不同载体固定化胰蛋白酶酶学特性的研究   总被引:4,自引:0,他引:4  
目的:研究以壳聚糖、复合硅胶、阴离子交换树脂为载体固定化胰蛋白酶的酶学特性。方法:通过测定不同载体固定化胰蛋白酶的活力得其最适反应温度值、最适反应pH值和米氏常数(Km)值。结果:以壳聚糖、复合硅胶、阴离子交换树脂为载体制备固定化胰蛋白酶的最适反应温度分别为70℃、60℃、60℃;最适反应pH值分别为7.5、8.0、8.0;表观米氏常数K’m分别为22.72mg/ml、25.12mg/ml、29.04mg/ml。结论:与游离酶相比,固定化胰蛋白酶均表现出一定的热稳定性、酸碱稳定性,利于工业化生产。  相似文献

ABSTRACT. The presence of nonvariant antigens (NVAs) limited to bloodstream forms of Trypanosoma brucei brucei and Trypanosoma brucei rhodesiense was demonstrated for the first time by immunodiffusion and Immunoelectrophoresis. Noncloned and cloned populations were employed in preparation of polyclonal antisera in rabbits and of antigens to be used in the immunologic reactions. The NVAs could be shown best in systems in which hyperimmune rabbit sera (adsorbed with procyclic forms to eliminate antibodies against antigens common to bloodstream form and procyclic stages) were reacted with trypanosomes characterized by heterologous variant-specific antigens (VSAs). The NVAs demonstrated in this study are very likely different from the common parts of VSAs. As has been suggested by experiments with living trypanosomes, at least a part of the NVAs appears to be located on the surface of the bloodstream forms. In these experiments involving the quantitative indirect fluorescent antibody test, the amount of fluorescence recorded for the heterologous system, i.e. ETat 5 trypanosomes incubated with anti-AmTat 1.1 serum, equalled ~3.0% of the fluorescence emitted by the AmTat 1.1 bloodstream forms treated with their homologous antiserum. Evidently, only small amounts of NVAs are present on the surfaces of T. brucei bloodstream forms. In addition to the NVAs, the electrophoresis results suggested the presence of antigenic differences between procyclic stages belonging to different T. brucei stocks.  相似文献

RuBP carboxylase-oxygenase protein in three C3 species (Nicotianatabacum L., Solanum tuberosum L., Triticum aestivum L.) andthree C4 species (Panicum miliaceum L., Panicum texanum Buckl.,Zea mays L.) was quantitatively determined by sucrose densitygradient centrifugation and by immunochemical assay using antibodyraised to crystallized tobacco leaf RuBP carboxylase-oxygenase.The C3 species had 3- to 6-fold higher concentrations of RuBPcarboxylase-oxygenase than the C4 species when expressed oneither a chlorophyll or a leaf area basis. The C3 species alsoallocated a higher fraction of their total soluble protein tothis enzyme (from 25 to 60% for the C3 species compared to 8to 23% for the C4 species). There was no RuBP carboxylase-oxygenaseprotein or activity in the C4 mesophyll cells, while the enzymeconstituted from 20 to 40% of the total soluble protein in theC4 bundle sheath cells. A close correlation (r = +0·91)was found between catalytic activity and level of the enzymeprotein in the species examined.  相似文献

Subterranean termites construct complicated tunnel networks for foraging below ground. Thus, they often encounter tunnel intersections during foraging activity. Directional selection by termites at intersections is likely to affect foraging efficiency because depending on their selection, the path length from food resources to the nest can vary significantly. In order to understand how termites are guided to choose the most economical path without the use of pheromones, we artificially excavated two tunnels of varying widths (W1 and W2; 2, 3, or 4 mm) that intersected at a 90° angle in each of nine soil‐filled arenas. We observed the directional selection of termites at intersections in arenas with combinations of W1 (mm) and W2 (mm) (W1, W2). For (W1, W2) = (2, 2) and (4, 4), termites statistically equally chose the three directions left, right, and straight, while for the (3, 3) arena combination, termites preferentially decided to go straight. On the other hand, for (W1, W2) = (2, 3) (2, 4), and (3, 4), termites advancing from narrower tunnels into intersections tended to turn right or left, while termites coming from the wider tunnel were more inclined to go straight. On the assumption that a wider tunnel is more frequently used by termites in the field, we can deduce that the wider tunnel may represent the most efficient/shortest path. Thus, this simple selection mechanism can prevent termites from deviating from the most economical path.  相似文献

本工作观察到在清醒、麻痹大鼠,电刺激黑质致密部(SN_C)或网状部(SN_R)均明显抑制中缝大核(NRM)神经元的自发放电;电针刺激双侧“足三里-三阴交”的同时,电刺激SN_C或SN_R可完全翻转电针对NRM神经元的兴奋效应,NRM神经元放电受抑制。将谷氨酸钠微量注入黑质,对中缝大核神经元亦具有抑制作用,电解损毁双侧中脑导水管周围灰质(PAG)腹外侧部或将多巴胺受体阻断剂氟哌啶醇注入该处,均可阻断此抑制效应。提示黑质对抗电针镇痛机制之一是通过其DA能投射纤维作用于PAG内的DA受体,进而抑制PAG-NRM系统而实现的。  相似文献

ABSTRACT. Mice infected with the protozoan parasite Trypanosoma cruzi , the causative agent of human Chagas'disease, develop immunosuppressed responses to heterologous antigens. Experiments were performed using infected mice in the acute stage of infection to assess immunoregulatory activities during induction of direct plaque-forming cells (DPFC) to sheep erythrocytes (SRBC), hapten-conjugated SRBC (TNP-SRBC), and horse erythrocytes (TNP-HRBC). Studies in vivo demonstrated that anti-SRBC responses were best enhanced when T. cruz -infected mice were injected with primed T cells derived from normal or infected mice immunized four days previously. The presence of enhancing capacities for DPFC responses by T cells from T. cruzi -infected mice were also supported by experiments examining the hapten-carrier effect. Preimmunization of infected mice with SRBC or HRBC four days before injection of hapten-homologous (TNP-SRBC or TNP-HRBC) carrier resulted in markedly augmented anti-hapten antibody responses. These results show that functional help provided by T cells activated during priming and exposed to a challenge dose of antigen (SRBC) in a time-dependent mode can overcome the effect of immunosuppression in T. cruzi -infected mice.  相似文献

将L-谷氨酸钠(Glu)注入乌拉坦麻醉、箭毒化、人工呼吸大鼠的室旁核(NPV)或蓝斑内引起升压反应。蓝斑升压反应可被双侧室旁核内预先注射酚妥拉明或心得安明显衰减;双侧室旁核内预先注射酚妥拉明或荷包牡丹碱还可使Glu兴奋延髓A_1区引起的降压反应减小,但注射心得安对A_1-降压反应无明显影响。以上结果提示蓝斑-升压反应和A_1-降压反应均部分通过NPV实现,A_1降压过程中可能有NPV内GABA能抑制性中间神经元参与。  相似文献
