfer of Staphylococcus aureus in a sanctuary setting. The assertion that this in itself is enough to reconsider the conservation potential of ape reintroduction provides an opportunity to discuss risk analysis of pathogen transmission, following IUCN guidelines, using S. aureus as an example. It is concluded that ape reintroduction projects must have disease risk mitigation strategies that include effective biosecurity protocols and pathogen surveillance. These strategies will assist with creating a well planned and executed reintroduction. This provides one way to enforce habitat protection, to minimise human encroachment and the risks from the illegal wildlife trade. ABSTRACT The United States Air Force (USAF) uses part of Sonoran pronghorn (Antilocapra americana sonoriensis) habitat for bombing exercises (i.e., Barry M. Goldwater Air Force Range [BMGR], southwest AZ, USA) that could be detrimental to the endangered subspecies. To minimize injury or death to Sonoran pronghorn, the USAF and United States Fish and Wildlife Service developed a monitoring protocol that would eliminate live ordnance delivery in the vicinity of pronghorn. From 1998 to 2003, we searched for pronghorn on or near military targets prior to ordnance delivery. If we observed pronghorn within 5 km of a target, the target was closed for ≥24 hours. We monitored bombing ranges on BMGR and closed >5,000 targets for >1,000 days due to military activity. To our knowledge, no pronghorn were killed or injured. We recommend that the monitoring program continue as long as military activity occurs in pronghorn habitat.  相似文献

Developing genetic markers for the identification of recently diverged groups, such as ecotypes or species complexes, remains difficult due to challenges with incomplete lineage sorting, hybridization and introgression. Genome‐wide scans of single nucleotide polymorphisms (SNPs) have proven useful for inferring patterns of genetic differentiation at the population level. In combination with a new analytical technique, the discriminant analysis of principal components (DAPC), and within the framework of iterative taxonomy, it may also be possible to extract a combination of SNPs as markers for the delimitation of closely related groups. In addition, since DAPC identifies the loci contributing the most to group clustering, it may be possible to link putative biological function to differences that define group boundaries. We tested this technique on two ecotypes of the hemlock looper (Lambdina fiscellaria), which differ in terms of number of larval stadia, developmental rate and fecundity. It was possible to separately cluster the two ecotypes with 95% correct assignment using 27 SNPs. We also determined that a storage hexamerin carried eight of these SNPs, including the two highest contributing loci, of which the top contributor was nonsynonymous. Other studies have found this protein to be highly expressed just before metamorphosis, pointing to a possible connection between its role in clustering ecotypes and its biological function. These SNP markers can now be further developed for high throughput delimitation of individuals of unknown ecotype identity.  相似文献

Dysplastic Nevus Syndrome (DNS) has been defined as that trait characterized by the presence of at least one dysplastic melanocytic nevus. DNS was originally described in kindreds having multiple members with melanoma. Various types DNS have been described in other situations to include individuals with apparently sporadic cases, familial DNS without melanoma and individuals with apparently sporadic DNS with melanoma. These categories are based on historical information in general, and not on examination of family members. In all cases, the presence of dysplastic nevi appear to confer some increased risk of melanoma, which varies between the groups. Similarly cutaneous melanoma is thought to occur in several distinct populations-random individuals without DNS, individuals with sporadic DNS, and those with familial DNS. Genetic analysis of DNS has been largely confined to the classically ascertained kindreds associated with melanoma. These studies have usually used diagnostic criteria based on pathology of clinically selected material, and that evidence suggests that DNS is inherited as an autosomal dominant trait in these families. Surveys of the general population have detected rates of dysplastic nevi of 5% 20%. In our Utah-based studies, we have evaluated probands and family members from three groups. These included kindreds with multiple occurrences of melanoma, random individuals with at least one dysplastic nevus, and cases of melanoma with unknown family history. Controls were spouses of study subjects. We sought to determine the percentage of each group associated with dysplastic nevi and/or genetic DNS. The range of phenotype of patients with dysplastic nevi was large with some individuals having few nevi, none of which were clinically atypical, and others having greater than 100 nevi. The prevalence of dysplastic nevi in at least one of two biopsies in Utah population controls is presently Wtimated at 62%. Some probands with melanoma as well as some of their relatives had elevated numbers of nevi, suggesting that this predisposition to melanoma may be inherited.  相似文献
