Cocos Island is a small oceanic island midway between Costa Rica and the Galápagos Archipelago; about 2 Myr in age, it is the only tropical oceanic island in the eastern Pacific with tropical wet forest. We identified several hundred bark beetle specimens collected during recent expeditions by INBio, the National Biodiversity Institute of Costa Rica, and re-examined all specimens from earlier collections. We report 19 species in ten genera, seven or eight of which are endemic, making scolytines the largest group of beetles known from the island. We describe as new Pycnarthrum pseudoinsulare , Xyleborinus cocoensis , and Xyleborus sparsegranulosus , resurrect Xyleborus bispinatus as separate from X. ferrugineus , and report six other species as new to Cocos Island. Three-quarters of the scolytines reproduce by brother–sister mating, and we argue that inbreeders are superior island colonists because they are less affected than are outbreeders by problems of mate location and inbreeding depression. The fauna and flora of Cocos Island arrived by dispersal and human transport. We examine natural colonization patterns for the fauna, using the distributions of the relatives of island endemics: most colonization came from the Americas, but the closest relatives to some endemics are found on Caribbean or Galápagos islands. © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 89 , 729–743.  相似文献

Haptoglobin (Hp) is an acute-phase protein that is produced by the liver to capture the iron that is present in the blood circulation, thus avoiding its accumulation in the blood. Moreover, Hp has been detected in a wide variety of tissues, in which it performs various functions. In addition, this protein is considered a potential biomarker in many diseases, such as cancer, including ovarian carcinoma; however, its participation in the cancerous processes has not yet been determined. The objective of this work was to demonstrate the expression of Hp and its receptor CCR2 in the ovarian cancer cells and its possible involvement in the process of cell migration through changes in the rearrangement of the actin cytoskeleton using western blot and wound-healing assays and confirming by confocal microscopy. Ovarian cancer cells express both Hp and its receptor CCR2 but only after exposure to ascitic fluid, inducing moderated cell migration. However, when the cells are exposed to exogenous Hp, the expression of CCR2 is induced together with drastic changes in the actin cytoskeleton rearrangement. At the same time, Hp induced cell migration in a much more efficient manner than did ascitic fluid. These effects were blocked when the CCR2 synthetic antagonist RS102895 was used to pretreat the cells. These results suggest that Hp-induced changes in the cell morphology, actin cytoskeleton structure, and migration ability of tumor cells, is possibly “preparing” these cells for the potential induction of the metastatic phenotype.  相似文献

1. We examined the ecological genetics of the invasive cladoceran Daphnia lumholtzi in a reservoir (Lake Texoma) in the southern USA. This species originates from the Old World subtropics and has spread across North America since the late 1980s after its inadvertent introduction to a reservoir in northeastern Texas. 2. The population genetic structure of D. lumholtzi was examined seasonally on 22 dates over a 3‐year period along a natural temperature and salinity gradient. 3. A two‐allele polymorphism at the phosphoglucose isomerase (PGI) locus was detected, while other loci tested showed no variation. Significant temporal heterogeneity existed in genotype frequencies with a major shift between summer and autumn. 4. Results of a multiple linear regression analysis revealed that a significant amount of variation was explained by day length, temperature and conductivity. Additionally, significant spatial heterogeneity of genotype frequencies between lake stations was observed, but was restricted to the summer. 5. Clonal isolates were used in controlled laboratory temperature and salinity tolerance experiments. Results suggest that salinity and temperature tolerance differed between PGI genotypes. 6. Genotype × environment interactions may play a significant role in the micro‐evolutionary dynamics of this invasive species and may have facilitated its rapid expansion across the North American continent.  相似文献

