The idea that many processes in arid and semi-arid ecosystems are dormant until activated by a pulse of rainfall, and then decay from a maximum rate as the soil dries, is widely used as a conceptual and mathematical model, but has rarely been evaluated with data. This paper examines soil water, evapotranspiration (ET), and net ecosystem CO2 exchange measured for 5 years at an eddy covariance tower sited in an AcaciaCombretum savanna near Skukuza in the Kruger National Park, South Africa. The analysis characterizes ecosystem flux responses to discrete rain events and evaluates the skill of increasingly complex “pulse models”. Rainfall pulses exert strong control over ecosystem-scale water and CO2 fluxes at this site, but the simplest pulse models do a poor job of characterizing the dynamics of the response. Successful models need to include the time lag between the wetting event and the process peak, which differ for evaporation, photosynthesis and respiration. Adding further complexity, the time lag depends on the prior duration and degree of water stress. ET response is well characterized by a linear function of potential ET and a logistic function of profile-total soil water content, with remaining seasonal variation correlating with vegetation phenological dynamics (leaf area). A 1- to 3-day lag to maximal ET following wetting is a source of hysteresis in the ET response to soil water. Respiration responds to wetting within days, while photosynthesis takes a week or longer to reach its peak if the rainfall was preceded by a long dry spell. Both processes exhibit nonlinear functional responses that vary seasonally. We conclude that a more mechanistic approach than simple pulse modeling is needed to represent daily ecosystem C processes in semiarid savannas. The neural supply of the dorsal lingitudinal muscles in successive segments from the prothorax to the pregenital abdomen of adult and larval instar locusts (Locusta migratoria and Schistocerca gregaria) has been studied. Stainings have also been carried out for embryos. The whole complement consists of three muscles, of which one or both of the smaller ones degenerate in the pterothoracic segments during early imaginal life. Based on morphological criteria, several motoneurone types can be distinguished. The neural set is almost identical for all segments, independent of the general organization of each segment. At about 65% of embryogenesis, all neurone types can be identified with respect to soma position and basic features of the central branching pattern. By the end of embryogenesis, a dendritic pattern is established which resembles the adult pattern in all major aspects. The reiteration of homonomous elements suggests that they form part of the basic segmental neural Bauplan generated early in embryogenesis. Summary The development of the flight motor pattern was studied by recording from the thoracic muscles of locusts of various developmental stages. In response to a short wind stimulus, larval locusts generate unpatterned motor activity, whereas newly moulted adults generate the flight pattern (Fig. 1A). The latter is equivalent to the mature adult flight pattern, although more irregular and of lower frequency. Experiments with highly deafferentated locusts indicate that the switch from the larval tonic to adult phasic flight pattern and subsequent increase in frequency are not dependent on phasic peripheral feedback from moving body structures (Fig. 1B). By using octopamine, flight motor activity could be released without need of the wind stimulus (Fig. 2). This corresponded to the normal wind released flight pattern of intact locusts, although the frequency was lower (Fig. 8). Following octopamine treatment, the response to wind stimulation was enhanced. Wind then released in deafferentated adults long flight sequences of significantly elevated frequency (Fig. 3). Although flight is essentially an adult specific behaviour, octopamine was finally found to release flight motor activity in all larval stages (Fig. 7).We conclude that major steps in the development of the flight motor circuitry are completed by the end of embryogenesis. Thus, in contrast to previous assumptions (cf. Bentley and Hoy 1970; Kutsch 1974a; Altman 1975), postembryonic changes in neither the central, nor peripheral nervous system appear to be of major importance for the ontogeny of the locust flight motor program. Whether developmental changes in the wind sensory system of the head, or levels of neurohormones such as octopamine, are related to the newly acquired responsiveness of freshly moulted adult locusts to the normal flight releasing stimulus is discussed.  相似文献

Synchronization of oocyte maturation in vitro has been shown to produce higher in vitro fertilization (IVF) rates than those observed in oocytes matured in vitro without synchronization. However, the increased IVF rates never exceeded those observed in oocytes matured in vivo without synchronization. This study was therefore designed to define the effect of in vivo synchronization of oocyte maturation on IVF rates. Mice were superovulated and orally treated with 7.5 mg cilostazol (CLZ), a phosphodiesterase 3A (PDE3A) inhibitor, to induce ovulation of immature oocytes at different stages depending on frequency and time of administration of CLZ. Mice treated with CLZ ovulated germinal vesicle (GV) or metaphase I (MI) oocytes that underwent maturation in vitro or in vivo (i.e. in the oviduct) followed by IVF. Superovulated control mice ovulated mature oocytes that underwent IVF directly upon collection. Ovulated MI oocytes matured in vitro or in vivo had similar maturation rates but significantly higher IVF rates, 2–4 cell embryos, than those observed in control oocytes. Ovulated GV oocytes matured in vitro showed similar maturation rates but significantly higher IVF rates than those observed in control oocytes. However, ovulated GV oocytes matured in vivo had significantly lower IVF rates than those noted in control oocytes. It is concluded that CLZ is able to synchronize oocyte maturation and improve IVF rates in superovulated mice. CLZ may be capable of showing similar effects in humans, especially since temporal arrest of human oocyte maturation with other PDE3A inhibitors in vitro was found to improve oocyte competence level. The capability of a clinically approved PDE3A inhibitor to improve oocyte fertilization rates in mice at doses extrapolated from human therapeutic doses suggests the potential scenario of the inclusion of CLZ in superovulation programs.
