Soy‐Derived Phytochemical Genistein Modifies Chromatome Topology to Restrict Cancer Cell Proliferation

Epidemiological data indicate that human cancer risk is significantly reduced by the consumption of soy‐based foods containing the “phytoestrogen” genistein, which can signal via host cell estrogen receptors. While additional chemoprotective effects of genistein induced by epigenetic factors have also been reported, the key molecules and mechanisms involved are poorly defined. We therefore investigated genistein effects on chromatin‐bound proteins in the estrogen receptor‐deficient cell line MDA‐MB‐231 which is insensitive to phytoestrogen signaling. After exposure to low‐dose genistein for >1 month, MDA‐MB‐231 cells exhibited stable epigenetic alterations that are analyzed via partial MNase digestion and TMT‐based quantitative proteomics. 3177 chromatin‐bound proteins are identified with high confidence, including 882 molecules that displayed altered binding topology after cell conditioning with genistein. Prolonged phytochemical exposure conferred heritable changes in the binding topology of key epigenetic regulators including ATRX, SUV39H1/H2, and HP1BP3 that are preserved in untreated progeny, resulting in sustained downregulation of proliferation genes and reduced cell growth. These data indicate that soy derivative genistein exerts complex estrogen receptor‐independent effects on the epigenome likely to influence tumorigenesis by restricting cell growth. 相似文献

Introduction: C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) which are used for assessment of axial spondyloarthritis (AxSpA) related disease activity have poor specificity and sensitivity. Neutrophil to lymphocyte ratio (NLR), platelet to lymphocyte ratio (PLR) and mean platelet volume (MPV) have not been investigated as disease activity markers among Asian AxSpA patients.Methodology: A retrospective, cross-sectional study was conducted in Singapore General Hospital from January 2013 to December 2015 to investigate the role of NLR, PLR and MPV as disease activity markers in AxSpA patients.Results: The mean age of patients (n?=?122) was 37.0?±?12.5 years old and majority of them were male (n?=?93, 76.2%). No significant differences were found between patients with disease with regards to age, gender, ethnicity, HLAB-27 status, age at onset of diagnosis of AxSpA, duration of disease and comorbidities such as cardiac disease (p?>?0.05).There were no significant differences in the ESR, NLR, PLR and MPV between the four disease activity groups (p?>?0.05). However, patients with very high disease activity had higher ESR and CRP compared to patients with inactive disease and moderate disease activity (p?0.05).Conclusion: NLR, PLR and MPV were not associated with disease activity in Asian AxSpA patients. 相似文献

There are conflicting views on whether collagen X is a purely structural molecule, or regulates bone mineralization during endochondral ossification. Mutations in the human collagen α1(X) gene (COL10A1) in Schmid metaphyseal chondrodysplasia (SMCD) suggest a supportive role. But mouse collagen α1(X) gene (Col10a1) null mutants were previously reported to show no obvious phenotypic change. We have generated collagen X deficient mice, which shows that deficiency does have phenotypic consequences which partly resemble SMCD, such as abnormal trabecular bone architecture. In particular, the mutant mice develop coxa vara, a phenotypic change common in human SMCD. Other consequences of the mutation are reduction in thickness of growth plate resting zone and articular cartilage, altered bone content, and atypical distribution of matrix components within growth plate cartilage. We propose that collagen X plays a role in the normal distribution of matrix vesicles and proteoglycans within the growth plate matrix. Collagen X deficiency impacts on the supporting properties of the growth plate and the mineralization process, resulting in abnormal trabecular bone. This hypothesis would accommodate the previously conflicting views of the function of collagen X and of the molecular pathogenesis of SMCD. 相似文献

Diatoms are potentially the most important biomonitors of environmental change in high arctic lakes and ponds, but to date few autecological data are available. Because of the shallow nature of many of these water bodies, a large proportion of taxa are periphytic and planktonic diatoms are absent for the most part. By determining the microhabitat and substrate preferences of these benthic diatom taxa, the potential exists to infer past changes in available habitats from fossil diatom assemblages collected from sediment cores and ultimately to reconstruct past environmental and climatic changes responsible for these shifts in habitat availability. To refine our understanding of high arctic diatom habitat preference, the common diatom taxa found on submerged moss (bryophyte), sediment, and rock substrates from lakes and ponds on Bathurst Island, Nunavut, Canadian High Arctic were examined. The relationships among key limnological variables and the common taxa from each habitat were examined. Many diatom taxa exhibited varying degrees of microhabitat preference, with moss representing the more unique habitat. In addition, the following limnological variables significantly ( P ≤ 0.05) explained the species variance for each of the three substrates: Na + and total nitrogen for moss; total phosphorus (filtered) and pH for rock; and Fe3 + , total phosphorus (unfiltered), total nitrogen, temperature, and pH for sediment. These data can be used to help interpret monitoring and paleolimnological studies in this environmentally sensitive region. 相似文献

