Metabolic approaches to understanding climate change impacts on seasonal host‐macroparasite dynamics
Climate change is expected to alter the dynamics of infectious diseases around the globe. Predictive models remain elusive due to the complexity of host–parasite systems and insufficient data describing how environmental conditions affect various system components. Here, we link host–macroparasite models with the Metabolic Theory of Ecology, providing a mechanistic framework that allows integrating multiple nonlinear environmental effects to estimate parasite fitness under novel conditions. The models allow determining the fundamental thermal niche of a parasite, and thus, whether climate change leads to range contraction or may permit a range expansion. Applying the models to seasonal environments, and using an arctic nematode with an endotherm host for illustration, we show that climate warming can split a continuous spring‐to‐fall transmission season into two separate transmission seasons with altered timings. Although the models are strategic and most suitable to evaluate broad‐scale patterns of climate change impacts, close correspondence between model predictions and empirical data indicates model applicability also at the species level. As the application of Metabolic Theory considerably aids the a priori estimation of model parameters, even in data‐sparse systems, we suggest that the presented approach could provide a framework for understanding and predicting climatic impacts for many host–parasite systems worldwide. 相似文献

Many northern Canadians have continued a subsistence lifestyle of wildlife harvesting and, therefore, value sustainable wildlife
populations. At a regional wildlife workshop in the Sahtu Settlement Area, Northwest Territories in 2002, elders and community
leaders raised concerns regarding wildlife health, food safety, and the effects of climate change on wildlife. They requested
that efforts be put toward training youth in science and increasing involvement of hunters and youth in wildlife research.
In response, we initiated a long-term, integrated approach to foster community-based wildlife health monitoring and research.
Annual trips were made to all schools in the Sahtu from 2003 to 2009 to provide hands-on learning for 250–460 students on
a range of wildlife topics. In addition, interviews were conducted with 31 hunters and elders to document their local ecological
knowledge of wildlife health and local hunters were trained as monitors to collect tissue samples and measurements to assess
body condition and monitor health of harvested caribou (n = 69) and moose (n = 19). In 2007 the program was extended to include participation in the annual caribou hunt held by one community. Each year
since 2005, a graduate student and/or a postdoctoral trainee in the veterinary or biological sciences has participated in
the program. The program has evolved during the last 6 years in response to community and school input, results of empirical
research, hunter feedback, local knowledge, and logistical constraints. The continuity of the program is attributed to the
energetic collaboration among diverse partners and a unified approach that responds to identified needs. 相似文献

Early evolution of metazoan serine/threonine and tyrosine kinases: identification of selected kinases in marine sponges 总被引:14,自引:1,他引:13
The phylum Porifera (sponges) was the first to diverge from the common
ancestor of the Metazoa. In this study, six cDNAs coding for protein-
serine/threonine kinases (PS/TKs) are presented; they have been isolated
from libraries obtained from the demosponges Geodia cydonium and Suberites
domuncula and from the calcareous sponge Sycon raphanus. Sequence
alignments of the catalytic domains revealed that two major families of
PS/TK, the "conventional" (Ca(2+)-dependent) protein kinase C (PKC), the
cPKC subfamily, as well as the "novel" (Ca(2+)- independent) PKC (nPKC),
form two separate clusters. In each cluster, the sequence from S. raphanus
diverges first. To approach the question about the origin of
protein-tyrosine kinases (PTK), which are found only in Metazoa, we
analyzed two additional PS/TKs which have been cloned from S. domuncula:
the stress-responsive protein kinase (KRSvSD) and the
protein-kinase-C-related kinase (PRKvSD). The construction of the
phylogenetic tree, comprising the eight PS/TKs and the PTK cloned
previously from G. cydonium, revealed that the PTK derived from the branch
including the KRSvSD kinase. These data facilitate the first molecular
approach to elucidate the origin of metazoan PTK within the PS/TK
superfamily.

A cultured rat ovarian cell line (31 A-F(2)) was used to study the effect of growth factors (epidermal growth factor [EGF] and fibroblast growth factor [FGF]), a survival factor (ovarian growth factor [OGF]), a hormone (insulin), and an iron-binding protein (transferring) on cell proliferation and steroid production under defined culture conditions. EGF and insulin were shown to be mitogenic (half-maximal response at 0.12 nM and 0.11 muM, respectively) for 31A-F(2) cells incubated in serum-free medium. EGF induced up to three doublings in the cell population, whereas insulin induced an average of one cell population doubling. FGF, OGF, and transferrin were found not to have any prominent effect on cell division when incubated individually with 31A-F(2) cells in serum-free medium. However, a combination of EGF, OGF, insulin, and transferrin stimulated cell division to the same approximate extent as cells incubated in the presence of 5 percent fetal calf serum. EGF or insulin did not significantly affect total cell cholesterol levels (relative to cells incubated in serum-free medium) when incubated individually with 31A-F(2) cells. However, cell cholesterol levels were increased by the addition of OGF (250 percent), FGF (370 percent), or a combination of insulin and EGF (320 percent). Progesterone secretion from 31A-F(2) cells was enhanced by EGF (25 percent), FGF (80 percent), and insulin (115 percent). However, the addition of a mitogenic mixture of EGF, OGF, insulin, and transferrin suppressed progesterone secretion 150 percent) below that of control cultures. These studies have permitted us to determine that EGF and insulin are mitogenic factors that are required for the growth of 31A-F(2) cells and that OGF and transferrin are positive cofactors that enhance growth. Also, additional data suggest that cholesterol and progesterone production in 31A-F(2) cells can be regulated by peptide growth factors and the hormone insulin. 相似文献