Estimation of heterozygosity for single-probe multilocus DNA fingerprints   总被引:8,自引:0,他引:8  
In spite of the increasing application of DNA fingerprinting to natural populations and to the genetic identification of humans, explicit methods for estimation of basic population genetic parameters from DNA fingerprinting data have not been developed. Contributing to this omission is the inability to determine, for multilocus fingerprinting probes, relatively important genetic information, such as the number of loci, the number of alleles, and the distribution of these alleles into specific loci. One of the most useful genetic parameters that could be derived from such data would be the average heterozygosity, which has traditionally been employed to measure the level of genetic variation within populations and to compare genetic variation among different loci. We derive here explicit formulas for both the estimation of average heterozygosity at multiple hypervariable loci and a maximum value for this estimate. These estimates are based upon the DNA restriction-pattern matrices that are typical for fingerprinting studies of humans and natural populations. For several empirical data sets from our laboratory, estimates of average and maximal heterozygosity are shown to be relatively close to each other. Furthermore, variances of these statistics based on simulation studies are relatively small. These observations, as well as consideration of the effect of missing alleles and alternate numbers of loci, suggest that the average heterozygosity can be accurately estimated using phenotypic DNA fingerprint patterns, because this parameter is relatively insensitive to the lack of certain genetic information.   相似文献

Transmission electron micrographs of starchy endosperms of germinatingsorghum indicate that the protein bodies are degraded predominantlyby progressive hydrolysis of prolamin from their surface. Theappearance of holes within partially degraded protein bodiesindicates that some internal hydrolysis also takes place. Chemicalanalyses of protein bodies isolated at different stages duringgermination showed that their amino acid composition and electrophoreticpattern remained relatively unchanged during hydrolysis. Theend result of protein body degradation was that the organellescompletely disappeared leaving empty starchy endosperm cells.The protein bodies did not swell prior to or during degradation.This mode of protein body degradation differs from that in germinatingdicotyledonous seeds and in the aleurone layer and embryo ofcereal seeds but was identical to the mode of prolamin proteinbody degradation in the starchy endosperm of germinating riceseeds. ABSTRACT It has been suggested that several Trypanosoma cruzi antigens have possible protective epitopes which may be suitable vaccine candidates. We found previously that animals resistant to T. cruzi infection produced antibodies against the 75-77-kDa parasite antigen. To test the ability of the recombinant form of this antigen to protect animals from T. cruzi infection, the cDNA which encodes a portion of the 75-77-kDa antigen was cloned using a cDNA library constructed in an orientation-specific bacteriophage expression vector (λgt11) from poly (A)+ RNA of Brazil strain epimastigotes. One clone, named SFS-40, was selected by screening the library using affinity purified antibodies specific for the 75-77-kDa parasite antigen as probe. The cDNA corresponding to the 1.7-kilobase insert of SFS-40 was subcloned into plasmid vectors and characterized. The cDNA sequence encodes a polypeptide of about 40 kDa. The putative product of the cDNA was homologous to members of the 70-kDa stress protein family. When epimastigotes were shifted from 29° C to 37° C, there was no change in the level of SFS-40 mRNA. In contrast, the 70-kDa heat shock protein mRNA of the parasite was increased about four fold by this treatment.  相似文献

In this study we investigated the causes of annual variability in peak aboveground biomass production, net ecosystem productivity (NEP) and gross ecosystem productivity (GEP) during an 8‐year period (1999–2006) in a northern Great Plains grassland near Lethbridge, Alberta, Canada. In particular, we tested for a significant relationship between growing season precipitation and productivity and determined whether soil moisture carry‐over from the previous fall–winter could alter this relationship. We also investigated the interaction between soil moisture availability and temperature in controlling grassland productivity. There was a very strong correlation between total precipitation input and average soil moisture content during the May–October growing season. However, the growing season average soil moisture contents in 2003 and 2006 were very similar to those recorded in 1999, despite lower than normal precipitation occurring in these 2 years. This resulted from a positive difference between precipitation and evapo‐transpiration that allowed significant soil moisture to be carried‐over from the previous fall–winter during both 2003 and 2006. Strong logistic relationships were observed between soil moisture and annual productivity based on data from all years except 2003 and 2006, years which had higher productivity than was predicted from the logistic regression. Interaction between temperature and soil moisture explained this difference. Productivity values in 2003 and 2006 were high compared with 1999, a year with approximately the same soil moisture content, and this resulted from the higher average growing season temperatures that were apparent in 2003 and 2006. Analysis of weather records indicated that precipitation in the month of June was significantly higher during El Niño years than during La Niña years in Lethbridge. During the study period, aboveground biomass, NEP and GEP were generally higher in El Niño years and lower in La Niña years because of associated variation in summer precipitation.  相似文献
