The N-terminal signal sequence of glucitol pcrmease of Escherichia Coli (Gut22) and its analogue (Gut22Ana) were synthesized. The analogue had a Pro residue substituting for the His at the 7th position of Gut22 and a Val residue substituting for the Glu at the 10th position. The intrinsic fluorescence emission spectra indicated that the binding of Gut22 with lipid bilayer was much stronger than that of Gut22Ana. The leakage experiments with calcein-loaded liposomes showed that Gut22 strongly perturbed lipid bilayers while Gut22Ana did not. The apparent partition constant of Gut22 for partitioning into phosphatidylserine/phosphatidylcholine bilayers was measured; the effect of membrane potential on the interaction of Gut22 with lipid bilayers was studied and the conformation changes of Gut22 and Gut22Ana upon interacting with liposomes were studied by the method of circular dichroism analysis.  相似文献

Repair activities of thymine radical anion by echinocoside, isolated from Pedicularis plicata. were studied using pulse radiolysis technique. The thymine radical anion was produced by the reaction of hydrated electron with thymine. Echinocoside. one of the polyphenols of phenylpropanoid glycoside, was added to the thymine aqueous solution saturated with N2. Kinetic analysis by transient absorption spectrum showed that thymine radical anion was formed at first, and then after several decades of microseconds of pulse radiolysis. the spectrum of thymine radical anion was changed to that of echinocoside radical anion. The evidence indicated that thymine radical anion was repaired through one-electron-transfer between the DNA base radical anion and echinocoside. The rate constant of electron transfer by echinocoside was 1.45× 109 dm3 · mol1 · s 1.  相似文献

本文利用时间序列方法(三次指数平滑法),并结合季节指数法,建立了板齿鼠种群数量中长期预测(6个月~1年)的时间序列模型,依据1991年11月~1993年12月在广东省博罗县石湾镇里波水管理区对板齿鼠种群数量动态的调查数据,分别预测了1994年和1995年板齿鼠种群数量的发生,预测准确率达到了86.56%。  相似文献

应用超薄层聚丙烯酰胺凝胶等电聚焦电泳法, 分析了潮汕地区216例无血缘关系、临床上诊断为食管癌的病人和216例健康人的运铁蛋白(Tf)亚型分布情况, 结果发现:食管癌病人组TfC1C1纯合子频率为0.2639,Tf*Cl基因频率为0.4745,显著低于正常人组(分别为0.4352和0.6227,均为P<0.0 01);同时,食管癌病人组TfC2C2纯合子频率为0.2278,Tf*C2基因频率为0. 4977,显著高于正常对照组(分别为0.1852,P<0.05,和0.3634,P<0.001)。应用超薄层聚丙烯酰胺凝胶等电聚焦结合免疫固定法,分析了潮汕地区21 7例无血缘关系的临床上诊断为食管癌病人和 217例健康人的组特异性成份(Gc)亚型的分布,发现两组间无显著性差异。  相似文献

Agkistrodotoxin, a neutral phospholipase A2 with high presynaptic neurotoxicity from the venom of Agkistrodon halys Pallas, has been crystallized by hanging drop vapor diffusion method. The crystal belongs to P21 space group with the cell dimensions a = 10.836 nm, b=8.486nm, c = 7.082nm, β=109.87$ showing C2 pseu-do-symmetry. Diffraction data to 0. 26 nm resolution have been collected on a Siemens X-200B area detector. C2 pseu-do-symmetry suggests that there exists a non-crystallographic two-fold axis parallel to crystallographic b axis. Self-rotation function calculation with different integrated radius and resolution ranges using the program POLARRFN yields four stable high peaks corresponding to three more non-crystallographic two-fold axis and one special non-crystal-lographic symmetry. The molecules in the asymmetric unit are suggested to be arranged in a manner of "dimer of dimers" by inference.  相似文献

本实验在31只清醒自由行动的雄性SD大鼠进行。结果如下:(1)双侧中脑腹侧被盖区(VTA)微量注射3.3nmol溴隐亭后第2—3h觉醒时间显著增加(P<0.01);6.6nmol溴隐亭有类似效果;0.66和1.33nmol溴隐亭无明显作用。(2)同样方法VTA微量注射2nmol和4nmolSCH23390后第2—3h觉醒时间明显减少(分别为P<0.01和P<0.05),但注射3.4nmol舒必利则无效。(3)红藻氨酸(0.3μg)双侧VTA注后一周,大鼠白天觉醒减少,夜间无明显变化。以上结果表明溴隐亭微量注射到VTA可能通过D1受体使多巴胺神经元活动增加而产生致觉醒作用;另外VTA中DA神经元对于维持日间的觉醒可能有紧张性作用。  相似文献

Tall fescue (Festuca arundinacea Schreb.) and white clover (Trifoliumrepens L.) were examined for Se assimilation and nutrient elementuptake. Substantial Se tolerance difference was found betweenthe two species. An inverse relationship between Se accumulationand Se tolerance suggests an exclusion mechanism that restrictsSe uptake by the plant with greater Se tolerance. A positiverelationship between the increase of protein Se concentrationand growth inhibition in the plants suggests that assimilationof Se into proteins is responsible for the Se toxic effect andthis study indicates that the antagonistic effect of sulphateis responsible for reducing Se toxicity at the protein level.No evidence that a Se exclusion mechanism which excludes Sefrom incorporating into protein, such as that found in Astragalusspecies, plays any major role of Se tolerance in these two species. Under Se treatment, plant tissue Ca concentration was increased,but P concentration was decreased. A higher Fe concentrationwas found in white clover which increased with increasing tissueSe concentration. Copper, Mn, and Zn concentrations only increasedin the white clover under conditions of severe growth inhibition.
