From a gene bank of theAcinetobacter calcoaceticus genome a plasmid was isolated that complements four different classes of PQQ- mutants. Subclones of this plasmid revealed that the four corresponding PQQ genes are located on a fragment of 5 kilobases. The nucleotide sequence of this 5 kb fragment was determined and by means of Tn5 insertion mutants the reading frames of the PQQ genes could be identified. Three of the PQQ genes code for proteins of Mr 29700 (gene I), Mr 10800 (gene II) and Mr 43600 (gene III) respectively. In the DNA region where gene IV was mapped however the largest possible reading frame encodes for a polypeptide of only 24 amino acids. A possible role for this small polypeptide will be discussed. Finally we show that expression of the four PQQ genes inAcinetobacter lwoffi andEscherichia coli lead to the synthesis of the coenzyme in these organisms. 相似文献

Summary DNA markers in the pericentromeric region of human chromosome 21 have shown linkage to a gene for Familial Alzheimer disease (FAD; St. George Hyslop et al. 1987). The limited informativeness of probes for the loci D21S13 and D21S16 have hindered precise mapping of the FAD locus and analysis of non-allelic heterogeneity in FAD (Schellenberg et al. 1988; St. George-Hyslop et al. 1987). We recently described a new EcoRII polymorphism at the D21S13 locus that was very informative in a large FAD pedigree (Pulst et al. 1990a, b). We now report another polymorphism for the D21S13 locus that further increases the informativeness of this locus. 相似文献

为进一步研究脊髓 P 物质(SP)在调节心血管活动中的作用,在大鼠脊髓蛛网膜下腔注射(ith)辣淑素(cap),以刺激脊髓 SP 能神经末梢释放 SP,结果引起血浆去甲肾上腺素(NA)和肾上腺素(AD)含量增高,及具有剂量依赖性的动脉血压上升,心率升高。ith 具有高度特异性的 SP 受体拮抗剂或 SP 抗血清均可阻断 cap 引起的升压效应,免疫组化测定也观察到注入的cap 剂量越大,脊髓胸段 SP 样免疫阳性反应物的致密度越低,这些观察结果支持 cap 可以引起脊髓内 SP 的释放的说法。在第一颈段(C_1)横断脊髓后 ith cap 所引起的升压效应与完整动物 ith cap 的升压效应无显著差异。以上结果提示脊髓 SP 能神经末梢释放的 SP 可以通过交感肾上腺髓质系统引起心血管兴奋效应,SP 可能是引起交感节前神经元兴奋的神经递质。 相似文献

从腹侧面暴露家兔延髓,脑内微量注射1%普鲁卡因阻滞面神经后核内侧区(mNRF),全部动物(n=20)一次注射(0.3—1.0μl)后即能可逆地消除呼吸节律。区域对照显示此区非常局限,范围约1.0×1.0×1.0mm。组织学检查表明为面神经后核内侧区。本文分析了 mNRF的呼吸相关神经元(RRNs)的放电形式。在 mNRF 有较多的呼气(E)神经元和呼气-吸气跨时相(E-IPS)神经元。在阻滞 mNRF 引起呼吸停止期间,观察到低位延髓背侧呼吸群(DRG)和腹侧呼吸群(VRG)尾端区 RRNs 放电的节律性消失,表现连续放电或停止放电。电刺激DRG,VRG 尾端区,只能诱发短串的膈神经放电,而不能产生节律性发放。说明这些区域的RRNs 无自动节律性活动的能力。结果表明,面神经后核内侧区与呼吸节律发生有关,它可能是呼吸节律发生器的一个重要的所在部位。 相似文献

电刺激大鼠下丘脑室旁核(PVH),在同侧中脑中央灰质(CG)内寻找逆行及顺行反应单位,然后观察它们对躯体感觉刺激的反应。实验结果表明:CG 及邻近网状结构内有10%(32/318)的单位呈逆行反应。逆行传导速度平均为0.37±0.24m/s(均数±标准差);推测这种CG→PVH 投射纤维属于细有髓或无髓神经纤维。这些单位分布于 CG 的腹外侧及背外侧亚核。50%(14/28)的逆行单位对坐骨(胫)神经的强电刺激和夹尾等损伤性刺激起反应,但对触毛或低强度的神经干刺激无明显反应。以上结果表明:外周躯体感觉,特别是损伤性信息传入 PVH 时,CG 是其中枢驿站之一。电刺激 PVH 还能顺行激活7.55(24/318)、抑制0.7%(2/318)的 CG 单位。有69%(18/26)的顺行反应单位对外周躯体神经强电刺激及夹尾起反应。提示 PVH 可能通过影响 CG神经元的活动而参与中枢痛觉的整合。 相似文献