As part of our analysis of the role of a uniquely clustered set of dam methylation sites (the motif GATC) within the origin of DNA replication in Escherichia coli, we have studied the effect of GATCs in various methylation states on the intrinsic curvature of DNA. We have designed a
set of DNA linkers and used commercially available linkers containing GATC motifs. The linkers were ligated and the electrophoretic
mobility of the resulting multimers in different states of methylation was tested relative to reference fragments. We report
that properly phased GATCs in certain sequence environments modulate DNA curvature and that these effects may be enhanced
by N6-adenine methylation of the GATCs. These structural alterations may in turn affect DNA-protein interactions, especially those
involving proteins that rely on both primary sequence and structure for recognition. We present an example, where introduction
of a GATC within an integration host factor (IHF) binding site, which does not alter the consensus sequence, reduces the binding
affinity of the protein for the modified site.

Determining the relationship between protein folding pathways on and off the ribosome remains an important area of investigation in biology. Studies on isolated domains have shown that alteration of the separation of residues in a polypeptide chain, while maintaining their spatial contacts, may affect protein stability and folding pathway. Due to the vectorial emergence of the polypeptide chain from the ribosome, chain connectivity may have an important influence upon cotranslational folding. Using MATH, an all β-sandwich domain, we investigate whether the connectivity of residues and secondary structure elements is a key determinant of when cotranslational folding can occur on the ribosome. From Φ-value analysis, we show that the most structured region of the transition state for folding in MATH includes the N and C terminal strands, which are located adjacent to each other in the structure. However, arrest peptide force-profile assays show that wild-type MATH is able to fold cotranslationally, while some C-terminal residues remain sequestered in the ribosome, even when destabilized by 2–3?kcal?mol?1. We show that, while this pattern of Φ-values is retained in two circular permutants in our studies of the isolated domains, one of these permutants can fold only when fully emerged from the ribosome. We propose that in the case of MATH, onset of cotranslational folding is determined by the ability to form a sufficiently stable folding nucleus involving both β-sheets, rather than by the location of the terminal strands in the ribosome tunnel. 相似文献

While there are persistent calls for developing more marine protected areas (MPAs) for Asian horseshoe crab conservation in response to population declines in Asia–Pacific region, most existing horseshoe crab MPAs were designated without prior comprehensive population assessment and habitat characterization. This study collected biological and habitat information in Hong Kong, in order to identify priority sites for conservation and management of Asian horseshoe crab populations. The territory-wide surveys at 18 spawning/nursery beaches displayed a persistently low mean juvenile density from 0.16 to 2.19 ind/100 m2 in 2012 and 0.08 to 1.41 ind/100 m2 in 2014. These density data were within the same range of that in 2002 and 2007 (0.10–1.97 ind/100 m2), apart from a low survey return of 0.08–0.31 ind/100 m2 in 2004. The current population of juvenile T. tridentatus and C. rotundicauda in Hong Kong is estimated about 2100–4300 and 2400–3000 individuals, respectively. From the 2012–2014 data, no new recruitments of 1st–3rd instar juveniles were found, as the shores were occupied mostly by 6th–9th instar juveniles of prosomal width between 23 and 45 mm, in which considerably high mortality rates were noted. The present findings of existing small and discrete juvenile populations, coupled with relatively few recruits, suggest that the status of juvenile horseshoe crabs in Hong Kong is fragile and vulnerable to local extirpation, especially if no urgent protection measures are implemented. Based on available population data and habitat characteristics, three MPAs, ranging from 5 to 7 km2, are proposed, so as to conserve over 60 and 70 % of the existing juvenile populations of T. tridentatus and C. rotundicauda, respectively. The approach adopted in this study may serve as a case study for proposing other horseshoe crab MPAs in Asia, where increasing human disturbances and over-harvest are imminent. 相似文献
