An experimental procedure and method of analysis are presented for calibration of a thin-beam force transducer. The beam transducer can be produced and calibrated with a minimum coefficient of 10 ng (10(-5) dyne) force per micron (10(-4) cm) deflection, i.e., kB approximately 0.1 dyne/cm. Since beam deflections on the order of 0.1 micron can be detected, forces of a few nanograms can be resolved. Such forces are common in mechanical experiments on microscopic bodies, e.g., biological cells, artificial membrane capsules, droplets, etc.  相似文献

Modulation of cholesterol 7 alpha-hydroxylase catalytic activity by adenine nucleotides was studied in rat liver microsomal preparations. Inactivation of cholesterol 7 alpha-hydroxylase showed specific requirements of ATP and ADP. AMP and cyclic AMP were stimulatory and cyclic AMP had no effect in the ATP inactivation. The inactivation reactions by ATP were dependent on Mg2+ ions, a cytosolic factor, and time. Ca2+ ions were less effective whereas Mn2+ ions were highly inhibitory to hydroxylase activity. The inactivation could be reversed in a time-dependent reaction requiring a cytosolic activator that was precipitable by ammonium sulphate of saturation up to 65%. The current data suggest that cholesterol 7 alpha-hydroxylase can exist in two catalytic forms that are reversible.  相似文献

Proteolytic cleavage of pyridoxal kinase into two structural domains   总被引:1,自引:0,他引:1  
Chymotryptic digestion of sheep brain pyridoxal kinase, a dimer of identical subunits each of 40 kDa, yields 2 fragments of 24 and 16 kDa with concomitant loss of catalytic activity. These fragments were separated by chromatographic techniques and analyzed for interaction with the ATP analogue, trinitrophenyl-ATP, using fluorescence spectroscopy. The absorption and fluorescence properties of trinitrophenyl-ATP bound to the fragment of 24 kDa (emission maximum, 540 nm, emission anisotropy at 460 nm, 0.30, and fluorescence lifetime, gamma = lns) are indistinguishable from those of the ATP analogue bound to the native enzyme. The fragment of 16 kDa does not bind trinitrophenyl-ATP. The results are consistent with the hypothesis that monomeric pyridoxal kinase is folded into 2 domains connected by a single polypeptide chain sensitive to proteolytic cleavage.  相似文献

Reduced, disordered hen egg lysozyme rapidly regains enzymic activity in a nonenzymic system previously reported from this laboratory. When such regenerations are carried out at high temperatures (where the native enzyme is unstable), native enzyme is formed as a transient intermediate. Thermal inactivation does not depend on irreversible changes in the protein, as shown by experiments wherein thermally inactivated regenerated lysozyme spontaneously reactivates at 37°. These findings are inconsistent with thermodynamic determinism of protein structure.  相似文献

Pentaerythritol tetranicotinate (Perycit), at an oral dosage of 750 mg daily, was given to 12 patients with idiopathic hyperlipidemia and to 12 patients with hyperlipidemia superimposed with diabetes mellitus (DM). With 2 months off-drug period as the baseline, each patient then received 3 months of placebo and 3 months of Perycit. The sequence of treatment was randomized and balanced in frequency. Blood glycosylated hemoglobin (Hb A1) and fasting plasma glucose (FPG) were used as indices of diabetic control. Serum triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), and TC/HDL-C ratio were measured and calculated in order to compare the antilipemic effectiveness of Perycit with that of placebo. The non-parametric Wilcoxon test was used for the statistical analysis. The results showed that in the idiopathic group, Perycit significantly lowered the serum level of TG and the ratio of TC/HDL-C, and elevated the serum level of HDL-C. In the diabetic group, although there was a similar improvement in diabetic control in both periods of placebo and Perycit treatments, there was no change in the serum levels of TG and HDL-C. There was a slight increase of the serum levels of TC in the periods of Perycit treatment, whereas a small increase of HDL-C resulted in a mild decrease of the TC/HDL-C ratio. There was mild and transient facial flushing during the Perycit treatment in 6 out of 12 diabetic patients. Otherwise, there was no side effects in either group. Pooling the two groups' data together, Perycit increased the serum levels of HDL-C and decreased the TC/HDL-C ratio. It is concluded that Perycit has antilipemic effects in patients with idiopathic hyperlipidemia, and may be helpful in reducing the atherogenic risks in these patients. In patients with hyperlipidemia superimposed with DM, although the serum lipids composition was not significantly changed after Perycit, the atherogenic risks might also be reduced as demonstrated by the decrease of the TC/HDL-C ratio.  相似文献

The differentiation of male specific sensory rays in the nematode Caenorhabditis elegans is a complex process regulated by multiple genetic components. A novel approach with heat shock treatment was employed to show that multistep regulation is involved in this process. Intervention in this stepwise regulation resulted in phenocopy of specific gene mutations. The results suggest that differential gene function acting at a precise time frame is necessary to guide the normal differentiation of sensory rays.  相似文献

A TdR carrier-transport system, believed to be facilitated diffusion, has been shown to exist in Ehrlich ascites tumour cells. It is suggested that this system is the predominant transport mechanism at low extracellular concentrations (less than 1-5 micron). The transport system was damaged considerably by 5 krad X-radiation, resulting in a 30-35 per cent reduction in the initial total TdR uptake rat at low extracellular concentrations and 15-20 min after irradiation. The extent of the damage was dependent on the age of the cells as was reflected by relative decreases in V max and Km. It can be concluded that the enhanced depression in 14C-TdR incorporation into DNA of irradiated cells when low precursor concentrations were used for monitoring, is partly attributed to the radiation-induced damage to the carrier-transport system. The permeability constant for passive diffusion in asynchronous E.A.T. cells and the endogenous natural rate of dTTP synthesis in S-phase cells were estimated.  相似文献
