New synthetic methods of imidazo[4,5-e][1,4]diazepine nucleosides 8-11 and 3-substituted 3-deazainosines 14 from inosine have been reported. Treatment of N1-substituted inosines 1-3 with aqueous NaOH gave 5-amino-4-(N-substituted carbamoyl)imidazole ribosides 5-7, followed by appropriate manipulations to afford ring-expanded guanosine, inosine, and xanthosine analogues. Additionally, the 5-amino group of 4-N-allylcarbamoyl derivative 12 was converted into corresponding 5-iodo nucleoside 13. We found that 13 was cyclized to form 3-deaza-3-methylinosine (14) in the presence of Pd catalysts.  相似文献

The effect of ester bond on the conformation of peptide molecule was studied by designing and synthesizing a model tetradepsipeptide cyclo(-L-Ala-L-Hmb-)2 and by analyzing the conformation both theoretically and experimentally. Theoretical analysis showed that both ester and peptide bonds in the calculated low-energy conformations within 3 kcal/mol of the global minimum take a trans but distorted configuration. The distortion is larger in ester bonds than in peptide bonds. Further, the four carbonyls project from one side of the plane of the cyclic backbone, whereas the side chains project from the other side. These results are consistent with the experimental results obtained by NMR measurement; first, the coupling constant deduced from 1H-NMR species in DMSO-d6 is consistent with the dihedral angles of the calculated low-energy conformations; second, results of NOE measurement can reproduce the calculated configuration of the carbonyls and side chains. From the consistency between theoretical and experimental results, it is concluded that this model tetradepsipeptide takes an all-trans backbone conformation in solution and this backbone conformation is stabilized by large distortion in the ester bond, which compensates the strain resulted from the 12-membered cyclic backbone structure consisting only of L-residues.  相似文献

Immunohistochemical analysis of the distribution of type VI collagen in the lingual mucosa of rats during the morphogenesis of filiform papillae. —Acta Zoologica (Stockholm) 93 : 80–87. We examined the distribution after immunostaining of immunofluorescence of type VI collagen, differential interference contrast (DIC) images, and images obtained using confocal laser‐scanning microscopy in transmission mode, after toluidine blue staining, during morphogenesis of the filiform papillae, keratinization of the lingual epithelium and myogenesis in the rat tongue on semi‐ultrathin sections of epoxy resin‐embedded samples. Immunoreactivity specific for type VI collagen was dispersed over a relatively wide range of connective tissue in the mesenchyme of fetuses on day 15 after conception (E15), at which time the lingual epithelium was composed of one or two layers of cuboidal cells and the lingual muscle was barely recognizable. Slight immunoreactivity specific for type VI collagen was scattered within the lamina propria in fetuses on E17 and on E19, and immunoreactivity was relatively distinct on the connective tissue around the lingual muscle during myogenesis. In fetuses on E19, the epithelium was already stratified squamous. At postnatal stages from P0 to P14, keratinization of the lingual epithelium advanced gradually as morphogenesis of the filiform papillae proceeded during postnatal development. In newborns on P0, myogenesis of the tongue was almost completed. The intensity of immunoreactivity specific for type VI collagen at postnatal stages was mainly restricted on the endomysium and perimysium around the lingual muscle, while scant immunoreactivity was evident in the connective tissue in the lamina propria. Immunoreactivity around the fully mature lingual muscle on P7 and P14 was weaker than that on E19 and P0. Thus, type VI collagen appeared in the connective tissue that surrounded the lingual muscles such as the endomysium and perimysium, in parallel with changes in extracellular components during myogenesis of the tongue.  相似文献

We examined the renin-angiotensin-aldosterone system in seven patients with Shy-Drager syndrome by studying their response to the stimulation of 1 mg/kg furosemide injection followed by sitting for 1 hour. Six of the seven patients showed a low response of plasma renin activity to the stimulation. However, in five of the low responders, the plasma aldosterone levels after stimulation were observed to be similar to those of the control subjects; in addition, an increment in the plasma cortisol level appeared although no such increment was observed in normal subjects. Next, we studied the aldosterone response to angiotensin II. The five patients who showed a low plasma renin activity response and a normal aldosterone response to furosemide administration also showed low plasma aldosterone response to angiotensin II. Furthermore, in the patients who demonstrated a low plasma renin activity response and a normal aldosterone response to furosemide administration, the pretreatment with 2 mg dexamethasone for 2 days caused a marked inhibition of aldosterone response to the stimulation. These findings suggested that in most patients with Shy-Drager syndrome, the plasma aldosterone response to the stimulation of furosemide injection followed by sitting for 1 hour might be controlled by ACTH but not by plasma renin activity.  相似文献