本文报道用人造寄主卵繁殖平腹小蜂Anastatus japonicus Ashmead***成功的结果.筛选出最佳卵壳材料为32—36μm的聚丙烯膜,培养基为柞蚕蛹血淋巴44.4%、10%麦乳精33.3%、鸡蛋黄11.1%、尼氏盐11.1%.体外连代培养平腹小蜂的结果表明,除蛹化率(72—83%)外,各代间在寄生率(40—44%)、孵化率(94—96%)、羽化率(91—96%)、展翅率(97—99%)方面无明显的差别,且人造卵育出的各代蜂在身体大小、寿命及繁殖力方面均与柞蚕卵育出蜂基本相似或优于柞蚕卵育出蜂.筛选出的人工培养基的氨基酸种类与蓖麻蚕卵和柞蚕卵相同,但量上差异比较明显.本文还报道了平腹小蜂在人造寄主卵上的产卵过程. 相似文献

The effect of dynamic exercise on the NADH content of human type I (slow-twitch) and II (fast-twitch) muscle fibres was investigated. Muscle biopsy samples were obtained from the quadriceps femoris of seven healthy subjects at rest and after bicycle exercise at 40, 75 and 100% of the maximal oxygen uptake [VO2(max.)]. At rest and after exercise at 100% VO2(max.), muscle NADH content was significantly higher (P less than 0.05) in type I than in type II fibres. After exercise at 40% VO2(max.), muscle NADH decreased in type I fibres (P less than 0.01), but was not significantly changed in type II fibres. After exercise at 75 and 100% VO2(max.), muscle NADH increased above the value at rest in both type I and II fibres (P less than 0.05). Muscle lactate was unchanged at 40% VO2(max.), but increased 20- and 60-fold after exercise at 75 and 100% VO2(max.) respectively. The finding that NADH decreased only in type I fibres at 40% VO2(max.) supports the idea that type I is the fibre type predominantly recruited during low-intensity exercise. The increase of NADH in both fibre types after exercise at 75% and 100% VO2(max.) suggests that the availability of oxygen relative to the demand is decreased in both fibre types at high exercise intensities. 相似文献

The results reported in this paper show two distinct ways for the incorporation ofN-acetylglucosamine into mitochondrial outer membranes. The first one is the glycosylation of dolichol acceptors, which is indicated by the inhibition of the synthesis of these products by the inhibitors of the dolichol intermediates (tunicamycin and GDP). The second one is the incorporation ofN-acetylglucosamine into protein acceptors directly from UDP-N-acetylglucosamine. This second way of glycosylation is only localized in mitochondria outer membranes.The existence of a direct route forN-glycoprotein biosynthesis has been based on the following evidence. First, the synthesis of theN-acetylglucosaminylated protein acceptors was not inhibited by tunicamycin or GDP. Second, the addition of exogenous dolichol-phosphate did not change the rate of biosynthesis of glycosylated protein material. Third, the sequential incorporation ofN-acetylglucosamine and mannose from their nucleotide derivatives in the presence of GDP and tunicamycin led to the synthesis of glycosylated protein material which entirely bound to Concanavalin A-Sepharose. The oligosaccharide moiety of the glycosylated protein material resulting from the direct transfer of sugars from their nucleotide derivatives to the protein acceptor is of theN-glycan type. On sodium dodecylsulphate polyacrylamide gel electrophoresis, this glycosylated material migrated as a marker protein with a molecular weight between 45 000 and 63 000. HPLC chromatofocusing analysis revealed that the fraction studied was anionic. The oligosaccharide moiety of the glycoprotein material can only be elongated by the incorporation ofN-acetylglucosamine and galactose from their nucleotide derivatives. 相似文献

Effect of Inorganic Phosphate on the Biosynthesis of Purine and Pyrimidine Nucleotides in Suspension-Cultured Cells of Catharanthus roseus 总被引:1,自引:0,他引:1
The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (+Pi)or phosphate-deficient (Pi) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in +Pimedium than in the cells grown in Pi medium.The levels of almost all other nucleotides were slightly higherin the cells in +Pi medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [214C]glycine, NaH14CO3, [614C]orotate,[814C]adenine, [814C]adenosine, [214C]uraciland [214C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in +Pi medium than in the cells in Pimedium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin +Pi medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed.