Parasite richness and prevalence in wild animals can be used as indicators of population and ecosystem health. In this study, the gastrointestinal parasites of ursine colobus monkeys (Colobus vellerosus) at the Boabeng‐Fiema Monkey Sanctuary (BFMS), Ghana, were investigated. BFMS is a sacred grove where monkeys and humans have long lived in relatively peaceful proximity. Fecal samples (n = 109) were collected opportunistically from >27 adult and subadult males in six bisexual groups and one all‐male band from July 2004 to August 2005. Using fecal floatation, we detected three protozoans (two Entamoeba sp., Isospora sp.), five nematodes (Ascaris sp., Enterobius sp., Trichuris sp., two strongyle sp.), and one digenean trematode. Using fluorescein labeled antibodies, we detected an additional protozoan (Giardia sp.), and with PCR techniques, we characterized this as G. duodenalis Assemblage B and also identified a protistan (Blastocystis sp., subtype 2). The most prevalent parasite species were G. duodenalis and Trichuris sp. Parasites were more prevalent in the long wet season than the long dry. Parasite prevalence did not vary by age, and average parasite richness did not differ by rank for males whose status remained unchanged. However, males that changed rank tended to show higher average parasite richness when they were lower ranked. Individuals that spent more time near human settlements had a higher prevalence of Isospora sp. that morphologically resembled the human species I. belli. The presence of this parasite and G. duodenalis Assemblage B indicates possible anthropozoonotic and/or zoonotic transmission between humans and colobus monkeys at this site. Standardized flotation techniques were used to survey 56 grizzly bear (Ursus arctos) fecal samples for parasites. The samples were collected during the spring and autumn of 1995 and 1996 in the central Arctic of the Northwest Territories (Canada). Parasites of the genera Nematodirus, gastrointestinal coccidia, and an unidentified first stage protostrongylid larva are reported for the first time from grizzly bear feces in North America. Parasites of the genera Diphyllobothrium and Baylisascaris also were collected. Prevalence of gastrointestinal parasites were significantly different between the spring and autumn seasons (31% and 58% respectively). Thus, we provide evidence supporting the theory that bears void gastrointestinal parasites before hibernation. 相似文献

The purpose of this study was to investigate the prevalence, intensity, and possible transmission routes of the trematode, Controrchis spp. (Dicrocoeliidae), in a population of black howler monkeys (Alouatta pigra) near Monkey River, Belize. Alouatta pigra are arboreal primates that are typically folivorous and frugivorous. Controrchis is a dicrocoeliid trematode and, as such, should require a gastropod and an ant (Formicidae) intermediate host for transmission. From January to July 2005 and February 2006 to June 2007, we collected fecal samples and focal animal data from 18 individual primates in four social groups. Feces were stored in either 10% buffered formalin or 95% ethanol and examined using a double-centrifugation sugar-flotation technique. The prevalence of Controrchis spp. was 89%, and the mean intensity was 2.29 eggs per gram (epg), with a range of 1.00-6.57 epg. A general linear mixed model to examine the effect of sex, rainfall, and time spent feeding on various plant species, while controlling for individual and group identity, revealed that both group and the amount of time spent feeding on trumpet trees (Cecropia peltata) by individuals predicted the prevalence and intensity of Controrchis spp. infestation. Cecropia peltata has a mutualistic relationship with Azteca spp. ants and constitutes an important food source for A. pigra. Our findings provide strong, circumstantial evidence that A. pigra are infected with Controrchis spp. through the ingestion of metacercariae in Azteca spp. when feeding on C. peltata. Because C. peltata is a pioneer tree species and typically occurs in disturbed forests, results from this study suggest an important link between habitat disturbance and parasitism in a wild mammal. 相似文献

Discovery of the ostertagiine nematode Teladorsagia boreoarcticus n. sp. in muskoxen, Ovibos moschatus, from the central Canadian Arctic highlights the paucity of knowledge about the genealogical and numerical diversity of nematode faunas characteristic of artiodactyls at high latitudes across the Holarctic. Teladorsagia boreoarcticus is a dimorphic cryptic species distinguished from Teladorsagia circumcincta/Teladorsagia trifurcata in domestic sheep by a 13% divergence in the ND4 region of mitochondrial DNA, constant differences in the synlophe, and significantly longer esophageal valve, spicules, gubernaculum, and bursa. Teladorsagia boreoarcticus represents an archaic component of the North American fauna and may have a Holarctic distribution in muskoxen and caribou. Recognition of T. boreoarcticus in muskoxen, in part, corroborates hypotheses for the existence of a cryptic species complex of Teladorsagia spp. among Caprinae and Cervidae at high latitudes and indicates the importance of climatological determinants during the late Tertiary and Pleistocene on diversification of the fauna. Also reinforced is the concept of the North American fauna as a mosaic of endemic and introduced species. Discovery of a previously unrecognized species of Teladorsagia has additional implications and clearly indicates that (1) our knowledge is incomplete relative to potentially pathogenic nematodes that could be exchanged among domestic and wild caprines; (2) we do not have sufficient knowledge of the fauna to understand the ecological control mechanisms (limitations) on dissemination and host range; and (3) an understanding of historical and geographical influences on the genealogical diversity and distribution of nematode faunas in domestic and wild ruminants is requisite to define the interface between agricultural and natural ecosystems across the Holarctic. 相似文献