Abstract

It was found that synthetic amphiphilic α-helix peptide could bind stabilize double or triple stranded DNA. The stabilization effect was significant for cationic α-helix peptides which indicated the importance of electrostatic interaction of positive charge of peptide and negative charge of DNA. Phosphoramidon, N-(α-l-rhamnopyranosyloxyhydroxyphosphinyl)-l-leucyl-l-tryptophan, and its analog, N-phosphoryl-l-leucyl-l-tryptophan, inhibited thermolysin in a competitive manner and Ki values were calculated to be 2.8 × 10?8 and 2.0 × 10?9m, respectively. The l-rhamnose moiety in phosphoramidon was suggested to be not involved in inhibition of thermolysin. A phosphoramidon analog containing histidine instead of tryptophan showed weaker inhibition. Spectrophotometric titration based on difference ultraviolet absorption spectra of the enzyme-inhibitor complex showed equimolar binding of the inhibitor to the enzyme.  相似文献

Harris et al. [P.V. Harris, O.M. Mazina, E.A. Leonhardt, R.B. Case, J.B. Boyd, K.C. Burtis, Molecular cloning of Drosophila mus308, a gene involved in DNA cross-link repair with homology to prokaryotic DNA polymerase I genes, Mol. Cell. Biol., 16 (1996) 5764-5771.] reported the molecular cloning of Drosophila mus308 gene, and its nucleotide and protein sequences similar to DNA polymerase I. In the present study, we attempted to find and isolate the gene product by purifying a DNA polymerase fraction not present in mus308 flies. A new DNA polymerase with properties different from those of any known polymerase species was identified and partially purified from the wild-type fly embryos through ten column chromatographies. The enzyme was resistant to aphidicolin, but sensitive to ddTTP and NEM. Human proliferating cell nuclear antigen (PCNA) and Drosophila replication protein A (RP-A) did not affect the polymerase activity. It preferred poly(dA)/oligo(dT) as a template-primer. The molecular mass was about 230 kDa with a broad peak region of 200 to 300 kDa in HiPrep16/30 Sephacryl S-300 gel filtration. These properties a different from those of all reported Drosophila polymerase classes such as alpha, beta, gamma, delta, epsilon and zeta and closely resemble those of the gene product expected from the nucleotide sequence. The new polymerase species appears to have ATPase and 3'-5' exonuclease activities as shown by the chromatographies.  相似文献

We developed a novel flow particle analyzer that automatically classifies airborne pollen grains. The design of the particle counter (model KP-1000) is based on that of a flow cytometer, applied to the measurement of airborne particles. The counter classifies pollen species by simultaneously detecting both scattered light and the characteristic fluorescence excited by ultraviolet light in the flow cell. We observed airborne pollen using KP-1000 pollen counters and Durham samplers to compare their performance at three study sites in Japan during the spring pollen season. The pollen counter followed the variation in pollen concentrations, and its daily pollen counts were significantly correlated with the results of the Durham sampling method at all study sites. Although the counter over- or under-counted 2 target pollen species (Cryptomeria japonica and Chamaecyparis obtusa) when they coexisted, a data correction based on the Durham sampling results improved the accuracy of pollen classification of the counter. Our results indicate that the new pollen counter has a strong potential for counting and identifying airborne pollen grains in real time, and it requires further improvement, field trials, and tests with other common airborne pollen grains.  相似文献

Although masting in Southeast Asia is characterized by the interspecific synchronization of reproduction, little is known about the variation in regeneration strategies among sympatric tree species during major masting events. Herein, we examined the hypothesis that non-abundant species achieve greater seed survival per seed production at the pre- and post-dispersal stages by synchronizing reproduction with abundant species. During the masting event from May to September 2010, we installed seed traps in a primary forest plot of the Deramakot Forest Reserve, Borneo. To identify the possible causes of post-dispersal seed mortality, we conducted a seed-sowing experiment from September 2010 to July 2011 in a primary forest plot with high community-level dipterocarp seed density and two surrounding secondary forests with low seed densities. An abundant species (Shorea multiflora) produced more seeds than other species and exhibited a lower survival rate during the pre-dispersal stage. The ratio of aborted seeds was greater in species with greater seed production, while the ratio of seeds predated by insects was not, suggesting that resource limitations and/or greater inbreeding frequency may explain inefficient seed production. Interspecific variation was rarely observed for post-dispersal seed survival rates. Our study highlights the density-dependent variation in reproductive success between abundant and non-abundant dipterocarp species at the pre-dispersal stage. Non-abundant species achieved greater reproductive success by synchronizing reproduction with the masting of abundant species, which might be an important mechanism for sustainability of rare species populations.  相似文献
