Impedance spectroscopy is a technique that reveals information, such as macromolecular charges and related properties about protein suspensions and other materials. Here we report on impedance measurements over the frequency range of 1 Hz to 1 MHz of alpha-beta tubulin heterodimers suspended in a buffer. These and other polyelectrolyte suspensions show enormous dielectric responses at low frequencies, due both to the motion of charges suspended in the medium and to an electrical double layer that forms at each electrode-medium interface. We propose an equivalent circuit model to minimize electrode polarization effects and extract the intrinsic response of the bulk medium. At megaHertz frequencies, the conductivity increases with concentration below the critical concentration of approximately 1 mg/ml for microtubule polymerization, above which the conductivity decreases. This suggests that such measurements can be used to monitor the dynamics of microtubule polymerization. Finally, we obtain the net charge number per tubulin dimer of /Z/ = 306 in the saline buffer, which, if maintained as the dimers polymerized, would yield a linear charge density of 3.8 e/angstroms for the assembled microtubules. These results are potentially important for fundamental electrostatic processes in biomolecules and suggest the possibility of developing future bioelectronic applications. 相似文献

We report two experiments designed to investigate the nature of any cross-modal interactions between olfactory and tactile information processing. In Experiment 1, we assessed the influence of olfactory cues on the tactile perception of fabric softness using computer-controlled stimulus presentation. The results showed that participants rated fabric swatches as feeling significantly softer when presented with a lemon odor than when presented with an animal-like odor, demonstrating that olfactory cues can modulate tactile perception. In Experiment 2, we assessed whether this modulatory effect varied as a function of the particular odors being used and/or of the spatial coincidence between the olfactory and tactile stimuli. The results replicated those reported in Experiment 1 thus further supporting the claim that people's rating of tactile stimuli can be modulated by the presence of an odor. Taken together, the results of the two experiments reported here support the existence of a cross-modal interaction between olfaction and touch. 相似文献

Weber's law--constancy of the coefficient of variation--is an apparently ubiquitous feature of time perception, and forms the foundation of several theories of timing. We sought evidence for Weber's law in temporal production and categorization experiments. The production task required pigeons to switch between keys within a specified temporal window. The categorization task required them to classify a stimulus duration as either short or long. Weber fractions did not descend to a horizontal asymptote, but were U-shaped: they decreased as a function of target duration, and increased again at intermediate and long durations. This pattern conforms neither to Weber's law, nor to its generalized form (Getty, D.J., 1975. Discrimination of short temporal intervals: a comparison of two models. Percept. Psychophys. 18, 1-8). We derive an analytical model of the potential differences induced across plasma and internal organelle membranes in suspended cells exposed to oscillatory electric fields. Multiple shells are modeled using iterative applications of the single-shell calculation with mobile charges. This work is motivated, in part, by recent results suggesting the ability to use alternating current (ac) fields to noninvasively monitor enzyme activity within internal membranes, particularly the mitochondrial electron transport chain. Previous work, on induced transmembrane voltages in cells subjected to ac fields, has mainly been limited to oscillatory potentials across the plasma membrane. Here we first develop a three-membrane model, consisting of a plasma membrane surrounding inner and outer membranes representing an internal organelle, such as a mitochondrion. Frequency-dependent transmembrane potentials are modeled for spherical, weakly conducting membrane shells enclosing a conductive cytoplasm surrounding an idealized internal organelle. We then use a two-shell model to simulate induced ac membrane potentials of a suspended isolated mitochondrion in which the outer membrane is usually much more permeable than the inner membrane. 相似文献

Undermatching and overmatching in concurrent schedules of reinforcement have been traditionally described as changes in the slope of the Generalized Matching Law function. More recently, it has been suggested that deviations from strict matching may be better described as following a policy of mostly fixing on the preferred schedule, and occasionally sampling the alternative schedule. So far, no model of local performance predicts the global outcome of this policy. We describe one such model; it assumes immediate and long-term effects of reinforcement on local performance. Self/nonself perception and recognition mechanisms in plants: a comparison of self-incompatibility and innate immunity 总被引:2,自引:0,他引:2
Analyses of emerging concepts indicate that parallels exist between self-incompatibility and pathogen recognition. In the case of surveillance of 'nonself', plant immune responses are triggered either by pattern recognition receptors (PRRs) that detect conserved pathogen-associated molecular patterns (PAMPs) or by resistance (R) proteins recognizing isolate-specific pathogen effectors. PAMP detection is an important component of innate immunity in plants and serves as an early warning system for the presence of potential pathogens and activation of plant defense mechanisms. In the Brassicaceae, the recognition of 'self' and self-incompatibility are components of a receptor-ligand based mechanism that utilizes an S receptor kinase (SRK) to perceive and reject 'self'-pollen. SRK is an S-domain receptor-like kinase (RLK), which in turn is part of the RLK family, some members of which represent PRRs involved in the detection of PAMPs. S-domain RLKs also occur in species that do not exhibit self-incompatibility and are up-regulated in response to wounding, PAMPs and pathogen recognition. Although evolution may have driven expansion of certain RLK families to serve roles in particular physiological processes, this may not exclude these receptor types from functioning in different programs. Recent findings on self/nonself recognition are reviewed and conceptual and mechanistic links between microbial recognition and self-incompatibility are discussed. 相似文献

Nitrite-dependent anaerobic oxidation of methane (n-damo) and ammonium (anammox) are two recently discovered processes in the nitrogen cycle that are catalyzed by n-damo bacteria, including "Candidatus Methylomirabilis oxyfera," and anammox bacteria, respectively. The feasibility of coculturing anammox and n-damo bacteria is important for implementation in wastewater treatment systems that contain substantial amounts of both methane and ammonium. Here we tested this possible coexistence experimentally. To obtain such a coculture, ammonium was fed to a stable enrichment culture of n-damo bacteria that still contained some residual anammox bacteria. The ammonium supplied to the reactor was consumed rapidly and could be gradually increased from 1 to 20 mM/day. The enriched coculture was monitored by fluorescence in situ hybridization and 16S rRNA and pmoA gene clone libraries and activity measurements. After 161 days, a coculture with about equal amounts of n-damo and anammox bacteria was established that converted nitrite at a rate of 0.1 kg-N/m(3)/day (17.2 mmol day(-1)). This indicated that the application of such a coculture for nitrogen removal may be feasible in the near future. 相似文献
